A rat model against chemotherapy plus radiation-induced oral mucositis

AbstractObjectivesPresent study was aimed at developing an experimental model of oral mucositis in rats using a combination of chemotherapeutic agent and radiation.Study designFemale Wistar rats (150–200g) were divided into 3 groups (n=6). Rats in group 1 (normal control) and group 2 (mucositis control) were treated with vehicle. Rats in group 3 were treated with l-glutamine (1g/kg, p.o.; 15days) before and after mucositis induction. Oral mucositis was induced by busulfan (6mg/kg, p.o.; 4days) and the tongue exposed to infrared (IR) radiation of intensity 40mV/cm2 for 5 s on the 1st, 4th and 10th days of challenge using a tail flick apparatus. Parameters monitored were body weight, food intake, blood count and survival. Oral mucositis score (OMS) was recorded daily. Histological changes of the irradiated tongue were assessed by hematoxylin and eosin staining.ResultsBusulfan and IR radiation significantly reduced body weight and food intake of the mucositis control group as compared to normal control. Clear ulceration of the tongue reflected in the OMS. Histopathology of the tongue revealed intense lymphocytic infiltration, decreased thickness of squamous epithelial cell layer, decrease in number of blood vessels, and necrosis of cells along with pseudo-membrane formation in the mucositis control group. These findings suggested that oral mucositis was successfully induced and treatment with l-glutamine partially reversed these conditions.ConclusionOral mucositis was established successfully in rats by the combination of chemotherapeutic agent and IR radiation. This may be a useful model for screening drugs in the treatment of oral mucositis

Free radical scavenging activity of gossypin and nevadensin: An in-vitro evaluation

Objectives: The antioxidant potential of gossypin and nevadensin, two flavonoid compounds, were evaluated by in vitro methods. Materials and Methods: Gossypin, nevadensin, and the reference standard, butylated hydroxyl toluene (BHT), were evaluated for DPPH (1, 1-diphenyl-2-picrylhydrazyl), nitric oxide, superoxide, and hydroxyl radical scavenging activity. Results: Gossypin and BHT showed the potential for significant DPPH radical inhibition of up to 88.52 and 91.45% at 100 µg/ml concentration. With a 100 µg/ml concentration of gossypin, the in vitro nitric oxide, superoxide, and hydroxyl radical scavenging activity was found to be 74.00, 74.22, and 67.15%, respectively; and with 100 µg/ml of BHT the corresponding values were 82.24, 81.76, and 73.03% of inhibition, respectively. Conclusion: The study results showed that gossypin has significant antioxidant activity

Anti-arthritic activity of Indigofera tinctoria L on adjuvant induced arthritis

Indigofera tinctoria L has long been traditionally employed for various diseases, including pain and inflammation. The present study is an attempt to evaluate the potential anti-arthritic activity of Indigofera tinctoria L. The antiarthritic activity for petroleum ether and ethanol extracts (100, 200 and 300 mg/kg) of Indigofera tinctoria L was carried out by inducing Freunds Complete Adjuvant (FCA) at the tibiotorsal joint. Behavioral changes, vascular permeability, histamine, hematological parameters, ESR, evaluation of development of arthritis, biochemical parameters, and Interleukin (IL) were estimated. In vitro proliferation of Spleen cells (MTT assay), Radiographic analysis and histopathological assessments were carried out. The petroleum ether and ethanol extracts of Indigofera tinctoria L leaf showed anti-arthritic activity, by significant changes in the behavioral activity, decreased vascular permeability, histamine level, IL and TNF, proliferation of spleenocytes dose-dependently. Similarly the significant increased SOD, GSH and reduced level of lipid peroxidation and decreased ESR, WBC’s and lymphocytes were observed. The soft tissue swelling and bone resorption were also reduced. The infiltration of leucocytes and loss of articular cartilage was significantly reduced in groups treated with the extracts. Global perspective of familial hypercholesterolaemia: a cross-sectional study from the EAS Familial Hypercholesterolaemia Studies Collaboration (FHSC)

none724siBackground The European Atherosclerosis Society Familial Hypercholesterolaemia Studies Collaboration (FHSC) global registry provides a platform for the global surveillance of familial hypercholesterolaemia through harmonisation and pooling of multinational data. In this study, we aimed to characterise the adult population with heterozygous familial hypercholesterolaemia and described how it is detected and managed globally.Methods Using FHSC global registry data, we did a cross-sectional assessment of adults (aged 18 years or older) with a clinical or genetic diagnosis of probable or definite heterozygous familial hypercholesterolaemia at the time they were entered into the registries. Data were assessed overall and by WHO regions, sex, and index versus non-index cases.Findings Of the 61 612 individuals in the registry, 42 167 adults (21 999 [53.6%] women) from 56 countries were included in the study. Of these, 31 798 (75.4%) were diagnosed with the Dutch Lipid Clinic Network criteria, and 35 490 (84.2%) were from the WHO region of Europe. Median age of participants at entry in the registry was 46.2 years (IQR 34.3-58.0); median age at diagnosis of familial hypercholesterolaemia was 44.4 years (32.5-56.5), with 40.2% of participants younger than 40 years when diagnosed. Prevalence of cardiovascular risk factors increased progressively with age and varied by WHO region. Prevalence of coronary disease was 17.4% (2.1% for stroke and 5.2% for peripheral artery disease), increasing with concentrations of untreated LDL cholesterol, and was about two times lower in women than in men. Among patients receiving lipid-lowering medications, 16 803 (81.1%) were receiving statins and 3691 (21.2%) were on combination therapy, with greater use of more potent lipid-lowering medication in men than in women. Median LDL cholesterol was 5.43 mmol/L (IQR 4.32-6.72) among patients not taking lipid-lowering medications and 4.23 mmol/L (3.20-5.66) among those taking them. Among patients taking lipid-lowering medications, 2.7% had LDL cholesterol lower than 1.8 mmol/L; the use of combination therapy, particularly with three drugs and with proprotein convertase subtilisin-kexin type 9 inhibitors, was associated with a higher proportion and greater odds of having LDL cholesterol lower than 1.8 mmol/L. Compared with index cases, patients who were non-index cases were younger, with lower LDL cholesterol and lower prevalence of cardiovascular risk factors and cardiovascular diseases (all p<0.001).Interpretation Familial hypercholesterolaemia is diagnosed late. Guideline-recommended LDL cholesterol concentrations are infrequently achieved with single-drug therapy. Cardiovascular risk factors and presence of coronary disease were lower among non-index cases, who were diagnosed earlier. Earlier detection and greater use of combination therapies are required to reduce the global burden of familial hypercholesterolaemia. Copyright (C) 2021 Elsevier Ltd.
