    Effects of tumour necrosis factor-α, interleukin-1 α, macrophage colony stimulating factor and transforming growth factor β on trophoblastic matrix metalloproteinases

    The aim of this study was to determine the effects of tumour necrosis factor α (TNF), interleukin-1 α (IL-1α), macrophage colony-stimulating factor (MCSF) and transforming growth factor β (TGFβ) on the secretion of matrix metalloproteinases (MMP), human chorionic gonadotrophin (HCG) and fetal fibronectin (fFN) by purified first trimester cytotrophoblastic cells (CTB) in vitro. CTB were obtained from legal abortions and cultured in vitro in the presence or absence of the different cytokines. Secreted gelatinases were analysed in the culture supernatants by zymography, by measurements of the total gelatinolytic activity and by enzyme immunoassays. HCG and fFN were measured by commercially available immunoassays. TNF increased the total gelatinolytic activity by increasing MMP-9 activity (P = 0.025-0.0177) but decreased MMP-2 activity (P < 0.03) and immunoreactivity (P < 0.05), fFN (P < 0.02) and HCG (P < 0.01). IL-1α significantly increased the secretion of fFN (P < 0.02), the activity (P < 0.02) and immunoreactivity (P < 0.05) of MMP-9 but had no effect on the other parameters. MCSF increased MMP-9 immunoreactivity (P < 0.05) and moderately decreased HCG. TGFβ inhibited total gelatinolytic activity, MMP-9 activity and immunoreactivity, but was without effect on MMP-2 concentrations and activity. TGFβ decreased HCG (P < 0.041) and increased fFN (P < 0.042). Our results indicate that TGFβ, TNF and IL-1α are important regulators of trophoblastic MMP secretio

    Biomechanics analysis of the ski jump is highly required. Some parameters and their interrelations have been reported in previous research studies limited to few athletes. The generalization of these parameters to athletes of various levels and under training conditions should be assessed, since they have the potential to be used for daily evaluation. This study proposed a new 3D approach based on inertial sensors to evaluate relevant kinematic and aerodynamic parameters of stable flight phase. The proposed wearable system can easily be used for daily training. Aerodynamic forces and body segments 3D angles were extracted during the stable flight phase of 86 jumps. Then, their correlations with respect to distance as well as their interrelations were analyzed. Their combination expressed 55% of the total distance variance

    Removal of spermatozoa with externalized phosphatidylserine from sperm preparation in human assisted medical procreation: effects on viability, motility and mitochondrial membrane potential

    <p>Abstract</p> <p>Background</p> <p>Externalization of phosphatidylserine (EPS) occurs in apoptotic-like spermatozoa and could be used to remove them from sperm preparations to enhance sperm quality for assisted medical procreation. We first characterized EPS in sperms from infertile patients in terms of frequency of EPS spermatozoa as well as localization of phosphatidylserine (PS) on spermatozoa. Subsequently, we determined the impact of depleting EPS spermatozoa on sperm quality.</p> <p>Methods</p> <p>EPS were visualized by fluorescently-labeled annexin V binding assay. Double staining with annexin V and Hoechst differentiates apoptotic from necrotic spermatozoa. We used magnetic-activated cell sorting using annexin V-conjugated microbeads (MACS-ANMB) technique to remove EPS spermatozoa from sperm prepared by density gradient centrifugation (DGC). The impact of this technique on sperm quality was evaluated by measuring progressive motility, viability, and the integrity of the mitochondrial membrane potential (MMP) by Rhodamine 123.</p> <p>Results</p> <p>Mean percentages of EPS spermatozoa were 14% in DGC sperm. Four subpopulations of spermatozoa were identified: 70% alive, 3% early apoptotic, 16% necrotic and 11% late apoptotic or necrotic. PS were localized on head and/or midpiece or on the whole spermatozoa. MACS efficiently eliminates EPS spermatozoa. MACS combined with DGC allows a mean reduction of 70% in EPS and of 60% in MMP-disrupted spermatozoa with a mean increase of 50% in sperm survival at 24 h.</p> <p>Conclusion</p> <p>Human ejaculates contain EPS spermatozoa which can mostly be eliminated by DGC plus MACS resulting in improved sperm long term viability, motility and MMP integrity. EPS may be used as an indicator of sperm quality and removal of EPS spermatozoa may enhance fertility potential in assisted medical procreation.</p
