    Characterization of primary cilia during the differentiation of retinal ganglion cells in the zebrafish

    Background: Retinal ganglion cell (RGC) differentiation in vivo is a highly stereotyped process, likely resulting from the interaction of cell type-specific transcription factors and tissue-derived signaling factors. The primary cilium, as a signaling hub in the cell, may have a role during this process but its presence and localization during RGC generation, and its contribution to the process of cell differentiation, have not been previously assessed in vivo. Methods: In this work we analyzed the distribution of primary cilia in vivo using laser scanning confocal microscopy, as well as their main ultrastructural features by transmission electron microscopy, in the early stages of retinal histogenesis in the zebrafish, around the time of RGC generation and initial differentiation. In addition, we knocked-down ift88 and elipsa, two genes with an essential role in cilia generation and maintenance, a treatment that caused a general reduction in organelle size. The effect on retinal development and RGC differentiation was assessed by confocal microscopy of transgenic or immunolabeled embryos. Results: Our results show that retinal neuroepithelial cells have an apically-localized primary cilium usually protruding from the apical membrane. We also found a small proportion of sub-apical cilia, before and during the neurogenic period. This organelle was also present in an apical position in neuroblasts during apical process retraction and dendritogenesis, although between these stages cilia appeared highly dynamic regarding both presence and position. Disruption of cilia caused a decrease in the proliferation of retinal progenitors and a reduction of neural retina volume. In addition, retinal histogenesis was globally delayed albeit RGC layer formation was preferentially reduced with respect to the amacrine and photoreceptor cell layers. Conclusions: These results indicate that primary cilia exhibit a highly dynamic behavior during early retinal differentiation, and that they are required for the proliferation and survival of retinal progenitors, as well as for neuronal generation, particularly of RGCs

    Local models-based regression trees for very short-term wind speed prediction

    This paper evaluates the performance of different types of Regression Trees (RTs) in a real problem of very short-term wind speed prediction from measuring data in wind farms. RT is a solidly established methodology that, contrary to other soft-computing approaches, has been under-explored in problems of wind speed prediction in wind farms. In this paper we comparatively evaluate eight different types of RTs algorithms, and we show that they are able obtain excellent results in real problems of very short-term wind speed prediction, improving existing classical and soft-computing approaches such as multi-linear regression approaches, different types of neural networks and support vector regression algorithms in this problem.We also show that RTs have a very small computation time, that allows the retraining of the algorithms whenever new wind speed data are collected from the measuring towers.Ministerio de Ciencia y Tecnología ECO2010-22065-C03-02Ministerio de Ciencia y Tecnología TIN2011-28956-C02Junta de Andalucía P12-TIC-1728Universidad Pablo de Olavide APPB81309

    Evolutionary association rules for total ozone content modeling from satellite observations

    In this paper we propose an evolutionary method of association rules discovery (EQAR, Evolutionary Quan titative Association Rules) that extends a recently published algorithm by the authors and we describe its ap plication to a problem of Total Ozone Content (TOC) modeling in the Iberian Peninsula. We use TOC data from the Total Ozone Mapping Spectrometer (TOMS) on board the NASA Nimbus-7 satellite measured at three lo cations (Lisbon, Madrid and Murcia) of the Iberian Peninsula. As prediction variables for the association rules we consider several meteorological variables, such as Outgoing Long-wave Radiation (OLR), Temperature at 50 hPa level, Tropopause height, and wind vertical velocity component at 200 hPa. We show that the best as sociation rules obtained by EQAR are able to accurate modeling the TOC data in the three locations consid ered, providing results which agree to previous works in the literatur

    Longitudinally extensive transverse myelitis with AQP4 antibodies revealing ovarian teratoma.

    Paraneoplastic myelitis is a rare inflammatory disorder most frequently associated with solid tumors or lymphoproliferative disorders. Patients often harbor onconeuronal antibodies and their prognosis is usually poor. Here we report a 42-year old woman with longitudinally extensive transverse myelitis and aquaporin-4 (AQP4) antibodies that led to the diagnosis of ovarian teratoma. After tumor removal and immune therapy (including corticosteroids, plasma exchange, intravenous immunoglobulins and rituximab) the patient progressively improved achieving complete recovery. Histological study of the teratoma demonstrated neural tissue containing AQP4 expressing cells and intense inflammatory infiltrates, providing evidence for a possible paraneoplastic link between both disorders

    Chemical composition, in vitro cytotoxic and antioxidant activities of the essential oil of Peruvian Minthostachys mollis Griseb

    The composition of the essential oil obtained by hydrodistillation from Minthostachys mollis Griseb (Lamiaceae) aerial parts was determined by GC and GC/MS. Menthone (13.2%), pulegone (12.4%), cis-dihydrocarvone (9.8%) and carvacrol acetate (8.8%) were the main essential oil components. The cytotoxic activity of the essential oil was in vitro measured using the MTT colorimetric assay. IC50values were calculated on healthy non-tumor cells (HEK-293) and three human cancer cell lines (T24, DU-145 and MCF-7). In such latter cells, the estimated values were around 0.2 mg/mL. In addition, the antioxidant activity was determined by interaction with the stable freeradical 2,2”-diphenyl-1-picrylhydrazyl. The essential oil was almost devoid of antioxidant activity indicating that its anti-proliferative action relies on other unknown mechanism

    COPD Clinical Control: predictors and long-term follow-up of the CHAIN cohort

    CHAIN Study Investigators.[Background] Control in COPD is a dynamic concept that can reflect changes in patients’ clinical status that may have prognostic implications, but there is no information about changes in control status and its long-term consequences.[Methods] We classified 798 patients with COPD from the CHAIN cohort as controlled/uncontrolled at baseline and over 5 years. We describe the changes in control status in patients over long-term follow-up and analyze the factors that were associated with longitudinal control patterns and related survival using the Cox hazard analysis.[Results] 134 patients (16.8%) were considered persistently controlled, 248 (31.1%) persistently uncontrolled and 416 (52.1%) changed control status during follow-up. The variables significantly associated with persistent control were not requiring triple therapy at baseline and having a better quality of life. Annual changes in outcomes (health status, psychological status, airflow limitation) did not differ in patients, regardless of clinical control status. All-cause mortality was lower in persistently controlled patients (5.5% versus 19.1%, p = 0.001). The hazard ratio for all-cause mortality was 2.274 (95% CI 1.394–3.708; p = 0.001). Regarding pharmacological treatment, triple inhaled therapy was the most common option in persistently uncontrolled patients (72.2%). Patients with persistent disease control more frequently used bronchodilators for monotherapy (53%) at recruitment, although by the end of the follow-up period, 20% had scaled up their treatment, with triple therapy being the most frequent therapeutic pattern.[Conclusions] The evaluation of COPD control status provides relevant prognostic information on survival. There is important variability in clinical control status and only a small proportion of the patients had persistently good control. Changes in the treatment pattern may be relevant in the longitudinal pattern of COPD clinical control.
