    Anti-Infective Mechanism-Based Drug Discovery via Sortase A

    Sortase A is a transmembrane protein prominent in gram-negative bacterial strains. It is a virulence factor that anchors other proteins, which facilitate MRSA infections. In the long term, we plan to utilize this protein to create anti-infective drugs as antibiotic resistance continues to become a global health issue. Starting with five NIH drug candidates, we decided to first study Sortase A with B12 due to its historical past of ancient civilizations using natural sources of B12 to treat infections. Our senior design is further centered around understanding how vitamin B12 interacts with Sortase A in vitro, particularly the binding affinity between the two. The in vitro experiments will verify the previous in vivo studies

    Bifurcation in the history of Uranus and Neptune: the role of giant impacts

    Despite many similarities, there are significant observed differences between Uranus and Neptune: while Uranus is tilted and has a regular set of satellites, suggesting their accretion from a disk, Neptune’s moons are irregular and are captured objects. In addition, Neptune seems to have an internal heat source, while Uranus is in equilibrium with solar insulation. Finally, structure models based on gravity data suggest that Uranus is more centrally condensed than Neptune. We perform a large suite of high resolution SPH simulations to investigate whether these differences can be explained by giant impacts. For Uranus, we find that an oblique impact can tilt its spin axis and eject enough material to create a disk where the regular satellites are formed. Some of the disks are massive and extended enough, and consist of enough rocky material to explain the formation of Uranus’ regular satellites. For Neptune, we investigate whether a head-on collision could mix the interior, and lead to an adiabatic temperature profile, which may explain its larger flux and higher moment of inertia value. We find that massive and dense projectiles can penetrate towards the centre and deposit mass and energy in the deep interior, leading to a less centrally concentrated interior for Neptune. We conclude that the dichotomy between the ice giants can be explained by violent impacts after their formation

    Availability, Use, and Cultivation of Support Networks as Predictors of the Well-Being of Middle-Aged and Older Chinese: A Panel Study

    Objectives. To examine the impact of the availability, use, and cultivation of a support network on the well-being of community-dwelling, middle-aged, and older Chinese. Methods. A total of 2,970 Hong Kong Chinese aged 40–74 years were interviewed using a structured questionnaire in 2004. Out of the original group of interviewees, 2,120 (71.4%) were interviewed again in 2005. Results. Structural equation modeling revealed a good fit of the model employing Wave 1 support network data and demographic characteristics to predict Wave 2 well-being. As hypothesized, the availability of important social ties and the cultivation of one's support networks were found to predict well-being one year later, but not the use of support networks to meet emotional, financial, or companion needs after controlling for demographic variables and baseline well-being. Discussion. Cultivating support networks can be interpreted as positive and active coping. Such cultivation is in line with what socioemotional selectivity theory predicts; specifically, when people age, they become more selective and concentrate on strengthening their relationship with those they are emotionally close to. We argue that network cultivation deserves more attention in theory, practice, and research to strengthen the resilience and adaptability of individuals approaching and experiencing old age

    Did Uranus' regular moons form via a rocky giant impactor?

    The formation of Uranus' regular moons has been suggested to be linked to the origin of its enormous spin axial tilt (~98^o). A giant impact between proto-Uranus and a 2-3 M_Earth impactor could lead to a large tilt and to the formation of an impact generated disc, where prograde and circular satellites are accreted. The most intriguing features of the current regular Uranian satellite system is that it possesses a positive trend in the mass-distance distribution and likely also in the bulk density, implying that viscous spreading of the disc after the giant impact plays a crucial role in shaping the architecture of the final system. In this paper, we investigate the formation of Uranus' satellites by combining results of SPH simulations for the giant impact, a 1D semi-analytic disc model for viscous spreading of the post-impact disc, and N-body simulations for the assembly of satellites from a disc of moonlets. Assuming the condensed rock (i.e., silicate) remains small and available to stick onto the relatively rapid growing condensed water-ice, we find that the best case in reproducing the observed mass and bulk composition of Uranus' satellite system is a pure-rocky impactor with 3 M_Earth colliding with the young Uranus with an impact parameter b = 0.75. Such an oblique collision could also naturally explain Uranus' large tilt and possibly, its low internal heat flux. The giant impact scenario can naturally explain the key features of Uranus and its regular moons. We therefore suggest that the Uranian satellite system formed as a result of an impact rather than from a circumplanetary disc.Comment: Accepted for publication in Icaru

    High expression FUT1 and B3GALT5 is an independent predictor of postoperative recurrence and survival in hepatocellular carcinoma.

