    A discrete element model to investigate sub-surface damage due to surface polishing

    International audienceLarge high-power laser facilities such as megajoule laser (LMJ) or National Ignition Facility (NIF) are designed to focus about 2 MJ of energy at the wavelength of 351 nm, in the center of an experiment chamber. The final optic assembly of these systems, operating at 351 nm is made of large fused silica optics working in transmission. When submitted to laser at the wavelength of 351 nm, fused silica optics can exhibit damage, induced by the high amount of energy traversing the part. The created damage is a set of micro-chips that appear on the optic surface. Current researches have shown that this damage could be initiated on pre-existing sub-surface damages created during the optics manufacturing process. It is then very important to understand, for various set of manufacturing parameters, what are the key parameters for sub-surface damage. The presented work details the development of a simplified model to investigate the polishing process. Both silica (the material to be polished) and the abrasive particles are modeled using a discrete element approach. This numerical tool allows following the evolution of micro-cracks inside the material during the abrasion process. It is shown how the mechanical properties (pressure), the abrasive properties (shape and quantity of abrasive particles) and the system properties (filtration) have an influence on the sub-surface properties at the end of the process

    Non-Peer ReviewedDue to the advent of relatively cheap chemical N fertilizers, the use of livestock manure as a source of nutrients to crops was downplayed (James, et al., 1996). However, the expansion of the livestock industry, the need to utilize of the manure in an environmentally friendly and economically viable manner, and the desire to minimize the use of chemical N fertilizers have rekindled interest in the use of livestock manure as a fertilizer. Of particular interest is the need to utilize manure from large-scale intensive livestock operations (Chang and Janzen, 1996). This report is part of an on-going study initiated in 1996 to examine the soil and crop response to application of liquid swine manure and solid feedlot cattle manure at different rates and methods of application. Furthermore, the study seeks to evaluate nutrient forms and amounts in the manure and the effect of rate and method of manure application on soil fertility, nutrient utilization and crop yield. This paper puts together and summarizes the results of crop response to annual application of the two types of manure compared to that of urea fertilizer observed over the past four years, 1997 to 2000

    The changing paradigm of intron retention: regulation, ramifications and recipes

    Intron retention (IR) is a form of alternative splicing that has long been neglected in mammalian systems although it has been studied for decades in non-mammalian species such as plants, fungi, insects and viruses. It was generally assumed that mis-splicing, leading to the retention of introns, would have no physiological consequence other than reducing gene expression by nonsense-mediated decay. Relatively recent landmark discoveries have highlighted the pivotal role that IR serves in normal and disease-related human biology. Significant technical hurdles have been overcome, thereby enabling the robust detection and quantification of IR. Still, relatively little is known about the cis- and trans-acting modulators controlling this phenomenon. The fate of an intron to be, or not to be, retained in the mature transcript is the direct result of the influence exerted by numerous intrinsic and extrinsic factors at multiple levels of regulation. These factors have altered current biological paradigms and provided unexpected insights into the transcriptional landscape. In this review, we discuss the regulators of IR and methods to identify them. Our focus is primarily on mammals, however, we broaden the scope to non-mammalian organisms in which IR has been shown to be biologically relevant

    Cryptic photosynthesis, Extrasolar planetary oxygen without a surface biological signature

    On the Earth, photosynthetic organisms are responsible for the production of virtually all of the oxygen in the atmosphere. On the land, vegetation reflects in the visible, leading to a red edge that developed about 450 Myr ago and has been proposed as a biosignature for life on extrasolar planets. However, in many regions of the Earth, and particularly where surface conditions are extreme, for example in hot and cold deserts, photosynthetic organisms can be driven into and under substrates where light is still sufficient for photosynthesis. These communities exhibit no detectable surface spectral signature to indicate life. The same is true of the assemblages of photosynthetic organisms at more than a few metres depth in water bodies. These communities are widespread and dominate local photosynthetic productivity. We review known cryptic photosynthetic communities and their productivity. We link geomicrobiology with observational astronomy by calculating the disk-averaged spectra of cryptic habitats and identifying detectable features on an exoplanet dominated by such a biota. The hypothetical cryptic photosynthesis worlds discussed here are Earth-analogs that show detectable atmospheric biomarkers like our own planet, but do not exhibit a discernable biological surface feature in the disc-averaged spectrum.Comment: 23 pages, 2 figures, Astrobiology (TBP) - updated Table 1, typo in detectable O2 correcte

