    Two-Dimensional Laser-Doppler Velocimetry in Turbulent Flows

    Localized measurement of the velocity of flow In fluids may be accomplished by detecting the Doppler shift In frequency from coherent, monochromatic light scattered from micrometer sized contaminant particles within the flow. The versatility of applications of this technique, first used several years ago, has been expanded by recent research. The basic geometry of measurement, in which the scattered energy is recombined with light energy at the original frequency on the face of the photocathode of a photomultiplier tube, can be rearranged in many alternative configurations to meet the needs of the experimenter. By using a single laser, additional arrangements are possible for obtaining two- or three-dimensional measurements of velocity components. Several different velocimeters are described for making two-dimensional flow measurements, including both backscatter and forward scatter systems. Effects of flow system geometries on capabilities for measuring specific components are investigated. Effects of laser beam polarization are discussed and conclusions reached on methods of optimizing signal strength in each of two orthogonal measurement systems. Results of the application of a two-dimensional measurement system used for obtaining velocity profiles in turbulent flow in a smooth walled four-inch diameter pipe are presented. Operating at a Reynolds number of about 1.1 x 106, relative turbulent intensities were measured in the axial direction and normal to that direction. Standard techniques were utilized. A second readout system was employed to make a measurement of this parameter without the requirement of adding scattering centers to the flow

It seeks to identify the factors contributing to successful management of air quality and the factors that have acted, or continue to do so, as barriers to progress. The public health catastrophe of the 1952 London Smog created the political momentum for the 1956 Act to be passed. The nature of the contemporary air pollution challenge is reviewed in terms of the public health burden, the economic cost and the governmental response. The contemporary response is considered inadequate for the scale and intensity of the problem

    A GIS-based assessment of the suitability of SCIAMACHY satellite sensor measurements for estimating reliable CO concentrations in a low-latitude climate

    An assessment of the reliability of the Scanning Imaging Absorption Spectrometer for Atmospheric Cartography (SCIAMACHY) satellite sensor measurements to interpolate tropospheric concentrations of carbon monoxide considering the lowlatitude climate of the Niger Delta region in Nigeria was conducted. Monthly SCIAMACHY carbon monoxide (CO) column measurements from January 2,003 to December 2005 were interpolated using ordinary kriging technique. The spatio-temporal variations observed in the reliability were based on proximity to the Atlantic Ocean, seasonal variations in the intensities of rainfall and relative humidity, the presence of dust particles from the Sahara desert, industrialization in Southwest Nigeria and biomass burning during the dry season in Northern Nigeria. Spatial reliabilities of 74 and 42 % are observed for the inland and coastal areas, respectively. Temporally, average reliability of 61 and 55%occur during the dry and wet seasons, respectively. Reliability in the inland and coastal areas was 72 and 38% during the wet season, and 75 and 46% during the dry season, respectively. Based on the results, the WFM-DOAS SCIAMACHY CO data product used for this study is therefore relevant in the assessment of CO concentrations in developing countries within the low latitudes that could not afford monitoring infrastructure due to the required high costs. Although the SCIAMACHY sensor is no longer available, it provided cost-effective, reliable and accessible data that could support air quality assessment in developing countries

    Validation of a commercially available indirect assay for SARS-CoV-2 neutralising antibodies using a pseudotyped virus assay.

    Objectives To assess whether a commercially available CE-IVD, ELISA-based surrogate neutralisation assay (cPass, Genscript) provides a genuine measure of SARS-CoV-2 neutralisation by human sera, and further to establish whether measuring responses against the RBD of S was a diagnostically useful proxy for responses against the whole S protein. Methods Serum samples from 30 patients were assayed for anti-NP responses, for ‘neutralisation’ by the surrogate neutralisation assay and for neutralisation by SARS-CoV-2 S pseudotyped virus assays utilising two target cell lines. Correlation between assays was measured using linear regression. Results The responses observed within the surrogate neutralisation assay demonstrated an extremely strong, highly significant positive correlation with those observed in both pseudotyped virus assays. Conclusions The tested ELISA-based surrogate assay provides an immunologically useful measure of functional immune responses in a much quicker and highly automatable fashion. It also reinforces that detection of anti-RBD neutralising antibodies alone is a powerful measure of the capacity to neutralise viral infection

    Protective effects of transforming growth factor β2 in intestinal epithelial cells by regulation of proteins associated with stress and endotoxin responses