    Cancer may arise from dedifferentiation of mature cells or maturation-arrested stem cells. Previously we reported that definitive endoderm from which liver was derived, expressed Globo H, SSEA-3 and SSEA-4. In this study, we examined the expression of their biosynthetic enzymes, FUT1, FUT2, B3GALT5 and ST3GAL2, in 135 hepatocellular carcinoma (HCC) tissues by qRT-PCR. High expression of either FUT1 or B3GALT5 was significantly associated with advanced stages and poor outcome. Kaplan Meier survival analysis showed significantly shorter relapse-free survival (RFS) for those with high expression of either FUT1 or B3GALT5 (P = 0.024 and 0.001, respectively) and shorter overall survival (OS) for those with high expression of B3GALT5 (P = 0.017). Combination of FUT1 and B3GALT5 revealed that high expression of both genes had poorer RFS and OS than the others (P < 0.001). Moreover, multivariable Cox regression analysis identified the combination of B3GALT5 and FUT1 as an independent predictor for RFS (HR: 2.370, 95% CI: 1.505-3.731, P < 0.001) and OS (HR: 2.153, 95% CI: 1.188-3.902, P = 0.012) in HCC. In addition, the presence of Globo H, SSEA-3 and SSEA-4 in some HCC tissues and their absence in normal liver was established by immunohistochemistry staining and mass spectrometric analysis

    Short version of the Smartphone Addiction Scale in Chinese adults: Psychometric properties, sociodemographic, and health behavioral correlates

    Background and aims: Problematic smartphone use (PSU) is an emerging but understudied public health issue. Little is known about the epidemiology of PSU at the population level. We evaluated the psychometric properties of the Smartphone Addiction Scale – Short Version (SAS-SV) and examined its associated sociodemographic factors and health behaviors in Chinese adults in Hong Kong. Methods: A random sample of 3,211 adults aged ≥18 years (mean ± SD: 43.3 ± 15.7, 45.3% men) participated in a population-based telephone survey in Hong Kong and completed the Chinese SAS-SV. Multivariable linear regressions examined the associations of sociodemographic factors, health behaviors, and chronic disease status with SAS-SV score. Data were weighted by age, sex, and education attainment distributions of the Hong Kong general population. Results: The Chinese SAS-SV is internally consistent (Cronbach’s α = .844) and stable over 1 week (intraclass correlation coefficient = .76, p < .001). Confirmatory factor analysis supported a unidimensional structure established by previous studies. The weighted prevalence of PSU was 38.5% (95% confidence interval: 36.9%, 40.2%). Female sex, younger age, being married/cohabitated or divorced/separated (vs. unmarried), and lower education level were associated with a higher SAS-SV score (all ps <.05). Current smoking, weekly to daily alcohol drinking, and physical inactivity predict greater PSU after controlling for sociodemographic factors and mutual adjustment. Discussion and conclusions: The Chinese SAS-SV was found valid and reliable for assessing PSU in Hong Kong adults. Several sociodemographic and health behavioral factors were associated with PSU at the population level, which may have implication for prevention of PSU and future research

    Understanding Dwarf Galaxies in order to Understand Dark Matter

    Much progress has been made in recent years by the galaxy simulation community in making realistic galaxies, mostly by more accurately capturing the effects of baryons on the structural evolution of dark matter halos at high resolutions. This progress has altered theoretical expectations for galaxy evolution within a Cold Dark Matter (CDM) model, reconciling many earlier discrepancies between theory and observations. Despite this reconciliation, CDM may not be an accurate model for our Universe.
