    Increased human pathogenic potential of Escherichia coli from polymicrobial urinary tract infections in comparison to isolates from monomicrobial culture samples

    The current diagnostic standard procedure outlined by the Health Protection Agency for urinary tract infections (UTIs) in clinical laboratories does not report bacteria isolated from samples containing three or more different bacterial species. As a result many UTIs go unreported and untreated, particularly in elderly patients, where polymicrobial UTI samples are especially prevalent. This study reports the presence of the major uropathogenic species in mixed culture urine samples from elderly patients, and of resistance to front-line antibiotics, with potentially increased levels of resistance to ciprofloxacin and trimethoprim. Most importantly, the study highlights that Escherichia coli present in polymicrobial UTI samples are statistically more invasive (P<0.001) in in vitro epithelial cell infection assays than those isolated from monomicrobial culture samples. In summary, the results of this study suggest that the current diagnostic standard procedure for polymicrobial UTI samples needs to be reassessed, and that E. coli present in polymicrobial UTI samples may pose an increased risk to human health

    Near-IR imaging of T Cha: evidence for scattered-light disk structures at solar system scales

    T Chamaeleontis is a young star surrounded by a transitional disk, and a plausible candidate for ongoing planet formation. Recently, a substellar companion candidate was reported within the disk gap of this star. However, its existence remains controversial, with the counter-hypothesis that light from a high inclination disk may also be consistent with the observed data. The aim of this work is to investigate the origin of the observed closure phase signal to determine if it is best explained by a compact companion. We observed T Cha in the L and K s filters with sparse aperture masking, with 7 datasets covering a period of 3 years. A consistent closure phase signal is recovered in all L and K s datasets. Data were fit with a companion model and an inclined circumstellar disk model based on known disk parameters: both were shown to provide an adequate fit. However, the absence of expected relative motion for an orbiting body over the 3-year time baseline spanned by the observations rules out the companion model. Applying image reconstruction techniques to each dataset reveals a stationary structure consistent with forward scattering from the near edge of an inclined disk.Comment: 6 pages, 3 figures, accepted for publication in MNRAS Letter

    DamID as a versatile tool for understanding gene regulation

    The interaction of proteins and RNA with chromatin underlies the regulation of gene expression. The ability to profile easily these interactions is fundamental for understanding chromatin biology in vivo. DNA adenine methyltransferase identification (DamID) profiles genome-wide protein-DNA interactions without antibodies, fixation or protein pull-downs. Recently, DamID has been adapted for applications beyond simple assaying of protein-DNA interactions, such as for studying RNA-chromatin interactions, chromatin accessibility and long-range chromosome interactions. Here, we provide an overview of DamID and introduce improvements to the technology, discuss their applications and compare alternative methodologies

    A longitudinal, observational study examining the relationships of patient satisfaction with services and mental well-being to their clinical course in young people with Type 1 diabetes mellitus during transition from child to adult health services

    AIM: We hypothesized that participant well-being and satisfaction with services would be positively associated with a satisfactory clinical course during transition from child to adult health care. METHODS: Some 150 young people with Type 1 diabetes mellitus from five diabetes units in England were recruited to a longitudinal study of transition. Each young person was visited at home four times by a research assistant; each visit was 1 year apart. Satisfaction with services (Mind the Gap; MTG) and mental well-being (Warwick-Edinburgh Mental Well-being Scale; WEMWBS) were captured. Change in HbA1c , episodes of ketoacidosis, clinic and retinal screening attendance were used to assess clinical course. In total, 108 of 150 (72%) young people had sufficient data for analysis at visit 4. RESULTS: Mean age at entry was 16 years. By visit 4, 81.5% had left paediatric healthcare services. Median HbA1c increased significantly (P = 0.01) from 69 mmol/mol (8.5%) at baseline to 75 mmol/mol (9.0%) at visit 4. WEMWBS scores were comparable with those in the general population at baseline and were stable over the study period. MTG scores were also stable. By visit 4, some 32 individuals had a 'satisfactory' and 76 a 'suboptimal' clinical course. There were no significant differences in average WEMWBS and MTG scores between the clinical course groups (P = 0.96, 0.52 respectively); nor was there a significant difference in transfer status between the clinical course groups. CONCLUSIONS: The well-being of young people with diabetes and their satisfaction with transition services are not closely related to their clinical course. Investigating whether innovative psycho-educational interventions can improve the clinical course is a research priority

    Discovery of Seven Companions To Intermediate-Mass Stars With Extreme Mass Ratios in the Scorpius-Centaurus Association

    We report the detection of seven low-mass companions to intermediate-mass stars (SpT B/A/F; M similar to 1.5-4.5M(circle dot)) in the Scorpius-Centaurus (Sco-Cen) Association using nonredundant aperture masking interferometry. Our newly detected objects have contrasts Delta L' approximate to 4-6, corresponding to masses as low as similar to 20 M-Jup and mass ratios of q approximate to 0.01-0.08, depending on the assumed age of the target stars. With projected separations rho approximate to 10-30 AU, our aperture masking detections sample an orbital region previously unprobed by conventional adaptive optics imaging of intermediate-mass Sco-Cen stars covering much larger orbital radii (similar to 30-3000 AU). At such orbital separations, these objects resemble higher-mass versions of the directly imaged planetary mass companions to the 10-30 Myr, intermediate-mass stars HR 8799, beta Pictoris, and HD 95086.     Open-framework zinc and cobalt phosphates synthesized by the tributylphosphate route

    By employing tributylphosphate as the source of phosphorus, several open-framework zinc and cobalt phosphates have been prepared hydrothermally. Of the three new zinc phosphates [C6N2H18][Zn(HPO4)2], I, has a linear chain structure while [C4N2H14][Zn5(PO4)4(H2O)], II, [NH4][H3O][Zn4(PO4)3]2·H2O, III, have three-dimensional structures. Of the three new cobalt phosphates described, [C5N2H14][Co(HPO4)2], IV, and [C5N2H14][Co(HPO4)2], V, have linear chain structures, while [C4N2H12]3 [Co2(OH)(HPO4)3]2, VI, has a complex double chain structure.     Optical Studies of Er-doped Yttrium Aluminium Garnet Phosphor Materials

    The need for materials application in solid-state lasers, medical devices, and optoelectronic devices has made the investigation of ceramic materials of increasing importance. A detail study of the optical properties of rare earth element typically from luminescent materials when intentionally doped inside the host materials and in particular crystal (such as YAG) is reported for the photoluminescence, power and lifetime measurement. The rare-earth dopants usually form trivalent lanthanide ions and the energy transfer and optical transitions involved originate from 4f-4f transitions of the ions and between these states and the host material. In order to understand the energy transfer processes in more detail we need to better understand the accompanying optical processes that give rise to the emission they display and it is this that forms the focus of the work presented. Following this second (and higher) order processes are considered that lead to upconversion in erbium-doped yttrium aluminum garnet (Er:YAG) materials