    Transforming growth factor (TGF)-β2 is an important anti-inflammatory protein in milk and colostrum. TGF-β2 supplementation appears to reduce gut inflammatory diseases in early life, such as necrotizing enterocolitis (NEC) in young mice. However, the molecular mechanisms by which TGF-β2 protects immature intestinal epithelial cells (IECs) remain to be more clearly elucidated before interventions in infants can be considered. Porcine IECs PsIc1 were treated with TGF-β2 and/or lipopolysaccharide (LPS), and changes in the cellular proteome were subsequently analyzed using two-dimensional gel electrophoresis-MS and LC-MS-based proteomics. TGF-β2 alone induced the differential expression of 13 proteins and the majority of the identified proteins were associated with stress responses, TGF-β and Toll-like receptor 4 signaling cascades. In particular, a series of heat shock proteins had similar differential trends as previously shown in the intestine of NEC-resistant preterm pigs and young mice. Furthermore, LC-MS-based proteomics and Western blot analyses revealed 20 differentially expressed proteins following treatment with TGF-β2 in LPS-challenged IECs. Thirteen of these proteins were associated with stress response pathways, among which five proteins were altered by LPS and restored by TGF-β2, whereas six were differentially expressed only by TGF-β2 in LPS-challenged IECs. Based on previously reported biological functions, these patterns indicate the anti-stress and anti-inflammatory effects of TGF-β2 in IECs. We conclude that TGF-β2 of dietary or endogenous origin may regulate the IEC responses against LPS stimuli, thereby supporting cellular homeostasis and innate immunity in response to bacterial colonization, and the first enteral feeding in early life

    Necrotizing enterocolitis is associated with acute brain responses in preterm pigs

    BACKGROUND: Necrotizing enterocolitis (NEC) is an acute gut inflammatory disorder that occurs in preterm infants in the first weeks after birth. Infants surviving NEC often show impaired neurodevelopment. The mechanisms linking NEC lesions with later neurodevelopment are poorly understood but may include proinflammatory signaling in the immature brain. Using preterm pigs as a model for preterm infants, we hypothesized that severe intestinal NEC lesions are associated with acute effects on the developing hippocampus. METHODS: Cesarean-delivered preterm pigs (n = 117) were reared for 8 days and spontaneously developed variable severity of NEC lesions. Neonatal arousal, physical activity, and in vitro neuritogenic effects of cerebrospinal fluid (CSF) were investigated in pigs showing NEC lesions in the colon (Co-NEC) or in the small intestine (Si-NEC). Hippocampal transcriptome analysis and qPCR were used to assess gene expressions and their relation to biological processes, including neuroinflammation, and neural plasticity. Microglia activation was quantified by stereology. The neuritogenic response to selected proteins was investigated in primary cultures of hippocampal neurons. RESULTS: NEC development rapidly reduced the physical activity of pigs, especially when lesions occurred in the small intestine. Si-NEC and Co-NEC were associated with 27 and 12 hippocampal differentially expressed genes (DEGs), respectively. These included genes related to neuroinflammation (i.e., S100A8, S100A9, IL8, IL6, MMP8, SAA, TAGLN2) and hypoxia (i.e., PDK4, IER3, TXNIP, AGER), and they were all upregulated in Si-NEC pigs. Genes related to protection against oxidative stress (HBB, ALAS2) and oligodendrocytes (OPALIN) were downregulated in Si-NEC pigs. CSF collected from NEC pigs promoted neurite outgrowth in vitro, and the S100A9 and S100A8/S100A9 proteins may mediate the neuritogenic effects of NEC-related CSF on hippocampal neurons. NEC lesions did not affect total microglial cell number but markedly increased the proportion of Iba1-positive amoeboid microglial cells. CONCLUSIONS: NEC lesions, especially when present in the small intestine, are associated with changes to hippocampal gene expression that potentially mediate neuroinflammation and disturbed neural circuit formation via enhanced neuronal differentiation. Early brain-protective interventions may be critical for preterm infants affected by intestinal NEC lesions to reduce their later neurological dysfunctions

    Development and annotation of perennial Triticeae ESTs and SSR markers.

    Triticeae contains hundreds of species of both annual and perennial types. Although substantial genomic tools are available for annual Triticeae cereals such as wheat and barley, the perennial Triticeae lack sufficient genomic resources for genetic mapping or diversity research. To increase the amount of sequence information available in the perennial Triticeae, three expressed sequence tag (EST) libraries were developed and annotated for Pseudoroegneria spicata, a mixture of both Elymus wawawaiensis and E. lanceolatus, and a Leymus cinereus x L. triticoides interspecific hybrid. The ESTs were combined into unigene sets of 8 780 unigenes for P. spicata, 11 281 unigenes for Leymus, and 7 212 unigenes for Elymus. Unigenes were annotated based on putative orthology to genes from rice, wheat, barley, other Poaceae, Arabidopsis, and the non-redundant database of the NCBI. Simple sequence repeat (SSR) markers were developed, tested for amplification and polymorphism, and aligned to the rice genome. Leymus EST markers homologous to rice chromosome 2 genes were syntenous on Leymus homeologous groups 6a and 6b (previously 1b), demonstrating promise for in silico comparative mapping.
