    Norm It! Lexical Normalization for Italian and Its Downstream Effects forDependency Parsing

    Lexical normalization is the task of translating non-standard social media data to a standard form. Previous work has shown that this is beneficial for many downstream tasks in multiple languages. However, for Italian, there is no benchmark available for lexical normalization, despite the presence of many benchmarks for other tasks involving social media data. In this paper, we discuss the creation of a lexical normalization dataset for Italian. After two rounds of annotation, a Cohen’s kappa score of 78.64 is obtained. During this process, we also analyze the inter-annotator agreement for this task, which is only rarely done on datasets for lexical normalization,and when it is reported, the analysis usually remains shallow. Furthermore, we utilize this dataset to train a lexical normalization model and show that it can be used to improve dependency parsing of social media data.     The Capacity of Magnesium to Induce Osteoclast Differentiation Is Greatly Enhanced by the Presence of Zoledronate

    Simple Summary A number of skeletal disorders, all characterized by a metabolic or neoplastic loss of bone tissue, are cured with drugs called Bisphosphonates (BPs), which exert their therapeutic effect by suppressing cells named osteoclasts, normally mediating bone resorption. Unfortunately, these drugs can also provoke a dangerous side effect known as osteonecrosis of the jaw (ONJ), a bone infection localized in the oral cavity and characterized by gingival ulceration, sometimes accompanied by suppuration and pain. This condition, occasionally arising spontaneously, is more often started by a tooth extraction. The reduced number of osteoclasts, determined by BPs, is thought to favor the bacterial invasion of healthy bone and the incapacity to eliminate infected bone, that are in turn responsible for the appearance of ONJ. Here we show that Magnesium, used for decades as dietary supplement, can invert the effect of BPs, transforming them, through a sort of paradox effect, into powerful activators of osteoclast production. These results suggest that Magnesium might be used in a topical approach aimed to cure or prevent ONJ. Notably, the capacity of Magnesium to activate osteoclast production was even observed in absence of BPs, suggesting its application also in ONJ forms caused by agents distinct to BPs.Abstract Bisphosphonates (BPs) are successfully used to cure a number of diseases characterized by a metabolic reduction in bone density, such as Osteoporosis, or a neoplastic destruction of bone tissue, such as multiple myeloma and bone metastases. These drugs exert their therapeutic effect by causing a systemic osteoclast depletion that, in turn, is responsible for reduced bone resorption. Unfortunately, in addition to their beneficial activity, BPs can also determine a frightening side effect known as osteonecrosis of the jaw (ONJ). It is generally believed that the inability of osteoclasts to dispose of inflamed/necrotic bone represents the main physiopathological aspect of ONJ. In principle, a therapeutic strategy able to elicit a local re-activation of osteoclast production could counteract ONJ and promote the healing of its lesions. Using an experimental model of Vitamin D3-dependent osteoclastogenesis, we have previously demonstrated that Magnesium is a powerful inducer of osteoclast differentiation. Here we show that, surprisingly, this effect is greatly enhanced by the presence of Zoledronate, chosen for our study because it is the most effective and dangerous of the BPs. This finding allows us to hypothesize that Magnesium might play an important role in the topical therapy of ONJ

    Lipid Cubic Mesophases Combined with Superparamagnetic Iron Oxide Nanoparticles: A Hybrid Multifunctional Platform with Tunable Magnetic Properties for Nanomedical Applications

    Hybrid materials composed of superparamagnetic iron oxide nanoparticles (SPIONs) and lipid self-assemblies possess considerable applicative potential in the biomedical field, specifically, for drug/nutrient delivery. Recently, we showed that SPIONs-doped lipid cubic liquid crystals undergo a cubic-to-hexagonal phase transition under the action of temperature or of an alternating magnetic field (AMF). This transition triggers the release of drugs embedded in the lipid scaffold or in the water channels. In this contribution, we address this phenomenon in depth, to fully elucidate the structural details and optimize the design of hybrid multifunctional carriers for drug delivery. Combining small-angle X-ray scattering (SAXS) with a magnetic characterization, we find that, in bulk lipid cubic phases, the cubic-to-hexagonal transition determines the magnetic response of SPIONs. We then extend the investigation from bulk liquid-crystalline phases to colloidal dispersions, i.e., to lipid/SPIONs nanoparticles with cubic internal structure (“magnetocubosomes”). Through Synchrotron SAXS, we monitor the structural response of magnetocubosomes while exposed to an AMF: the magnetic energy, converted into heat by SPIONs, activates the cubic-to-hexagonal transition, and can thus be used as a remote stimulus to spike drug release “on-demand”. In addition, we show that the AMF-induced phase transition in magnetocubosomes steers the realignment of SPIONs into linear string assemblies and connect this effect with the change in their magnetic properties, observed at the bulk level. Finally, we assess the internalization ability and cytotoxicity of magnetocubosomes in vitro on HT29 adenocarcinoma cancer cells, in order to test the applicability of these smart carriers in drug delivery applications

    Rapid Assessment of Susceptibility of Bacteria and Erythrocytes to Antimicrobial Peptides by Single-Cell Impedance Cytometry