    Hip fracture in the elderly multidisciplinary rehabilitation (FEMuR) feasibility study: testing the use of routinely collected data for future health economic evaluations

    Background: Health economic evaluations rely on the accurate measurement of health service resource use in order to calculate costs. These are usually measured with patient completed questionnaires using instruments such as the Client Service Receipt Inventory (CSRI). These rely on participants' recall and can be burdensome to complete. Health service activity data are routinely captured by electronic databases.The aim was to test methods for obtaining these data and compare with those data collected using the CSRI, within a feasibility study of an enhanced rehabilitation intervention following hip fracture (Fracture in the Elderly Multidisciplinary Rehabilitation: FEMuR). Methods: Primary care activity including prescribing data was obtained from the Secure Anonymised Information Linkage (SAIL) Databank and secondary care activity (Emergency Department attendances, out-patient visits and in-patient days) directly from Betsi Cadwaladr University Health Board (BCUHB), North Wales, UK. These data were compared with patient responses from the CSRI using descriptive statistics and the intraclass correlation coefficient (ICC). Results: It was possible to compare health service resource use data for 49 out of 61 participants in the FEMuR study. For emergency department (ED) attendances, records matched in 23 (47%) cases, 21 (43%) over-reported on electronic records compared with CSRI and five participants (10%) under-reported, with an overall ICC of 0.42. For out-patient episodes, records matched in only six cases, 28 participants over-reported on electronic records compared with CSRI and 15 (12%) under-reported, with an overall ICC of only 0.27. For in-patient days, records matched exactly in only five cases (10%), but if an error margin of 7 days was allowed, then agreement rose to 39 (66%) cases, and the overall ICC for all data was 0.88.It was only possible to compare prescribing data for 12 participants. For prescribing data, the SAIL data reported 117 out of 118 items (99%) and the CSRI only 89 (79%) items. Conclusions: The use of routinely collected data has the potential to improve the efficiency of trials and other studies. Although the methodology to make the data available has been demonstrated, the data obtained was incomplete and the validity of using this method remains to be demonstrated.     We use SLE(6) paths to construct a process of continuum nonsimple loops in the plane and prove that this process coincides with the full continuum scaling limit of 2D critical site percolation on the triangular lattice -- that is, the scaling limit of the set of all interfaces between different clusters.     Fast Non-Adiabatic Two Qubit Gates for the Kane Quantum Computer

    In this paper we apply the canonical decomposition of two qubit unitaries to find pulse schemes to control the proposed Kane quantum computer. We explicitly find pulse sequences for the CNOT, swap, square root of swap and controlled Z rotations. We analyze the speed and fidelity of these gates, both of which compare favorably to existing schemes. The pulse sequences presented in this paper are theoretically faster, higher fidelity, and simpler than existing schemes. Any two qubit gate may be easily found and implemented using similar pulse sequences. Numerical simulation is used to verify the accuracy of each pulse scheme

    Ctcf haploinsufficiency mediates intron retention in a tissue-specific manner

    CTCF is a master regulator of gene transcription and chromatin organisation with occupancy at thousands of DNA target sites genome-wide. While CTCF is essential for cell survival, CTCF haploinsufficiency is associated with tumour development and hypermethylation. Increasing evidence demonstrates CTCF as a key player in several mechanisms regulating alternative splicing (AS), however, the genome-wide impact of Ctcf dosage on AS has not been investigated. We examined the effect of Ctcf haploinsufficiency on gene expression and AS in five tissues from Ctcf hemizygous (Ctcf+/-) mice. Reduced Ctcf levels caused distinct tissue-specific differences in gene expression and AS in all tissues. An increase in intron retention (IR) was observed in Ctcf+/- liver and kidney. In liver, this specifically impacted genes associated with cytoskeletal organisation, splicing and metabolism. Strikingly, most differentially retained introns were short, with a high GC content and enriched in Ctcf binding sites in their proximal upstream genomic region. This study provides new insights into the effects of CTCF haploinsufficiency on organ transcriptomes and the role of CTCF in AS regulation
