    Muscular Strength and Body Composition Comparison Between the Charlotte-Mecklenburg Fire and Police Departments

    Firefighters and police have different physical job requirements and selection processes. Firefighters have on-duty exercise time and police do not. This could affect body composition and strength. The purpose of this investigation was to compare body composition and bench press strength between firefighters and police officers in the Charlotte-Mecklenburg departments. Current 2006 fitness records were analyzed to compare firefighters and police (N = 2330). Variables included: gender, age, body mass, percent body-fat, fat mass, lean mass, bench press strength, bench press/lean mass, bench press/body mass, and percent that were obese. Significant (p ≤ 0.05) differences were found between male firefighters and police in age (37.7 vs. 36.9 yrs), body mass (91.5 vs. 93.2 kg), percent body-fat (17.8% vs.18.5%), fat mass (16.9 vs. 18.0 kg), bench press strength (93.4 vs. 96.3 kg), bench press/body mass (1.03 vs.1.05) and percent that were obese (10% vs.17%), respectively. Significant differences between female firefighters and police were found in age (42.4 vs. 37.4 yrs), body mass (77.5 vs. 71.8 kg), lean mass (54.4 vs. 51.2 kg), bench press strength (52.3 vs. 43.9 kg), bench press/lean mass (0.99 vs. 0.86) and bench press/body mass (0.69 vs. 0.62), respectively. Male police were younger, weighed more, had higher percent body fat, higher percent of obesity, and had greater upper body strength than firefighters. Female police were younger, weighed less, had less lean mass and less upper body strength than their firefighter counterparts. Contributing factors might include pre-employment selection, recruit training, in-service physical fitness testing, and counseling provided by departments

    Novel self-assembling cyclic peptides with reversible supramolecular nanostructures

    Self-assembly peptides (SAPs) are an important class of hydrogels used in nanomedicine for tissue repair and neural regeneration. Due to their unique properties, SAPs may be used in a wide range of applications but some limitations, such as low bioavailability and rapid hydrolysis degradation, need to be overcome. Here, we describe the synthesis and characterization of two novel cyclic SAPs without the use of d/l-alternating amino acids, showing a reversible transition of their supramolecular nanostructures, from nanotubes/nanofibers into nanovesicles/nanospheres. The investigation, characterization and optimization are performed using atomic force microscopy (AFM), attenuated total reflectance-Fourier transform infrared (ATR-FTIR) spectroscopy, Raman analysis, circular dichroism (CD), and rheology measurements. Also, in vitro cell viability assays show negligible toxicity of the representative optimized cyclic SAP towards human neural stem cells (hNSCs). Our results suggest that linear SAP theoretical background can be applied to develop cyclic SAPs, with important implications in the scalable fabrication of inter-changeable nanostructures, as well as for biomedical applications, including tissue regeneration, drug-delivery, drug-design, sensing, imaging, and size selectivity

    Can Baseline IL-6 Levels Predict Long COVID in Subjects Hospitalized for SARS-CoV-2 Disease?

    The immune response to infection plays a crucial role in the pathogenesis of COVID-19, but several patients develop a wide range of persistent symptoms, which is becoming a major global health and economic burden. However, reliable indicators are not yet available to predict the persistence of symptoms typical of the so-called long COVID. Our study aims to explore an eventual role of IL-6 levels as a marker of long COVID. Altogether, 184 patients admitted to the COVID Medicine Unit of the University Hospital in Palermo, Italy, from the 1st of September 2020, were analyzed. Patients were divided into two groups according to the IL-6 serum levels (normal or elevated), considering the serum IL-6 levels measured during the first four days of hospitalization. In our study, higher serum IL-6 levels were associated with a doubled higher risk of long COVID (OR = 2.05; 95% CI: 1.04–4.50) and, in particular, they were associated with a higher incidence of mobility decline (OR = 2.55; 95% CI: 1.08–9.40) and PTSD (OR = 2.38; 95% CI: 1.06–8.61). The analysis of our case series confirmed the prominent role of IL-6 levels in response to SARS-CoV-2 infection, as predictors not only of COVID-19 disease severity and unfavorable outcomes, but also long COVID development trends

    Identification of a Novel Pseudo-Natural Product Type IV IDO1 Inhibitor Chemotype

    Natural product (NP)-inspired design principles provide invaluable guidance for bioactive compound discovery. Pseudo-natural products (PNPs) are de novo combinations of NP fragments to target biologically relevant chemical space not covered by NPs. We describe the design and synthesis of apoxidoles, a novel pseudo-NP class, whereby indole- and tetrahydropyridine fragments are linked in monopodal connectivity not found in nature. Apoxidoles are efficiently accessible by an enantioselective [4+2] annulation reaction. Biological evaluation revealed that apoxidoles define a new potent type IV inhibitor chemotype of indoleamine 2,3-dioxygenase 1 (IDO1), a heme-containing enzyme considered a target for the treatment of neurodegeneration, autoimmunity and cancer. Apoxidoles target apo-IDO1, prevent heme binding and induce unique amino acid positioning as revealed by crystal structure analysis. Novel type IV apo-IDO1 inhibitors are in high demand, and apoxidoles may provide new opportunities for chemical biology and medicinal chemistry research

