Habitual Choline Intakes across the Childbearing Years: A Review
Choline is an important nutrient during the first 1000 days post conception due to its roles
in brain function. An increasing number of studies have measured choline intakes at the population
level. We collated the evidence focusing on habitual choline intakes in the preconceptual, pregnancy,
and lactation life stages. We conducted a review including studies published from 2004 to 2021.
Twenty-six relevant publications were identified. After excluding studies with a high choline intake
(>400 mg/day; two studies) or low choline intake (<200 mg/day; one study), average choline intake
in the remaining 23 studies ranged from 233 mg/day to 383 mg/day, even with the inclusion of
choline from supplements. Intakes were not higher in studies among pregnant and lactating women
compared with studies in nonpregnant women. To conclude, during the childbearing years and
across the globe, habitual intakes of choline from foods alone and foods and supplements combined
appear to be consistently lower than the estimated adequate intakes for this target group. Urgent
measures are needed to (1) improve the quality of choline data in global food composition databases,
(2) encourage the reporting of choline intakes in dietary surveys, (3) raise awareness about the
role(s) of choline in foetalâmaternal health, and (4) consider formally advocating the use of choline
supplements in women planning a pregnancy, pregnant, or lactating
Pharmacokinetics of Sodium and Calcium Salts of (6S)-5-Methyltetrahydrofolic Acid Compared to Folic Acid and Indirect Comparison of the Two Salts
(6S)-5-Methyltetrahydrofolic acid ((6S)-5-Methyl-THF) salts and folic acid may differ in
their abilities to raise plasma (6S)-5-Methyl-THF levels. We compared the area under the curve
(AUC), Cmax, and Tmax of plasma (6S)-5-Methyl-THF after intakes of (6S)-5-Methyl-THF-Na salt
(ArcofolinÂź) and folic acid. Moreover, we compared the AUCs after intakes of (6S)-5-Methyl-THF-Na
and the calcium salt, (6S)-5-Methyl-THF-Ca, that were tested against folic acid in two independent
studies. The study was randomized, double blind, and cross over. Twenty-four adults (12 men and
12 women) received a single oral dose of 436 ”g (6S)-5-Methyl-THF-Na and an equimolar dose of
folic acid (400 ”g) on two kinetic days with two weeks washout period in between. The plasma
concentrations of (6S)-5-Methyl-THF were measured at 9 time points between 0 and 8 h. We found
that the AUC0â8 h of plasma (6S)-5-Methyl-THF (mean (SD) = 126.0 (33.6) vs. 56.0 (25.3) nmol/L*h) and
Cmax (36.8 (10.8) vs. 11.1 (4.1) nmol/L) were higher after administration of (6S)-5-Methyl-THF-Na than
after the administration of folic acid (p < 0.001 for both). These differences were present in men and
women. Only administration of folic acid resulted in a transient increase in plasma unmetabolized
folic acid (2.5 (2.0) nmol/L after 0.5 h and 4.7 (2.9) nmol/L after 1 h). Intake of (6S)-5-Methyl-THF-Na
was safe. The ratios of the AUC0â8 h for (6S)-5-Methyl-THF-Na and (6S)-5-Methyl-THF-Ca to the
corresponding folic acid reference group and the delta of these AUC0â8 h did not differ between the
studies. In conclusion, a single oral dose of (6S)-5-Methyl-THF-Na caused higher AUC0â8 h and Cmax of
plasma (6S)-5-Methyl-THF compared to folic acid. The Na- and Ca- salts of (6S)-5-Methyl-THF are not
likely to differ in their pharmacokinetics. Further studies may investigate whether supplementation
of the compounds for a longer time will lead to differences in circulating or intracellular/tissue
folate concentrations
Physiological effects of oral glucosamine on joint health: Current status and consensus on future research priorities
The aim of this paper was to provide an overview of the current knowledge and understanding of the potential
beneficial physiological effects of glucosamine (GlcN) on joint health. The objective was to reach a consensus on
four critical questions and to provide recommendations for future research priorities. To this end, nine scientists
from Europe and the United States were selected according to their expertise in this particular field and were
invited to participate in the Hohenheim conference held in August 2011. Each expert was asked to address a
question that had previously been posed by the chairman of the conference. Based on a systematic review of the
literature and the collection of recent data, the experts documented the effects of GlcN on cartilage ageing,
metabolic/kinetic and maintenance of joint health as well as reduction of risk of OA development. After extensive
debate and discussion the expert panel addressed each question and a general consensus statement was
developed, agreeing on the current state-of-the-art and future areas for basic and clinical studies. This paper
summarizes the available evidence for beneficial effects of GlcN on joint health and proposes new insight into the
design of future clinical trials aimed at identifying beneficial physiological effect of GlcN on joint tissuesHenrotin is the founder of Artialis SA a spin-off of the University of LiĂšge.

