    Natriuretic Peptides Synthesis and Secretion Profiles during the Evolution of Cardiac Hypertrophy in DOCA-Salt Hypertensive Rats

    Durante el desarrollo de la hipertensión arterial, las interacciones entre las sobrecargas de presión y de volumen conducen a diferentes patrones de hipertrofia cardíaca y a un aumento de los péptidos natriuréticos (PN). Los perfiles de síntesis y secreción de ANP y BNP se han investigado en modelos de hipertensión arterial; sin embargo, aún no se ha estudiado la evolución diferencial de estos perfiles durante períodos agudos y crónicos de la hipertrofia cardíaca producida por sobrecarga de volumen. Por este motivo estudiamos ratas Sprague- Dawley con el modelo DOCA-sal a las 2, 4, 6 y 12 semanas, correlacionando la evolución de dichos perfiles con la hipertrofia cardíaca y la hipertensión arterial. El grado de hipertrofia cardíaca se correlacionó positivamente con la expresión del ANP en el ventrículo izquierdo y con los niveles de ANP en plasma. La expresión del ANP aumentó a las 4 semanas de tratamiento, mientras que la de BNP se incrementó recién a las 6 semanas. Asimismo, el BNP plasmático se incrementó sólo en el grupo con 12 semanas de tratamiento, mientras que el ANP plasmático mostró un aumento a partir de las 2 semanas de tratamiento. Durante el desarrollo de la hipertrofia cardíaca producida en el modelo DOCA-sal, la síntesis y la secreción de los PN responden en forma diferencial, con incremento precoz del ANP. Además, el aumento de éste superó al de BNP en todos los grupos DOCA-sal, lo que permitiría considerar al ANP como un marcador más específico de la sobrecarga de volumen.The interactions between pressure and volume overload that occur in hypertension lead to different patterns of cardiac hypertrophy and to increase in natriuretic peptides (NPs). The profiles of ANP and BNP synthesis and secretion have been investigated in models of hypertension; however, the different evolution of these profiles during the acute and chronic periods of pressure overload-induced cardiac hypertrophy is still unknown. For this reason, we studied DOCA-salt treated Sprague-Dawley rats at weeks 2, 4, 6 and 12 and correlated the evolution of these profiles with cardiac hypertrophy and hypertension. Cardiac hypertrophy had a positive correlation with ANP expression in the left ventricle and with ANP plasma levels. BNP expression increased after 4 weeks of treatment while ANP increased significantly after 6 weeks. In addition, BNP plasma levels increased only in the group treated for 12 weeks, while ANP plasma levels increased from week 2. NP secretion has a differential response in the early stages of the development of cardiac hypertrophy induced by the DOCA-salt model, with an early increase in ANP.     La localidad de Potrerillos, en el departamento de Luján de Cuyo, se encuentra conformada por villas cordilleranas en un fondo de montaña a gran altura en cercanía del llamado “Cordón del Plata” perteneciente a la Cordillera Frontal de Los Andes, que fueron consideradas por sus características para este trabajo de investigación como unidad funcional. En los últimos 40 años la construcción de viviendas se fue incrementando a raíz del creciente turismo y principalmente desde el año 2005, surge la radicación de familias movilizadas por un cambio en el estilo de vida urbana por el espacio rural y la protección del ambiente. El crecimiento del asentamiento humano sin el debido control sobre los recursos naturales ha tenido efectos negativos en el ambiente frágil de dichas villas y en el incremento de los riesgos de origen natural, especialmente el aluvional, viento zonda y sísmico

    Photoreceptors UVR8 and phytochrome B cooperate to optimize plant growth and defense in patchy canopies

