    Ethics After Theory

    Today, architectural theory has atrophied. Despite a perceived need for different theoretical ways of clarifying, explaining and understanding the complex phenomena of contemporary architectural production prior to a designer's intervention, there are few existing theoretical frameworks. Without either the descriptive or projective enablement by theory, conscientious and critical practices in architecture will have few alternatives against the tide of neoliberal city-making. In this paper, I make a pun of Speaks's (2006) paper titled, ‘Intelligence after theory' by arguing that while (design) intelligence is necessary, it is however not sufficient. Ethics is also needed in architecture. Through a brief restatement of a canonical debate in the critical school of architectural theory, I argue why ethics is needed in architecture, and how ethics could be further developed through theory and theorization

    One-pot Synthesis of Lactams Using Domino Reactions: Combination of Schmidt Reaction with Sakuri and Aldol Reaction

    A series of domino reactions in which the intramolecular Schmidt reaction is combined with either a Sakurai reaction, an aldol reaction, or both is reported. The Sakurai reaction of an allylsilane with an azido-containing enone under Lewis acidic conditions followed by protonation of the resulting titanium enolate species allowed for a subsequent intramolecular Schmidt reaction. Alternatively, the intermediate titanium enolate could undergo an aldol reaction, followed by the intramolecular Schmidt reaction to form lactam products with multiple stereogenic centers. The stereochemical features of the titanium enolate aldol reaction with several 3-azidoaldehyde substrates during this domino process is discussed

    2BC Non-Structural Protein of Enterovirus A71 Interacts with SNARE Proteins to Trigger Autolysosome Formation

    Viruses have evolved unique strategies to evade or subvert autophagy machinery. Enterovirus A71 (EV-A71) induces autophagy during infection in vitro and in vivo. In this study, we report that EV-A71 triggers autolysosome formation during infection in human rhabdomyosarcoma (RD) cells to facilitate its replication. Blocking autophagosome-lysosome fusion with chloroquine inhibited virus RNA replication, resulting in lower viral titres, viral RNA copies and viral proteins. Overexpression of the non-structural protein 2BC of EV-A71 induced autolysosome formation. Yeast 2-hybrid and co-affinity purification assays showed that 2BC physically and specifically interacted with a N-ethylmaleimide-sensitive factor attachment receptor (SNARE) protein, syntaxin-17 (STX17). Co-immunoprecipitation assay further showed that 2BC binds to SNARE proteins, STX17 and synaptosome associated protein 29 (SNAP29). Transient knockdown of STX17, SNAP29, and microtubule-associated protein 1 light chain 3B (LC3B), crucial proteins in the fusion between autophagosomes and lysosomes) as well as the lysosomal-associated membrane protein 1 (LAMP1) impaired production of infectious EV-A71 in RD cells. Collectively, these results demonstrate that the generation of autolysosomes triggered by the 2BC non-structural protein is important for EV-A71 replication, revealing a potential molecular pathway targeted by the virus to exploit autophagy. This study opens the possibility for the development of novel antivirals that specifically target 2BC to inhibit formation of autolysosomes during EV-A71 infection.Peer reviewe

    One Size Doesn't Fit All: Improving Network QoS Through Preference-driven Web Caching

    In order to combat Internet congestion Web caches use replacement policies that attempt to keep the objects in a cache that are most likely to get requested in the future. We adopt the economic perspective that the objects with the greatest value to the users should be in a cache. Using trace driven simulations we implement an incentive compatible market-based Web cache for servers to push content into a cache. This system decentralizes the caching process as servers provide information in the form of bids for space in the cache. Truthful information from the server on valuations of objects and predictions of hit rates is obtained. This information is used in filling the cache, which can provide increased aggregate value and differential quality of service to servers when compared to LFU and LRU.http://deepblue.lib.umich.edu/bitstream/2027.42/50429/1/berlin.pd

    Experimental and meta-analytic evidence that source variability of misinformation does not increase eyewitness suggestibility independently of repetition of misinformation

    Considerable evidence has shown that repeating the same misinformation increases its influence (i.e., repetition effects). However, very little research has examined whether having multiple witnesses present misinformation relative to one witness (i.e., source variability) increases the influence of misinformation. In two experiments, we orthogonally manipulated repetition and source variability. Experiment 1 used written interview transcripts to deliver misinformation and showed that repetition increased eyewitness suggestibility, but source variability did not. In Experiment 2, we increased source saliency by delivering the misinformation to participants via videos instead of written interviews, such that each witness was visibly and audibly distinct. Despite this stronger manipulation, there was no effect of source variability in Experiment 2. In addition, we reported a meta-analysis (k = 19) for the repeated misinformation effect and a small-scale meta-analysis (k = 8) for the source variability effect. Results from these meta-analyses were consistent with the results of our individual experiments. Altogether, our results suggest that participants respond based on retrieval fluency rather than source-specifying information
