    How the 'seraphic' became 'geographic': women travellers in West Africa, 1840-1915

    This thesis brings together two important developments in contemporary geography; firstly, the recognition of the need to write critical histories of geographical thought and, particularly, the relationship between modern geography and European imperialism, and secondly, the attempt by feminist geographers to countervail the absence of women in these histories. Drawing on recent innovative attempts by geographers to construct alternative, contextual perspectives in (re)writing histories of geographical thought, the thesis analyzes the travel narratives of British women travellers in West Africa between 1840 and 1915. Recent attempts by feminists to include women in histories of geography and imperialism have, all too often, failed to analyze critically the role of women in imperial culture, or have reproduced gender dichotomies in their analysis. This thesis seeks to overcome these problems in three ways. Firstly, it explores the contributions of women travellers to imperial culture, primarily through their production of popular geographies. Secondly, it analyzes the ways in which these women were empowered in the imperial context by virtue of both race and class. Thirdly, it frames the accounts of each woman within the specific spatial and temporal context of their journeys in order to explore the complexities in the popular geographies they produced. The thesis illustrates that while gender was an important factor in the construction of images in the travel narratives of Victorian women travellers, this cannot be divorced from the wider context of their journey, nor from other elements in power relations based on difference such as race and class. Using this framework, the study explores in detail the production of popular geographies of the landscapes and peoples of West Africa by British women travellers, and formulates an argument on how women and their experiences can be included in histories of geographical thought

    Zinc disrupts central carbon metabolism and capsule biosynthesis in Streptococcus pyogenes.

    Neutrophils release free zinc to eliminate the phagocytosed bacterial pathogen Streptococcus pyogenes (Group A Streptococcus; GAS). In this study, we investigated the mechanisms underpinning zinc toxicity towards this human pathogen, responsible for diseases ranging from pharyngitis and impetigo, to severe invasive infections. Using the globally-disseminated M1T1 GAS strain, we demonstrate that zinc stress impairs glucose metabolism through the inhibition of the glycolytic enzymes phosphofructokinase and glyceraldehyde-3-phosphate dehydrogenase. In the presence of zinc, a metabolic shift to the tagatose-6-phosphate pathway allows conversion of D-galactose to dihydroxyacetone phosphate and glyceraldehyde phosphate, partially bypassing impaired glycolytic enzymes to generate pyruvate. Additionally, zinc inhibition of phosphoglucomutase results in decreased capsule biosynthesis. These data indicate that zinc exerts it toxicity via mechanisms that inhibit both GAS central carbon metabolism and virulence pathways

    Book Review: Limits to Decolonization: Indigeneity, Territory, and Hydrocarbon Politics in the Bolivian Chaco

    This book review symposium critically evaluates Penelope Anthias’ recent text Limits to Decolonization: Indigeneity, Territory, and Hydrocarbon Politics in the Bolivian Chaco (Cornell University Press 2018). Through deep ethnographic attention, Anthias’ text evaluates Indigenous struggles for territory in the context of “post-neoliberal” Bolivia under the Evo Morales administration, showing the variegated and nuanced politics of autonomy in an era of hydrocarbon extraction and increasingly contradictory state-Indigenous relations. The text examines the “limits” of rights and state-led territorial titling processes to radically challenge the racialized extractive geographies that shape the Bolivian Chaco region. In so doing, Anthias’ ethnography provides a rich analysis of how Guaraní Indigenous peoples are reshaping their relations with non-Indigenous landowners and the hydrocarbon industry to advance new forms of territorial autonomy and self-determination with significant ramifications on Indigenous studies in Latin America. This book review symposium draws from a session at the 2019 American Association of Geographers Conference, featuring two leading geographers who share their critical readings of Limits to Decolonization with a conclusion by Anthias that responds to the written reviews

    The Role of Copper and Zinc Toxicity in Innate Immune Defense against Bacterial Pathogens

    Zinc (Zn) and copper (Cu) are essential for optimal innate immune function, and nutritional deficiency in either metal leads to increased susceptibility to bacterial infection. Recently, the decreased survival of bacterial pathogens with impaired Cu and/or Zn detoxification systems in phagocytes and animal models of infection has been reported. Consequently, a model has emerged in which the host utilizes Cu and/or Zn intoxication to reduce the intracellular survival of pathogens. This review describes and assesses the potential role for Cu and Zn intoxication in innate immune function and their direct bactericidal function

