    A fate-alternating transitional regime in contracting liquid filaments

    The fate of a contracting liquid filament depends on the Ohnesorge number, the initial aspect ratio and surface perturbation. Generally, it is believed that there exists a critical aspect ratio such that longer filaments break up and shorter ones recoil into a single drop. Through computational and experimental studies, we report a transitional regime for filaments with a broad range of intermediate aspect ratios, where there exist multiple thresholds at which a novel breakup mode alternates with a no-break mode. We develop a simple model considering the superposition of capillary waves, which can predict the complicated new phase diagram. In this model, the breakup results from constructive interference between the capillary waves that originate from the ends of the filament

    Checkpoint inhibition reduces the threshold for Drug-Specific T-Cell priming and increases the incidence of sulfasalazine hypersensitivity

    An emerging clinical issue associated with immune-oncology agents is the collateral effects on the tolerability of concomitant medications. One report of this phenomenon was the increased incidence of hypersensitivity reactions observed in patients receiving concurrent immune checkpoint inhibitors (ICIs) and sulfasalazine (SLZ). Thus, the aim of this study was to characterize the T cells involved in the pathogenesis of such reactions, and recapitulate the effects of inhibitory checkpoint blockade on de-novo priming responses to compounds within in vitro platforms. A regulatory competent human dendritic cell/T-cell coculture assay was used to model the effects of ICIs on de novo nitroso sulfamethoxazole- and sulfapyridine (SP) (the sulfonamide component of SLZ) hydroxylamine-specific priming responses. The role of T cells in the pathogenesis of the observed reactions was explored in 3 patients through phenotypic characterization of SP/sulfapyridine hydroxylamine (SPHA)-responsive T-cell clones (TCC), and assessment of cross-reactivity and pathways of T-cell activation. Augmentation of the frequency of responding drug-specific T cells and intensity of the T-cell response was observed with PD-1/PD-L1 blockade. Monoclonal populations of SP- and SPHA-responsive T cells were isolated from all 3 patients. A core secretory effector molecule profile (IFN-γ, IL-13, granzyme B, and perforin) was identified for SP and SPHA-responsive TCC, which proceeded through Pi and hapten mechanisms, respectively. Data presented herein provides evidence that drug-responsive T cells are effectors of hypersensitivity reactions observed in oncology patients administered ICIs and SLZ. Perturbation of drug-specific T-cell priming is a plausible explanation for clinical observations of how an increased incidence of these adverse events is occurring

    Intravascular Large B-Cell Lymphoma Genomic Profile Is Characterized by Alterations in Genes Regulating NF-κB and Immune Checkpoints.

    Intravascular large B-cell lymphoma (IVLBCL) is an uncommon lymphoma with an aggressive clinical course characterized by selective growth of tumor cells within the vessels. Its pathogenesis is still uncertain and there is little information on the underlying genomic alterations. In this study, we performed a clinicopathologic and next-generation sequencing analysis of 15 cases of IVLBCL using a custom panel for the detection of alterations in 68 recurrently mutated genes in B-cell lymphomagenesis. Six patients had evidence of hemophagocytic syndrome. Four patients presented concomitantly a solid malignancy. Tumor cells outside the vessels were observed in 7 cases, 2 with an overt diffuse large B-cell cell lymphoma. In 4 samples, tumor cells infiltrated lymphatic vessel in addition to blood capillaries. Programmed death-ligand 1 (PD-L1) was positive in tumor cells in 4 of 11 evaluable samples and in macrophages intermingled with tumor cells in 8. PD-L1 copy number gains were identified in a higher proportion of cases expressing PD-L1 than in negative tumors. The most frequently mutated gene was PIM1 (9/15, 60%), followed by MYD88L265P and CD79B (8/15, 53% each). In 6 cases, MYD88L265P and CD79B mutations were detected concomitantly. We also identified recurrent mutations in IRF4 , TMEM30A , BTG2 , and ETV6 loci (4/15, 27% each) and novel driver mutations in NOTCH2 , CCND3 , and GNA13 , and an IRF4 translocation in 1 case each. The mutational profile was similar in patients with and without evidence of hemophagocytic syndrome and in cases with or without dissemination of tumor cells outside the vessels. Our results confirm the relevance of mutations in B-cell receptor/nuclear factor-κB signaling and immune escape pathways in IVLBCL and identify novel driver alterations. The similar mutational profile in tumors with extravascular dissemination suggests that these cases may also be considered in the spectrum of IVLBCL