    Antimicrobial peptides (AMPs) represent a promising classof compoundsto fight antibiotic-resistant infections. In most cases, they killbacteria by making their membrane permeable and therefore exhibitlow propensity to induce bacterial resistance. In addition, they areoften selective, killing bacteria at concentrations lower than thoseat which they are toxic to the host. However, clinical applicationsof AMPs are hindered by a limited understanding of their interactionswith bacteria and human cells. Standard susceptibility testing methodsare based on the analysis of the growth of a bacterial populationand therefore require several hours. Moreover, different assays arerequired to assess the toxicity to host cells. In this work, we proposethe use of microfluidic impedance cytometry to explore the actionof AMPs on both bacteria and host cells in a rapid manner and withsingle-cell resolution. Impedance measurements are particularly well-suitedto detect the effects of AMPs on bacteria, due to the fact that themechanism of action involves perturbation of the permeability of cellmembranes. We show that the electrical signatures of Bacillus megaterium cells and human red blood cells(RBCs) reflect the action of a representative antimicrobial peptide,DNS-PMAP23. In particular, the impedance phase at high frequency (e.g.,11 or 20 MHz) is a reliable label-free metric for monitoring DNS-PMAP23bactericidal activity and toxicity to RBCs. The impedance-based characterizationis validated by comparison with standard antibacterial activity assaysand absorbance-based hemolytic activity assays. Furthermore, we demonstratethe applicability of the technique to a mixed sample of B. megaterium cells and RBCs, which paves the wayto study AMP selectivity for bacterial versus eukaryotic cells inthe presence of both cell types

    Reducing healthcare-associated infections incidence by a probiotic-based sanitation system: A multicentre, prospective, intervention study

    Healthcare Associated Infections (HAI) are a global concern, further threatened by the increasing drug resistance of HAI-associated pathogens. On the other hand, persistent contamination of hospital surfaces contributes to HAI transmission, and it is not efficiently controlled by conventional cleaning, which does not prevent recontamination, has a high environmental impact and can favour selection of drug-resistant microbial strains. In the search for effective approaches, an eco-sustainable probiotic-based cleaning system (Probiotic Cleaning Hygiene System, PCHS) was recently shown to stably abate surface pathogens, without selecting antibiotic-resistant species. The aim of this study was to determine whether PCHS application could impact on HAI incidence. A multicentre, pre-post interventional study was performed for 18 months in the Internal Medicine wards of six Italian public hospitals (January 1st 2016-June 30th 2017). The intervention consisted of the substitution of conventional sanitation with PCHS, maintaining unaltered any other procedure influencing HAI control. HAI incidence in the pre and post-intervention period was the main outcome measure. Surface bioburden was also analyzed in parallel. Globally, 11,842 patients and 24,875 environmental samples were surveyed. PCHS was associated with a significant decrease of HAI cumulative incidence from a global 4.8% (284 patients with HAI over 5,930 total patients) to 2.3% (128 patients with HAI over 5,531 total patients) (OR = 0.44, CI 95% 0.35-0.54) (P<0.0001). Concurrently, PCHS was associated with a stable decrease of surface pathogens, compared to conventional sanitation (mean decrease 83%, range 70-96.3%), accompanied by a concurrent up to 2 Log drop of surface microbiota drug-resistance genes (P<0.0001; Pc = 0.008). Our study provides findings which support the impact of a sanitation procedure on HAI incidence, showing that the use of a probiotic-based environmental intervention can be associated with a significant decrease of the risk to contract a HAI during hospitalization. Once confirmed in larger experiences and other target populations, this eco-sustainable approach might be considered as a part of infection control and prevention (IPC) strategies.     Underwater intervention robotics: An outline of the Italian national project Maris

    The Italian national project MARIS (Marine Robotics for Interventions) pursues the strategic objective of studying, developing, and integrating technologies and methodologies to enable the development of autonomous underwater robotic systems employable for intervention activities. These activities are becoming progressively more typical for the underwater offshore industry, for search-and-rescue operations, and for underwater scientific missions. Within such an ambitious objective, the project consortium also intends to demonstrate the achievable operational capabilities at a proof-of-concept level by integrating the results with prototype experimental systems

    L’attività descritta nel presente elaborato è relativa alla prima fase di sviluppo di un innovativo sistema di caricamento automatico del filamento per una stampante 3D, prodotto da un’azienda che opera nel settore. In particolare, la progettazione meccanica qui illustrata è stata affrontata partendo dal progetto concettuale del nuovo dispositivo, per concludersi con il progetto costruttivo e il dimensionamento dei singoli componenti. In primo luogo, è stata effettuata un’analisi funzionale del problema, analizzando sistematicamente le specifiche tecniche richieste ed i vincoli di progetto dettati dall’azienda. Scomposto il problema in tal modo, sono state proposte ed analizzate soluzioni concettuali alternative, valutandone comparativamente punti di forza e criticità. Una volta individuata la soluzione ritenuta più vantaggiosa, si sono dimensionati opportunamente i componenti commerciali (e scelti a catalogo) e progettati quelli da realizzare ad hoc (con processo di produzione in additive manufacturing, secondo le richieste dell’azienda). L’elaborato è parte integrante di un progetto che prevede le successive fasi di prototipazione e programmazione del controller del sistema di caricamento automatico del filamento