    Light is a critical source of information for plants. Plants use the phytochromes (particularly phyB) to detect light signals associated with the proximity of competitors. A low ratio of red (R) to far-red (FR) radiation (R:FR) indicates increased competition intensity, and triggers morphological responses that allow the plant to escape shading from its neighbors (the shade avoidance syndrome, SAS). Recent evidence from studies on light regulation of plant immunity has suggested that plants may also use ultraviolet-B (UV-B, 290-315 nm) radiation as an indicator of competition intensity and light availability. In addition, recent studies have shown that UV-B radiation can strongly repress SAS responses triggered by low R:FR ratios. Ambient UV-B radiation causes damaging effects on plants, such as DNA damage, and also induces adaptive photomorphogenic responses acting through a specific UV-B photoreceptor (UVR8). Therefore, the possibility exists that plants integrate information perceived by phyB and UVR8 to make decisions about growth and defense when faced with a complex light environment, such as the one that characterizes vegetation canopies. In this Letter, we address this possibility and discuss how the interplay between UV-B and R:FR signaling fine tunes plant growth and defense to optimize resource utilization in patchy canopy environments.Fil: Mazza, Carlos Alberto.     Genome-based trait prediction in multi- environment breeding trials in groundnut

    Genomic selection (GS) can be an efficient and cost-effective breeding approach which captures both small- and large-effect genetic factors and therefore promises to achieve higher genetic gains for complex traits such as yield and oil content in groundnut. A training population was constituted with 340 elite lines followed by genotyping with 58 K ‘Axiom_Arachis’ SNP array and phenotyping for key agronomic traits at three locations in India. Four GS models were tested using three different random cross-validation schemes (CV0, CV1 and CV2). These models are: (1) model 1 (M1 = E + L) which includes the main effects of environment (E) and line (L); (2) model 2 (M2 = E + L + G) which includes the main effects of markers (G) in addition to E and L; (3) model 3 (M3 = E + L + G + GE), a naïve interaction model; and (4) model 4 (E + L + G + LE + GE), a naïve and informed interaction model. Prediction accuracy estimated for four models indicated clear advantage of the inclusion of marker information which was reflected in better prediction accuracy achieved with models M2, M3 and M4 as compared to M1 model. High prediction accuracies (> 0.600) were observed for days to 50% flowering, days to maturity, hundred seed weight, oleic acid, rust@90 days, rust@105 days and late leaf spot@90 days, while medium prediction accuracies (0.400–0.600) were obtained for pods/plant, shelling %, and total yield/plant. Assessment of comparative prediction accuracy for different GS models to perform selection for untested genotypes, and unobserved and unevaluated environments provided greater insights on potential application of GS breeding in groundnut

    Low Red/Far-Red Ratios Reduce Arabidopsis Resistance to Botrytis cinerea and Jasmonate Responses via a COI1-JAZ10-Dependent, Salicylic Acid-Independent Mechanism1[C][W][OA]

    Light is an important modulator of plant immune responses. Here, we show that inactivation of the photoreceptor phytochrome B (phyB) by a low red/far-red ratio (R:FR), which is a signal of competition in plant canopies, down-regulates the expression of defense markers induced by the necrotrophic fungus Botrytis cinerea, including the genes that encode the transcription factor ETHYLENE RESPONSE FACTOR1 (ERF1) and the plant defensin PLANT DEFENSIN1.2 (PDF1.2). This effect of low R:FR correlated with a reduced sensitivity to jasmonate (JA), thus resembling the antagonistic effects of salicylic acid (SA) on JA responses. Low R:FR failed to depress PDF1.2 mRNA levels in a transgenic line in which PDF1.2 transcription was up-regulated by constitutive expression of ERF1 in a coronatine insensitive1 (coi1) mutant background (35S::ERF1/coi1). These results suggest that the low R:FR effect, in contrast to the SA effect, requires a functional SCFCOI1-JASMONATE ZIM-DOMAIN (JAZ) JA receptor module. Furthermore, the effect of low R:FR depressing the JA response was conserved in mutants impaired in SA signaling (sid2-1 and npr1-1). Plant exposure to low R:FR ratios and the phyB mutation markedly increased plant susceptibility to B. cinerea; the effect of low R:FR was (1) independent of the activation of the shade-avoidance syndrome, (2) conserved in the sid2-1 and npr1-1 mutants, and (3) absent in two RNA interference lines disrupted for the expression of the JAZ10 gene. Collectively, our results suggest that low R:FR ratios depress Arabidopsis (Arabidopsis thaliana) immune responses against necrotrophic microorganisms via a SA-independent mechanism that requires the JAZ10 transcriptional repressor and that this effect may increase plant susceptibility to fungal infection in dense canopies
