Multiploidy occurrence in tomato calli from anther culture
Anther culture has long been used for the production of fully homozygous lines in order to produce, mainly, doubled haploid plants, which are of great interest in plant breeding. For tomato, a recalcitrant species for androgenesis production protocols have not been standardized. It is known that the genotype, anther size, the developmental stage of the microspore, and the medium composition are some factors that can influence the calli production. The present study aimed to adapt flow cytometry methodology to verify the microsporogenesis phases of anthers in order to assess the anther responsiveness of different tomato genotypes in an androgenesis-induction culture medium and to analyze the DNA ploidy level of calli produced by flow cytometry. Anthers from flower buds of length 1.0 to 5.9 mm, corresponding to the size range as analyzed by flow cytometry and cytogenetic methods, were inoculated into Murashige and Skoog (MS) basal medium containing the growth regulators 6-(y,ydimethylallylamino) purine and indole-3-acetic acid. The obtained calli were subsequently analyzed by flow cytometry to determine the DNA ploidy level. Surprisingly, despite no pretreatment with microtubule-depolymerizing agents, five classes of multiploid calli were observed, as follows: class I (2C-4C-8C-16C), class II (2C-4C-8C-16C-32C), class III (4C-8C), class IV (4C-8C-16C) and class V (8C-16C- 32C). Multiploid calli were identified in short-term (two month) culturing, suggesting that the variable culture duration did not directly influence the occurrence of endoreduplication. In this work, this type of somaclonal variation has been reported for the first time in tomato anther culture, and their possible origin has been discussed.Key words: Callogenesis, flow cytometry, polyploidy, Solanum lycopersicum, somaclonal variation
PARTICIPAÇÃO, VIABILIDADE E SUSTENTABILIDADE: DIMENSÕES DE DESENVOLVIMENTO LOCAL NUMA ASSOCIAÇÃO DE PRODUTORES RURAIS
The agribusiness unfolds in the rural Brazilian environment as one of the most efficient and competitive in the world but the rural poverty also blunts as one of the largest social blemishes for the society. The methodology applied was the study case, a milk producers’ association, comprehending some observation, semi-structured interviews and informal conversations. The study has evidenced that the association accomplishes several development indicators, job and income generation, access to rural credit, access to new technologies and important institutional partnerships. However, the association inclusion is still very limited to the associates and the social sustainability is fragile (there is an incidence of objective participation and low subjective participation), the administrative structure demands more attention regarding to the improvement intention. There is a possible social/economical instability in case of a competition incitation. However the reality is valid, provocative and highlighted into the region for the few associative enterprises successes and high level poverty, could be considered as an agent of great importance for the local development, but it still requires further preoccupation with the social side, with cooperative/associative education and more professionalism in the administration.participation, viability, sustainability and associativism.,

Gene therapy restores vision in rd1 mice after removal of a confounding mutation in Gpr179
The rd1 mouse with a mutation in the Pde6b gene was the first strain of mice identified with a retinal degeneration. However, AAV-mediated gene supplementation of rd1 mice only results in structural preservation of photoreceptors, and restoration of the photoreceptor-mediated a-wave, but not in restoration of the bipolar cell-mediated b-wave. Here we show that a mutation in Gpr179 prevents the full restoration of vision in rd1 mice. Backcrossing rd1 with C57BL6 mice reveals the complete lack of b-wave in a subset of mice, consistent with an autosomal recessive Mendelian inheritance pattern. We identify a mutation in the Gpr179 gene, which encodes for a G-protein coupled receptor localized to the dendrites of ON-bipolar cells. Gene replacement in rd1 mice that are devoid of the mutation in Gpr179 successfully restores the function of both photoreceptors and bipolar cells, which is maintained for up to 13 months. Our discovery may explain the failure of previous gene therapy attempts in rd1 mice, and we propose that Grp179 mutation status should be taken into account in future studies involving rd1 mice

Stressors in anaesthesiology: development and validation of a new questionnaire: A cross-sectional study of Portuguese anaesthesiologists
BACKGROUND:
Stress in anaesthesiologists is a common and multifactorial problem related to patients, colleagues and organisations. The consequences of stress include depression, work-home conflicts and burnout. Reduction in stress can be achieved by reducing the number and magnitude of stressors or by increasing resilience strategies.
OBJECTIVES:
We have created the self-reporting 'Stress Questionnaire in Anaesthesiologists' (SQA), to qualify the sources of stress in anaesthesiologists' professional lives, and measure the level of associated stress. Our study aimed to develop and validate the SQA using exploratory and confirmatory factor analyses. Construct validity was assessed through correlations between SQA and negative psychological outcomes as well as by comparing perception of stress among different known groups.
DESIGN:
A questionnaire-based cross-sectional, correlational, observational study.
SETTINGS:
The study was conducted between January 2014 and December 2014, throughout different anaesthesia departments in Portuguese hospitals. Data collection was from a representative subset at one specific time point.
PARTICIPANTS:
A sample of 710 anaesthesia specialists and residents from Portugal.
MAIN OUTCOME MEASURES:
The primary outcome measure was to identify specific stressors in anaesthesiologists. Secondary outcome was the association between stressors and burnout, depression symptoms, anxiety, stress, rumination, satisfaction with life and functional impairment.
RESULTS:
The exploratory analysis showed the SQA is a tri-dimensional instrument and confirmatory analysis showed the tri-dimensional structure presented good model fit. The three dimensions of SQA correlated positively with other stress measures and burnout, but negatively with satisfaction with life.
CONCLUSION:
SQA is a well adjusted measure for assessing stressors in anaesthesia physicians and includes clinical, organisational and team stress factors. Results showed that the SQA is a robust and reliable instrument.info:eu-repo/semantics/publishedVersio

