    After the 2014 fatal shooting of Michael Brown, an unarmed 18-year-old African American man, by a White police officer in Ferguson, Missouri, some news organizations included in their immediate coverage a photo of Brown taken from his Facebook page. In a now iconic image, Brown stands in a Nike tank top, unsmiling, and flashing a peace sign (misidentified by some news organizations as a gang sign). Later, less incendiary photos from Brown’s Facebook page surfaced. Within a few days of Brown’s death, the hashtag #IfTheyGunnedMeDown appeared on social media. Young African Americans posted two photos of themselves, representing positive and potentially negative images, questioning which photo would be used by the media if they were shot by police. These posts reflected an insight into the notion of media representation that likely escapes most audience members who regularly view news coverage of Black men. These young African Americans perceptively and concisely identified the problems inherent in dominant media representations of Black men as pathological criminals, a persistent representation that affects both racial attitudes and public policy decisions

    An integrated circuit for chip-based analysis of enzyme kinetics and metabolite quantification

    We have created a novel chip-based diagnostic tools based upon quantification of metabolites using enzymes specific for their chemical conversion. Using this device we show for the first time that a solid-state circuit can be used to measure enzyme kinetics and calculate the Michaelis-Menten constant. Substrate concentration dependency of enzyme reaction rates is central to this aim. Ion-sensitive field effect transistors (ISFET) are excellent transducers for biosensing applications that are reliant upon enzyme assays, especially since they can be fabricated using mainstream microelectronics technology to ensure low unit cost, mass-manufacture, scaling to make many sensors and straightforward miniaturisation for use in point-of-care devices. Here, we describe an integrated ISFET array comprising 216 sensors. The device was fabricated with a complementary metal oxide semiconductor (CMOS) process. Unlike traditional CMOS ISFET sensors that use the Si3N4 passivation of the foundry for ion detection, the device reported here was processed with a layer of Ta2O5 that increased the detection sensitivity to 45 mV/pH unit at the sensor readout. The drift was reduced to 0.8 mV/hour with a linear pH response between pH 2 – 12. A high-speed instrumentation system capable of acquiring nearly 500 fps was developed to stream out the data. The device was then used to measure glucose concentration through the activity of hexokinase in the range of 0.05 mM – 231 mM, encompassing glucose’s physiological range in blood. Localised and temporal enzyme kinetics of hexokinase was studied in detail. These results present a roadmap towards a viable personal metabolome machine

    Novel dimeric β-helical model of an ice nucleation protein with bridged active sites

    <p>Abstract</p> <p>Background</p> <p>Ice nucleation proteins (INPs) allow water to freeze at high subzero temperatures. Due to their large size (>120 kDa), membrane association, and tendency to aggregate, an experimentally-determined tertiary structure of an INP has yet to be reported. How they function at the molecular level therefore remains unknown.</p> <p>Results</p> <p>Here we have predicted a novel β-helical fold for the INP produced by the bacterium <it>Pseudomonas borealis</it>. The protein uses internal serine and glutamine ladders for stabilization and is predicted to dimerize via the burying of a solvent-exposed tyrosine ladder to make an intimate hydrophobic contact along the dimerization interface. The manner in which <it>Pb</it>INP dimerizes also allows for its multimerization, which could explain the aggregation-dependence of INP activity. Both sides of the <it>Pb</it>INP structure have tandem arrays of amino acids that can organize waters into the ice-like clathrate structures seen on antifreeze proteins.</p> <p>Conclusions</p> <p>Dimerization dramatically increases the 'ice-active' surface area of the protein by doubling its width, increasing its length, and presenting identical ice-forming surfaces on both sides of the protein. We suggest that this allows sufficient anchored clathrate waters to align on the INP surface to nucleate freezing. As <it>Pb</it>INP is highly similar to all known bacterial INPs, we predict its fold and mechanism of action will apply to these other INPs.</p

    A colorimetric CMOS-based platform for rapid total serum cholesterol quantification

    Elevated cholesterol levels are associated with a greater risk of developing cardiovascular disease and other illnesses, making it a prime candidate for detection on a disposable biosensor for rapid point of care diagnostics. One of the methods to quantify cholesterol levels in human blood serum uses an optically mediated enzyme assay and a bench top spectrophotometer. The bulkiness and power hungry nature of the equipment limits its usage to laboratories. Here, we present a new disposable sensing platform that is based on a complementary metal oxide semiconductor process for total cholesterol quantification in pure blood serum. The platform that we implemented comprises readily mass-manufacturable components that exploit colorimetric changes of cholesterol oxidase and cholesterol esterase reactions. We have shown that our quantification results are comparable to that obtained by a bench top spectrophotometer. Using the implemented device, we have measured cholesterol concentration in human blood serum as low as 29 ÎźM with a limit of detection at 13 ÎźM, which is approximately 400 times lower than average physiological range, implying that our device also has the potential to be used for applications that require greater sensitivity

    Extreme magnetic field-boosted superconductivity

    Applied magnetic fields underlie exotic quantum states, such as the fractional quantum Hall effect and Bose-Einstein condensation of spin excitations. Superconductivity, on the other hand, is inherently antagonistic towards magnetic fields. Only in rare cases can these effects be mitigated over limited fields, leading to reentrant superconductivity. Here, we report the unprecedented coexistence of multiple high-field reentrant superconducting phases in the spin-triplet superconductor UTe2. Strikingly, we observe superconductivity in the highest magnetic field range identified for any reentrant superconductor, beyond 65 T. These extreme properties reflect a new kind of exotic superconductivity rooted in magnetic fluctuations and boosted by a quantum dimensional crossover

    Systematic review and meta-analysis of effects of community-delivered positive youth development interventions on violence outcomes

    Background We systematically reviewed and meta-analysed evaluations testing the effectiveness of positive youth development (PYD) interventions for reducing violence in young people. Methods Two reviewers working independently screened records, assessed full-text studies for inclusion and extracted data. Outcomes were transformed to Cohen's d. Quality assessment of included evaluations was undertaken using the Cochrane risk of bias tool. Effect sizes were combined using multilevel meta-analysis. We searched 21 databases, including MEDLINE, PsycINFO, CINAHL and CENTRAL, and hand-searched key journals and websites. We included studies where the majority of participants were aged 11–18 years and where interventions were delivered in community (not clinical or judicial) settings outside of normal school hours. We excluded studies targeting predefined physical and mental health conditions or parents/carers alongside young people. We defined violence as perpetration or victimisation of physical violence including violent crime. Results Three randomised trials were included in this systematic review. Included evaluations each had design flaws. Meta-analyses suggested that PYD interventions did not have a statistically significant effect on violence outcomes across all time points (d=0.021, 95% CI −0.050 to 0.093), though interventions did have a statistically significant short-term effect (d=0.076, 95% CI 0.013 to 0.140). Conclusions Our meta-analyses do not offer evidence of PYD interventions in general having effects of public health significance in reducing violence among young people. Evaluations did not consistently report theories of change or implementation fidelity, so it is unclear if our meta-analyses provide evidence that the PYD theory of change is ineffective in reducing violence among young people