Effect of Two Different Multimicronutrient Supplements on Vitamin D Status in Women of Childbearing Age: A Randomized Trial
The German Nutrition Society raised in 2012 the recommended daily vitamin D intake from 200 to 800 international units (IU) to achieve 25-hydroxyvitamin D (25(OH)D) levels of at least 50 nmol/L, even when endogenous vitamin D synthesis is minimal such as in winter. We aimed to evaluate this recommendation in women of childbearing age. This is a single-center, randomized, open trial conducted from 8 January to 9 May 2016 in Esslingen, Germany. We randomized 201 apparently healthy women to receive for 8 weeks a daily multimicronutrient supplement containing either 200 IU (n = 100) or 800 IU vitamin D3 (n = 101). Primary outcome measure was serum 25(OH)D. 196 participants completed the trial. Increases in 25(OH)D (median with interquartile range) from baseline to study end were 13.2 (5.9 to 20.7) nmol/L in the 200 IU group, and 35.8 (18.2 to 52.8) nmol/L in the 800 IU group (p < 0.001 for the between group difference). At study end, levels of â„50 nmol/L were present in 70.4% of the 200 IU group and in 99% of the 800 IU group. Participants on hormonal contraceptives had higher baseline levels and a stronger increase in 25(OH)D. In conclusion, daily supplementation of 800 IU vitamin D3 during wintertime in Germany is sufficient to achieve a 25(OH)D level of at least 50 nmol/L in almost all women of childbearing age, whereas 200 IU are insufficient
Grape Seed Extract Positively Modulates Blood Pressure and Perceived Stress: A Randomized, Double-Blind, Placebo-Controlled Study in Healthy Volunteers
It is well established that maintaining healthy blood pressure is fundamental in order to avoid disorders to the heart and blood vessels. In prevention, and alongside pharmacological therapy, the use of natural substances has been proven to be extremely helpful for pre- and mild hypertensive subjects. Our study was therefore focused on the effects, both in vitro and in humans, of a grape seed extract, Enovita (GSEe), a highly standardized extract in polyphenols of Vitis vinifera L. The in vitro human umbilical vein endothelial cells (HUVEC) model was chosen to explore the extract properties related to vascular inflammation/vasodilation. A significant reduction of both soluble Inter-Cellular Adhesion Molecule-1 (sICAM) and endothelin-1 secretion/release was induced by GSEe in HUVEC cells. A randomized, double-blind, placebo-controlled clinical study in healthy volunteers was further performed to investigate GSEe benefits. In healthy volunteers, both supplementations significantly modulated blood pressure, with a pronounced effect after GSEe tablets (300 mg/day for 16 weeks) in respect to placebo. In the male gender subgroup, no placebo effect was observed as it was for the female group. As an additional outcome, an overall GSEe positive modulation emerged on mood related to stress perception. Thus, GSEe resulted in a benefit of modulating endothelial functionality and blood pressure. It was noteworthy that GSEe relieved the perceived stress, promising new future perspectives on mood comfort