International gestational age-specific centiles for umbilical artery Doppler indices: a longitudinal prospective cohort study of the INTERGROWTH-21st Project.
BACKGROUND: Reference values for umbilical artery Doppler indices are used clinically to assess fetal well-being. However, many studies that have produced reference charts have important methodologic limitations, and these result in significant heterogeneity of reported reference ranges. OBJECTIVES: To produce international gestational age-specific centiles for umbilical artery Doppler indices based on longitudinal data and the same rigorous methodology used in the original Fetal Growth Longitudinal Study of the INTERGROWTH-21st Project. STUDY DESIGN: In Phase II of the INTERGROWTH-21st Project (the INTERBIO-21st Study), we prospectively continued enrolling pregnant women according to the same protocol from 3 of the original populations in Pelotas (Brazil), Nairobi (Kenya), and Oxford (United Kingdom) that had participated in the Fetal Growth Longitudinal Study. Women with a singleton pregnancy were recruited at <14 weeks' gestation, confirmed by ultrasound measurement of crown-rump length, and then underwent standardized ultrasound every 5±1 weeks until delivery. From 22 weeks of gestation umbilical artery indices (pulsatility index, resistance index, and systolic/diastolic ratio) were measured in a blinded fashion, using identical equipment and a rigorously standardized protocol. Newborn size at birth was assessed using the international INTERGROWTH-21st Standards, and infants had detailed assessment of growth, nutrition, morbidity, and motor development at 1 and 2 years of age. The appropriateness of pooling data from the 3 study sites was assessed using variance component analysis and standardized site differences. Umbilical artery indices were modeled as functions of the gestational age using an exponential, normal distribution with second-degree fractional polynomial smoothing; goodness of fit for the overall models was assessed. RESULTS: Of the women enrolled at the 3 sites, 1629 were eligible for this study; 431 (27%) met the entry criteria for the construction of normative centiles, similar to the proportion seen in the original fetal growth longitudinal study. They contributed a total of 1243 Doppler measures to the analysis; 74% had 3 measures or more. The healthy low-risk status of the population was confirmed by the low rates of preterm birth (4.9%) and preeclampsia (0.7%). There were no neonatal deaths and satisfactory growth, health, and motor development of the infants at 1 and 2 years of age were documented. Only a very small proportion (2.8%-6.5%) of the variance of Doppler indices was due to between-site differences; in addition, standardized site difference estimates were marginally outside this threshold in only 1 of 27 comparisons, and this supported the decision to pool data from the 3 study sites. All 3 Doppler indices decreased with advancing gestational age. The 3rd, 5th 10th, 50th, 90th, 95th, and 97th centiles according to gestational age for each of the 3 indices are provided, as well as equations to allow calculation of any value as a centile and z scores. The mean pulsatility index according to gestational age = 1.02944 + 77.7456*(gestational age)-2 - 0.000004455*gestational age3. CONCLUSION: We present here international gestational age-specific normative centiles for umbilical artery Doppler indices produced by studying healthy, low-risk pregnant women living in environments with minimal constraints on fetal growth. The centiles complement the existing INTERGROWTH-21st Standards for assessment of fetal well-being
GESTÃO ESTRATÉGICA APLICADA AO COOPERATIVISMO SOLIDÁRIO: UMA ALTERNATIVA DE FORTALECIMENTO PARA OS AGRICULTORES FAMILIARES.
Este estudo teve como objetivo investigar, sob a ótica da administração, a Gestão Estratégica aplicada ao cooperativismo solidário como alternativa de fortalecimento organizacional. A intenção é conhecer as controvérsias a respeito da aplicação de ferramentas de gestão estratégica nos empreendimentos dessa natureza, mantendo suas características essenciais de cooperação, solidariedade e auto-gestão. Para isto buscou-se identificar um modelo de gestão capaz de facilitar o ganho de competitividade ao mesmo tempo em que mantenha as características de solidariedade e cooperação nos empreendimentos solidários. Utilizou-se o estudo de caso como método de pesquisa, tendo como objeto de análise uma pequena cooperativa de agricultores familiares. Onde se fez uma análise de como estão sendo aplicados na prática os princípios e valores dos Empreendimentos de Economia Solidária (EES) na Cooperativa de Agricultores Familiares de Itapuranga – COOPERAFI, bem como de seu modelo de gestão e estrutura organizacional. Analisou-se a aplicabilidade de instrumentos de gestão estratégica nas pequenas cooperativas, concluindo que os mesmos ajudam no desenvolvimento dos cooperados e da organização, sem necessariamente descaracterizar a cooperativa como empreendimento solidário.----------------------------------------------This study it had as objective to investigate, under the optics of the administration, the applied Strategical Management to the solidary cooperativism as an alternative to strengthen organizational. Where if it made an analysis of as they are being applied in practical the principles and values of the Enterprises of Economia Solidária (EES) in the Cooperative of Familiar Agriculturists of Itapuranga - COOPERAFI, as well as of its model of management and organizacional structure.