    On the analysis of the contact angle for impacting droplets using a polynomial fitting approach

    ractical considerations on the measurement of the dynamic contact angle and the spreading diameter of impacting droplets are discussed in this paper. The contact angle of a liquid is commonly obtained either by a polynomial or a linear fitting to the droplet profile around the triple phase point. Previous works have focused on quasi-static or sessile droplets, or in cases where inertia does not play a major role on the contact angle dynamics. Here, we study the effect of droplet shape, the order of the fitting polynomial, and the fitting domain, on the measurement of the contact angle on various stages following droplet impact where the contact line is moving. Our results, presented in terms of the optical resolution and the droplet size, show that a quadratic fitting provides the most consistent results for a range of various droplet shapes. As expected, our results show that contact angle values are less sensitive to the fitting conditions for the cases where the droplet can be approximated to a spherical cap. Our experimental conditions include impact events with liquid droplets of different sizes and viscosities on various substrates. In addition, validating past works, our results show that the maximum spreading diameter can be parameterised by the Weber number and the rapidly advancing contact angle

    Corrosion behavior of aluminum-carbon fiber/epoxy sandwich composite exposed on NaCl solution

    For years, the aeronautical industry has employed different types of materials to satisfy its high-performance requirements. Fiber-metal laminates are used due to their combination of lighter weight and the high mechanical properties of reinforced metal and carbon. We therefore made two different composites of laminate-metal and laminate-metal-laminate of carbon fiber-reinforced polymer and aluminum with an ALCLAD layer. The samples were characterized by salt fog (0, 48, and 96 h) at 5 wt% NaCl and electrochemical impedance spectroscopy (EIS) with an electrolyte of 3.5 wt% NaCl. All samples were studied by electron scanning microscopy (SEM). The results demonstrated that the samples of laminate-metal-laminate presented an adsorption process after 0 and 48 h of salt fog exposition; meanwhile, the samples of laminate-metal showed a capacitive behavior for all the samples; however, corrosion resistance decreased when the salt fog exposition time increased

    Cut-offs and response criteria for the Hospital Universitario la Princesa Index (HUPI) and their comparison to widely-used indices of disease activity in rheumatoid arthritis

    Objective To estimate cut-off points and to establish response criteria for the Hospital Universitario La Princesa Index (HUPI) in patients with chronic polyarthritis. Methods Two cohorts, one of early arthritis (Princesa Early Arthritis Register Longitudinal PEARL] study) and other of long-term rheumatoid arthritis (Estudio de la Morbilidad y Expresión Clínica de la Artritis Reumatoide EMECAR]) including altogether 1200 patients were used to determine cut-off values for remission, and for low, moderate and high activity through receiver operating curve (ROC) analysis. The areas under ROC (AUC) were compared to those of validated indexes (SDAI, CDAI, DAS28). ROC analysis was also applied to establish minimal and relevant clinical improvement for HUPI. Results The best cut-off points for HUPI are 2, 5 and 9, classifying RA activity as remission if =2, low disease activity if >2 and =5), moderate if >5 and <9 and high if =9. HUPI''s AUC to discriminate between low-moderate activity was 0.909 and between moderate-high activity 0.887. DAS28''s AUCs were 0.887 and 0.846, respectively; both indices had higher accuracy than SDAI (AUCs: 0.832 and 0.756) and CDAI (AUCs: 0.789 and 0.728). HUPI discriminates remission better than DAS28-ESR in early arthritis, but similarly to SDAI. The HUPI cut-off for minimal clinical improvement was established at 2 and for relevant clinical improvement at 4. Response criteria were established based on these cut-off values. Conclusions The cut-offs proposed for HUPI perform adequately in patients with either early or long term arthritis

    Wild species of vaccinium composition, nutritional value and utilization

    There are still, in many parts of the globe, some edible fruit collects from the wild for human feeding and other uses. These fruits are utilised either in their raw nature or after some form of processing. One of these wild fruits are Vaccinium species such as Vaccinium myrtoides (Blume) Miq., Vaccinium cylindraceum, Vaccinium padifolium, Vaccinium corymbosum, Vaccinium myrtillus and others from the plant family Ericaceae. The term wild infers non-cultivated plants found in plantation felids or the forest. The species Vaccinium myrtoides and the others are small trees (shrubs) that are well known in Southeast Asian countries such as Philippines, Indonesia and other neighbouring islands. The plants classified as wild growing shrubs. However, the locals have made it be fully utilized in some areas as the wood that are used in grafting utensils and cutleries, or as fuel and fruits. The small-sized berry-like fruits with an average diameter of about 4–5 mm turn black when ripe. They have a delicious flavour that makes it edible and used in the making of other delicacies such as tart and pies to add flavour and as preservatives as well. A few published researches were done on Vaccinium myrtoides showing that it is still used in folkloric medicine. Leaves and fruits extracts showed strong antioxidant activity when tested in-vitro using DPPH (2, 2-diphenyl-1-picrylhydrazyl) free radical scavenging activity as part of a preliminary phytochemical screening for V. myrtoides. The study revealed that the antioxidant activity is due to the presence of flavonoids and other phenolic compounds in the plant leaves and fruit extracts
