    Creative Placemaking as a Mechanism for Community Revitalization: A Case Study of the SALT District in the Near Westside of Syracuse, New York

    The purpose of this single qualitative case study was to identify the effects of creative placemaking interventions on the stakeholders of the SALT District in Syracuse, New York, and to explore how creative placemaking may help to revitalize other economically and socially blighted communities. Using the PolicyLink research and documentation framework to facilitate discovery, the researcher sought to understand the effects of creative placemaking on the people of the Near Westside. The participant sample included 12 stakeholders and their perceptions of creative placemaking in their community of Syracuse, New York. In addition to renovating warehouses and creating resident workspace for artists, the inquiry revealed that the initiative has created access to people and services, and it has created income-producing assets. Consistent with other research on the benefits of the arts, agency, self-efficacy, and empowerment emerged as a significant finding in the data. Arts participation and engagement, and representative leadership emerged while crime, as a result of the creative placemaking decreased. All 12 participants felt the initiative constructively impacted the area in some way. However, all of the participants thought it fell short of its promise. This research considered the impact of creative placemaking strategies on stakeholders of the Near Westside of Syracuse, through the lens of that community’s 50-year battle with systemic, perpetual poverty and scarcity. One recommendation for future research on creative placemaking in impoverished communities is to consider positioning similar studies with an eye toward urban thriving and elevating diversity as a value within the practice

    Autism spectrum disorders (ASD) affect one in 110 children in the U.S. Parents of children with ASD need clear and accurate information to communicate with professionals as they seek appropriate services, including applied behavior analysis (ABA) based intervention. Behavioral professionals can assist parents in this endeavor by recommending resources, including online courses. This paper describes the development and evaluation of an online course on ABA for parents of children with ASD. Parents completing a summative field test (N=21) made significant gains in knowledge and reported high levels of satisfaction. Implications include the potential for enhanced parent-professional collaboration in treatment decision-making

    Exome Genotyping, Linkage Disequilibrium and Population Structure in Loblolly Pine (\u3cem\u3ePinus taeda\u3c/em\u3e L.)

    Background: Loblolly pine (Pinus taeda L.) is one of the most widely planted and commercially important forest tree species in the USA and worldwide, and is an object of intense genomic research. However, whole genome resequencing in loblolly pine is hampered by its large size and complexity and a lack of a good reference. As a valid and more feasible alternative, entire exome sequencing was hence employed to identify the gene-associated single nucleotide polymorphisms (SNPs) and to genotype the sampled trees. Results: The exons were captured in the ADEPT2 association mapping population of 375 clonally-propagated loblolly pine trees using NimbleGen oligonucleotide hybridization probes, and then exome-enriched genomic DNA fragments were sequenced using the Illumina HiSeq 2500 platform. Oligonucleotide probes were designed based on 199,723 exons (≈49 Mbp) partitioned from the loblolly pine reference genome (PineRefSeq v. 1.01). The probes covered 90.2 % of the target regions. Capture efficiency was high; on average, 67 % of the sequence reads generated for each tree could be mapped to the capture target regions, and more than 70 % of the captured target bases had at least 10X sequencing depth per tree. A total of 972,720 high quality SNPs were identified after filtering. Among them, 53 % were located in coding regions (CDS), 5 % in 5’ or 3’ untranslated regions (UTRs) and 42 % in non-target and non-coding regions, such as introns and adjacent intergenic regions collaterally captured. We found that linkage disequilibrium (LD) decayed very rapidly, with the correlation coefficient (r 2) between pairs of SNPs linked within single scaffolds decaying to half maximum (r 2 = 0.22) within 55 bp, to r 2 = 0.1 within 192 bp, and to r 2 = 0.05 within 451 bp. Population structure analysis using unlinked SNPs demonstrated the presence of two main distinct clusters representing western and eastern parts of the loblolly pine range included in our sample of trees. Conclusions: The obtained results demonstrated the efficiency of exome capture for genotyping species such as loblolly pine with a large and complex genome. The highly diverse genetic variation reported in this study will be a valuable resource for future genetic and genomic research in loblolly pine

    Head and shoulder posture affect scapular mechanics and muscle activity in overhead tasks

    Forward head and rounded shoulder posture (FHRSP) is theorized to contribute to alterations in scapular kinematics and muscle activity leading to the development of shoulder pain. However, reported differences in scapular kinematics and muscle activity in those with forward head and rounded shoulder posture are confounded by the presence of shoulder pain. Therefore, the purpose of this study was to compare scapular kinematics and muscle activity in individuals free from shoulder pain, with and without FHRSP. Eighty volunteers were classified as having FHRSP or ideal posture. Scapular kinematics were collected concurrently with muscle activity from the upper and lower trapezius as well as the serratus anterior muscles during a loaded flexion and overhead reaching task using an electromagnetic tracking system and surface electromyography. Separate mixed model analyses of variance were used to compare three-dimensional scapular kinematics and muscle activity during the ascending phases of both tasks. Individuals with FHRSP displayed significantly greater scapular internal rotation with less serratus anterior activity, during both tasks as well as greater scapular upward rotation, anterior tilting during the flexion task when compared with the ideal posture group. These results provide support for the clinical hypothesis that FHRSP impacts shoulder mechanics independent of shoulder pain

