    Long-term spatial and temporal trends in frost indices in Kansas, USA

Climatic Change (2013) 120: 169. https://doi.org/10.1007/s10584-013-0794-4Frost indices such as number of frost days (nFDs), number of frost-free days (nFFDs), last spring freeze (LSF), first fall freeze (FFF), and growing-season length (GSL) were calculated using daily minimum air temperature (T[subscript min]) from 23 centennial weather stations across Kansas during four time periods (through 1919, 1920–1949, 1950–1979, and 1980–2009). A frost day is defined as a day with T[subscript min] < 0 °C. The long- and short-term trends in frost indices were analyzed at monthly, seasonal, and annual timescales. Probability of occurrence of the indices was analyzed at 5 %, 25 %, 50 %, 75 %, and 95 %. Results indicated a general increase in T[subscript min] from 1900 through 2009 causing a decrease in nFDs. LSF and FFF occurred earlier and later than normal in the year, respectively, thereby resulting in an increase in GSL. In general, northwest Kansas recorded the greatest nFD and lowest T[subscript min], whereas southeast Kansas had the lowest nFD and highest T[subscript min]; however, the magnitude of the trends in these indices varied with location, time period, and time scales. Based on the long-term records in most stations, LSF occurred earlier by 0.1–1.9 days/decade, FFF occurred later by 0.2–0.9 day/decade, and GSL was longer by 0.1–2.5 day/decade. At the 50 % probability level, Independence in the south-eastern part of Kansas had the earliest LSF (6 April), latest FFF (29 October) and longest GSL (207 days). Oberlin (north-western Kansas) recorded the shortest GSL (156 days) and earliest FFF (7 October) had the latest LSF (2 May) at the 50 % probability level. A positive correlation was observed for combinations of indices (LSF and GSL) and elevation, whereas a negative correlation was found between FFF and elevation

    PurposeTriple negative breast cancer (TNBC) is characterized by the presence of immune cells in the tumor microenvironment, however, the response to single-agent immune checkpoint inhibitor (ICI) therapy is modest. Preclinical models have demonstrated that intratumoral regulatory T cells (Tregs) dampen the antitumor response to ICI. We performed a single-arm phase II trial to evaluate the efficacy of a single low dose of cyclophosphamide (Cy) to deplete Tregs administered before initiating pembrolizumab.Patients and methods40 patients with pretreated metastatic TNBC were enrolled. The primary endpoints were progression-free survival (PFS) and change in peripheral blood Tregs after Cy. Secondary endpoints included overall response rate (ORR), duration of response, overall survival, treatment-related adverse events (AEs), and correlative evaluations.ResultsMedian PFS was 1.8 months, and the ORR was 21%. Tregs were not significantly decreased after Cy prior to ICI (−3.3%, p=0.19), and increased significantly after the first cycle of therapy (+21% between cycles 1 and 2, p=0.005). Immune-related AEs were similar to historical pembrolizumab monotherapy, and were associated with response to therapy (p=0.02). Patients with pretreatment tumors harboring increased expression of B cell metagene signatures and increased circulating B cell receptor repertoire diversity were associated with clinical response and immune-related toxicity (IRT).ConclusionsAmong patients with heavily pretreated TNBC, Cy prior to pembrolizumab did not significantly deplete Tregs, and in those with decreased numbers there was rapid recovery following therapy. Increased B cell gene expression in baseline samples was associated with clinical response and IRT.</jats:sec

    Evaluation of the BAX® System for Detection of Salmonella in Selected Foods: Collaborative Study

    Abstract A multilaboratory study was conducted to compare the automated BAX® System to the standard cultural methods for detection of Salmonella in selected foods. Five food types—frankfurters, raw ground beef, mozzarella cheese, raw frozen tilapia fish, and orange juice—at 3 inoculation levels, were analyzed by each method. A sixth food type, raw ground chicken, was tested using 3 naturally contaminated lots. A total of 16 laboratories representing government and industry participated. In this study, 1386 samples were analyzed, of which 1188 were paired samples and 198 were unpaired samples. Of the 1188 paired samples, 461 were positive by both methods and 404 were negative by both methods. Thirty-seven samples were positive by the BAX System but negative by the standard reference method, and 11 samples were positive by standard cultural method and negative by the BAX System. Of the 198 unpaired samples, 106 were positive by the BAX System and 60 were positive by the standard cultural method. A Chi square analysis of each of the 6 food types, at the 3 inoculation levels tested, was performed. For all foods, the BAX System demonstrated results comparable to those of the standard reference methods based on the Chi square results.</jats:p

    Evidence summaries tailored to health policy-makers in low- and middle-income countries

    Q1Artículo de investigación54-61Objective To describe how the SUPPORT collaboration developed a short summary format for presenting the results of systematic reviews to policy-makers in low- and middle-income countries (LMICs).Methods We carried out 21 user tests in six countries to explore users’ experiences with the summary format. We modified the summaries based on the results and checked our conclusions through 13 follow-up interviews. To solve the problems uncovered by the user testing, we also obtained advisory group feedback and conducted working group workshops.Findings Policy-makers liked a graded entry format (i.e. short summary with key messages up front). They particularly valued the section on the relevance of the summaries for LMICs, which compensated for the lack of locally-relevant detail in the original review. Some struggled to understand the text and numbers. Three issues made redesigning the summaries particularly challenging: (i) participants had a poor understanding of what a systematic review was; (ii) they expected information not found in the systematic reviews and (iii) they wanted shorter, clearer summaries. Solutions included adding information to help understand the nature of a systematic review, adding more references and making the content clearer and the document quicker to scan.Conclusion Presenting evidence from systematic reviews to policy-makers in LMICs in the form of short summaries can render the information easier to assimilate and more useful, but summaries must be clear and easy to read or scan quickly. They should also explain the nature of the information provided by systematic reviews and its relevance for policy decisions