    The role of zinc acquisition and zinc tolerance in group A streptococcal infection

    Zinc plays an important role in host innate immune function. However, the innate immune system also utilizes zinc starvation ('nutritional immunity') to combat infections. Here, we investigate the role of zinc import and export in protection of(Group A; GAS), a Gram-positive bacterial pathogen responsible for a wide spectrum of human diseases, against challenge from host innate immune defence. In order to determine the role of GAS zinc import and export during infection, we utilized the zinc import (Δ) and export (Δ) deletion mutants in competition with wild-type in bothandvirulence models. We demonstrate that nutritional immunity is deployed extracellularly while zinc toxicity is utilized upon phagocytosis of GAS by neutrophils. We also show that lysosomes and azurophilic granules in neutrophils contain zinc stores for use against intracellular pathogens

    Benchmarking the prevalence of care problems in UK care homes using the LPZ-i: a feasibility study

    Background: UK care homes have no mechanism for counting, collating or analyzing prevalence of care problems to benchmark quality of care. This study investigated the feasibility of the International Prevalence Measurement of Quality of Care (LPZ-i) for benchmarking in UK care homes and described facilitators and challenges to implementation. Method: The pressure care and incontinence modules of the LPZ-i were implemented in a purposive sample of UK care homes. Interviews and focus groups with regional co-ordinators, NHS and care home staff who had been involved in implementation. Data were analysed using a framework approach. Results: 26 care homes and 489 residents implemented the LPZ-I modules. 60 pressure ulcers were reported in 41 (8.4%) of residents.325(66% of residents were incontinent of urine, faeces or both. It was demonstrated that it was possible to use prevalence data to benchmark care between UK care homes and against homes overseas. 38 participants took part in focus groups and interviews which revealed the importance of existing relational working with care homes as a basis for implementation. Care homes reported improved identification and management of pressure ulcers and incontinence in response to the audit. However, care home staff could not interpret or analyse data from the audit or use it to drive quality improvement. Conclusion: Implementing the LPZ-i was possible, delivered data appropriate for benchmarking, was well received by care home and NHS staff, and appeared to lead to changes in practice. More widespread implementation of the LPZ-i is feasible, subject to improving the data and quality improvement competencies amongst care home staff

    Chemical Synergy between Ionophore PBT2 and Zinc Reverses Antibiotic Resistance.

    The World Health Organization reports that antibiotic-resistant pathogens represent an imminent global health disaster for the 21st century. Gram-positive superbugs threaten to breach last-line antibiotic treatment, and the pharmaceutical industry antibiotic development pipeline is waning. Here we report the synergy between ionophore-induced physiological stress in Gram-positive bacteria and antibiotic treatment. PBT2 is a safe-for-human-use zinc ionophore that has progressed to phase 2 clinical trials for Alzheimer's and Huntington's disease treatment. In combination with zinc, PBT2 exhibits antibacterial activity and disrupts cellular homeostasis in erythromycin-resistant group A Streptococcus (GAS), methicillin-resistant Staphylococcus aureus (MRSA), and vancomycin-resistant Enterococcus (VRE). We were unable to select for mutants resistant to PBT2-zinc treatment. While ineffective alone against resistant bacteria, several clinically relevant antibiotics act synergistically with PBT2-zinc to enhance killing of these Gram-positive pathogens. These data represent a new paradigm whereby disruption of bacterial metal homeostasis reverses antibiotic-resistant phenotypes in a number of priority human bacterial pathogens.IMPORTANCE The rise of bacterial antibiotic resistance coupled with a reduction in new antibiotic development has placed significant burdens on global health care. Resistant bacterial pathogens such as methicillin-resistant Staphylococcus aureus and vancomycin-resistant Enterococcus are leading causes of community- and hospital-acquired infection and present a significant clinical challenge. These pathogens have acquired resistance to broad classes of antimicrobials. Furthermore, Streptococcus pyogenes, a significant disease agent among Indigenous Australians, has now acquired resistance to several antibiotic classes. With a rise in antibiotic resistance and reduction in new antibiotic discovery, it is imperative to investigate alternative therapeutic regimens that complement the use of current antibiotic treatment strategies. As stated by the WHO Director-General, "On current trends, common diseases may become untreatable. Doctors facing patients will have to say, Sorry, there is nothing I can do for you.