    Loss of a 20S Proteasome Activator in Saccharomyces cerevisiae Downregulates Genes Important for Genomic Integrity, Increases DNA Damage, and Selectively Sensitizes Cells to Agents With Diverse Mechanisms of Action

    Cytoprotective functions of a 20S proteasome activator were investigated. Saccharomyces cerevisiae Blm10 and human 20S proteasome activator 200 (PA200) are homologs. Comparative genome-wide analyses of untreated diploid cells lacking Blm10 and growing at steady state at defined growth rates revealed downregulation of numerous genes required for accurate chromosome structure, assembly and repair, and upregulation of a specific subset of genes encoding protein-folding chaperones. Blm10 loss or truncation of the Ubp3/Blm3 deubiquitinating enzyme caused massive chromosomal damage and cell death in homozygous diploids after phleomycin treatments, indicating that Blm10 and Ubp3/Blm3 function to stabilize the genome and protect against cell death. Diploids lacking Blm10 also were sensitized to doxorubicin, hydroxyurea, 5-fluorouracil, rapamycin, hydrogen peroxide, methyl methanesulfonate, and calcofluor. Fluorescently tagged Blm10 localized in nuclei, with enhanced fluorescence after DNA replication. After DNA damage that caused a classic G2/M arrest, fluorescence remained diffuse, with evidence of nuclear fragmentation in some cells. Protective functions of Blm10 did not require the carboxyl-terminal region that makes close contact with 20S proteasomes, indicating that protection does not require this contact or the truncated Blm10 can interact with the proteasome apart from this region. Without its carboxyl-terminus, Blm10(−339aa) localized to nuclei in untreated, nonproliferating (G0) cells, but not during G1 S, G2, and M. The results indicate Blm10 functions in protective mechanisms that include the machinery that assures proper assembly of chromosomes. These essential guardian functions have implications for ubiquitin-independent targeting in anticancer therapy. Targeting Blm10/PA200 together with one or more of the upregulated chaperones or a conventional treatment could be efficacious

    Sustainable Human Presence on the Moon using In Situ Resources

    New capabilities, technologies and infrastructure must be developed to enable a sustained human presence on the moon and beyond. The key to having this permanent presence is the utilization of in situ resources. To this end, NASA is investigating how in situ resources can be utilized to improve mission success by reducing up-mass, improving safety, reducing risk, and bringing down costs for the overall mission. To ensure that this capability is available when needed, technology development is required now. NASA/Marshall Space Flight Center (MSFC) is supporting this endeavor, along with other NASA centers, by exploring how lunar regolith can be mined for uses such as construction, life support, propulsion, power, and fabrication. Efforts at MSFC include development of lunar regolith simulant for hardware testing and development, extraction of oxygen and other materials from the lunar regolith, production of parts and tools on the moon from local materials or from provisioned feedstocks, and capabilities to show that produced parts are "ready for use".     Discovery of an 86 AU Radius Debris Ring Around HD 181327

    HST/NICMOS PSF-subtracted coronagraphic observations of HD 181327 have revealed the presence of a ring-like disk of circumstellar debris seen in 1.1 micron light scattered by the disk grains, surrounded by a di use outer region of lower surface brightness. The annular disk appears to be inclined by 31.7 +/- 1.6 deg from face on with the disk major axis PA at 107 +/-2 deg . The total 1.1 micron flux density of the light scattered by the disk (at 1.2" < r < 5.0") of 9.6 mJy +/- 0.8 mJy is 0.17% +/- 0.015% of the starlight. Seventy percent of the light from the scattering grains appears to be confined in a 36 AU wide annulus centered on the peak of the radial surface brightness (SB) profile 86.3 +/- 3.9 AU from the star, well beyond the characteristic radius of thermal emission estimated from IRAS and Spitzer flux densities assuming blackbody grains (~ 22 AU). The light scattered by the ring appears bilaterally symmetric, exhibits directionally preferential scattering well represented by a Henyey-Greenstein scattering phase function with g = 0.30 +/- 0.03, and has an azimuthally medianed SB at the 86.3 AU radius of peak SB of 1.00 +/- 0.07 mJy arcsec^-2. No photocentric offset is seen in the ring relative to the position of the central star. A low surface brightness diffuse halo is seen in the NICMOS image to a distance of ~ 4" Deeper 0.6 micron HST/ACS PSF-subtracted coronagraphic observations reveal a faint outer nebulosity, asymmetrically brighter to the North of the star. We discuss models of the disk and properties of its grains, from which we infer a maximum vertical scale height of 4 - 8 AU at the 87.6 AU radius of maximum surface density, and a total maximum dust mass of collisionally replenished grains with minimum grain sizes of ~ 1 micron of ~ 4 M(moon).Comment: 45 pages, 15 figures, accepted for publication in Ap