    Gelation as arrested phase separation in short-ranged attractive colloid-polymer mixtures

    We present further evidence that gelation is an arrested phase separation in attractive colloid-polymer mixtures, based on a method combining confocal microscopy experiments with numerical simulations recently established in {\bf Nature 453, 499 (2008)}. Our results are independent of the form of the interparticle attractive potential, and therefore should apply broadly to any attractive particle system with short-ranged, isotropic attractions. We also give additional characterization of the gel states in terms of their structure, inhomogeneous character and local density.Comment: 6 figures, to be published in J. Phys. Condens.     Effects of simulated altitude (normobaric hypoxia) on cardiorespiratory parameters and circulating endothelial precursors in healthy subjects

    <p>Abstract</p> <p>Background</p> <p>Circulating Endothelial Precursors (PB-EPCs) are involved in the maintenance of the endothelial compartment being promptly mobilized after injuries of the vascular endothelium, but the effects of a brief normobaric hypoxia on PB-EPCs in healthy subjects are scarcely studied.</p> <p>Methods</p> <p>Clinical and molecular parameters were investigated in healthy subjects (n = 8) in basal conditions (T0) and after 1 h of normobaric hypoxia (T1), with Inspiratory Fraction of Oxygen set at 11.2% simulating 4850 mt of altitude. Blood samples were obtained at T0 and T1, as well as 7 days after hypoxia (T2).</p> <p>Results</p> <p>In all studied subjects we observed a prompt and significant increase in PB-EPCs, with a return to basal value at T2. The induction of hypoxia was confirmed by Alveolar Oxygen Partial Pressure (PAO<sub>2</sub>) and Spot Oxygen Saturation decreases. Heart rate increased, but arterial pressure and respiratory response were unaffected. The change in PB-EPCs percent from T0 to T1 was inversely related to PAO<sub>2 </sub>at T1. Rapid (T1) increases in serum levels of hepatocyte growth factor and erythropoietin, as well as in cellular PB-EPCs-expression of Hypoxia Inducible Factor-1α were observed.</p> <p>Conclusion</p> <p>In conclusion, the endothelial compartment seems quite responsive to standardized brief hypoxia, possibly important for PB-EPCs activation and recruitment.</p

    The topology of a discussion: the #occupy case

    We analyse a large sample of the Twitter activity developed around the social movement 'Occupy Wall Street' to study the complex interactions between the human communication activity and the semantic content of a discussion. We use a network approach based on the analysis of the bipartite graph @Users-#Hashtags and of its projections: the 'semantic network', whose nodes are hashtags, and the 'users interest network', whose nodes are users In the first instance, we find out that discussion topics (#hashtags) present a high heterogeneity, with the distinct role of the communication hubs where most the 'opinion traffic' passes through. In the second case, the self-organization process of users activity leads to the emergence of two classes of communicators: the 'professionals' and the 'amateurs'. Moreover the network presents a strong community structure, based on the differentiation of the semantic topics, and a high level of structural robustness when a certain set of topics are censored and/or accounts are removed. Analysing the characteristics the @Users-#Hashtags network we can distinguish three phases of the discussion about the movement. Each phase corresponds to specific moment of the movement: from declaration of intent, organisation and development and the final phase of political reactions.     Damage detection via shortest-path network sampling

    Large networked systems are constantly exposed to local damages and failures that can alter their functionality. The knowledge of the structure of these systems is, however, often derived through sampling strategies whose effectiveness at damage detection has not been thoroughly investigated so far. Here, we study the performance of shortest-path sampling for damage detection in large-scale networks. We define appropriate metrics to characterize the sampling process before and after the damage, providing statistical estimates for the status of nodes (damaged, not damaged). The proposed methodology is flexible and allows tuning the trade-off between the accuracy of the damage detection and the number of probes used to sample the network. We test and measure the efficiency of our approach considering both synthetic and real networks data. Remarkably, in all of the systems studied, the number of correctly identified damaged nodes exceeds the number of false positives, allowing us to uncover the damage precisely

    Poorly differentiated carcinoma arising from adenolymphoma of the parotid gland

    BACKGROUND: There is only one previous case report of a poorly differentiated carcinoma arising from an adenolymphoma of the parotid gland (Warthin's tumour). The absence of clinical symptoms, and the aspecificity of the radiological pattern make the diagnosis very difficult. CASE PRESENTATION: We here report the case of a 73-year-old man with Warthin's tumour who was brought to our attention because of a swelling in the parotid region. CONCLUSIONS: In this case with an atypical clinical presentation, the intra-operative examination of a frozen section of the parotid mass allowed us to diagnose the malignant tumour correctly and consequently undertake its radical excision
