Projeto vida: educação cientÃfica para estudantes do ensino fundamental anos iniciais / Life project: science education for elementary students early years
Em um contexto no qual a produção do conhecimento avança em ritmo acelerado, saber portar-se de maneira crÃtica e reflexiva frente aos desafios é uma caracterÃstica fundamental de cidadãos do século XXI. Nesse sentido, a educação cientÃfica tem se mostrado como uma importante aliada para a promoção da formação integral do sujeito. É nessa perspectiva que surgiu o Projeto Vida, com o intuito de estimular, desde a educação básica, indivÃduos a pensarem cientificamente. Através do trabalho com projetos, os estudantes mobilizam conhecimentos de várias áreas para resolucionar questões sociocientÃficas e socioambientais identificadas por eles mesmos, seja no ambiente escolar ou fora dele, sempre prezando por valores como sustentabilidade em todos os âmbitos: ambiental, social e cultural. Desta forma, as crianças desenvolvem diversas habilidades como a elaboração de hipóteses, a autonomia, a tomada de decisão, a argumentação, a cooperatividade, a criatividade, a investigação, entre outras, tornando-se protagonista da construção do seu processo cognoscitivo - a metacognição - aprendendo a aprender. Trata-se de uma pesquisa interventiva e formativa para a reconfiguração da prática docente de professoras de Pedagogia e Ciências Biológicas, de abordagem quantitativo/qualitativa. É exploratória, descritiva e analÃtica, tendo como público-alvo estudantes do ensino fundamental anos iniciais de uma escola privada do municÃpio de Natal/RN. Os instrumentos para coleta de dados são: observação por meio de categorias de análise, baseadas na metodologia de Laville e Dionne (1999), e também os projetos desenvolvidos pelas crianças. Os resultados expressam que as crianças já se familiarizaram com a linguagem da ciência, identificam situações-problema e propõem problemas do contexto local, sugerem estratégias de solução e apresentam atitudes positivas em relação à participação nos projetos.
Action potential variability in human pluripotent stem cell-derived cardiomyocytes obtained from healthy donors
Human pluripotent stem cells (PSC) have been used for disease modelling, after differentiation into the desired cell type. Electrophysiologic properties of cardiomyocytes derived from pluripotent stem cells are extensively used to model cardiac arrhythmias, in cardiomyopathies and channelopathies. This requires strict control of the multiple variables that can influence the electrical properties of these cells. In this article, we report the action potential variability of 780 cardiomyocytes derived from pluripotent stem cells obtained from six healthy donors. We analyze the overall distribution of action potential (AP) data, the distribution of action potential data per cell line, per differentiation protocol and batch. This analysis indicates that even using the same cell line and differentiation protocol, the differentiation batch still affects the results. This variability has important implications in modeling arrhythmias and imputing pathogenicity to variants encountered in patients with arrhythmic diseases. We conclude that even when using isogenic cell lines to ascertain pathogenicity to variants associated to arrythmias one should use cardiomyocytes derived from pluripotent stem cells using the same differentiation protocol and batch and pace the cells or use only cells that have very similar spontaneous beat rates. Otherwise, one may find phenotypic variability that is not attributable to pathogenic variants
R534C mutation in hERG causes a trafficking defect in iPSC-derived cardiomyocytes from patients with type 2 long QT syndrome
Patient-specific cardiomyocytes obtained from induced pluripotent stem cells (CM-iPSC) offer unprecedented mechanistic insights in the study of inherited cardiac diseases. The objective of this work was to study a type 2 long QT syndrome (LQTS2)-associated mutation (c.1600C > T in KCNH2, p.R534C in hERG) in CM-iPSC. Peripheral blood mononuclear cells were isolated from two patients with the R534C mutation and iPSCs were generated. In addition, the same mutation was inserted in a control iPSC line by genome editing using CRISPR/Cas9. Cells expressed pluripotency markers and showed spontaneous differentiation into the three embryonic germ layers. Electrophysiology demonstrated that action potential duration (APD) of LQTS2 CM-iPSC was significantly longer than that of the control line, as well as the triangulation of the action potentials (AP), implying a longer duration of phase 3. Treatment with the IKr inhibitor E4031 only caused APD prolongation in the control line. Patch clamp showed a reduction of IKr on LQTS2 CM-iPSC compared to control, but channel activation was not significantly affected. Immunofluorescence for hERG demonstrated perinuclear staining in LQTS2 CM-iPSC. In conclusion, CM-iPSC recapitulated the LQTS2 phenotype and our findings suggest that the R534C mutation in KCNH2 leads to a channel trafficking defect to the plasma membrane.Fil: Mesquita, Fernanda C. Fractional Distillation of Bio-Oil Produced by Pyrolysis of Açaà (Euterpe oleracea) Seeds
In this work, the seeds of açaà (Euterpe oleracea, Mart), a rich lignin-cellulose residue, has been submitted to pyrolysis to produce a bio-oil-like fossil fuels. The pyrolysis carried out in a reactor of 143 L, 450°C, and 1.0 atm. The morphology of Açaà seeds in nature and after pyrolysis is characterized by SEM, EDX, and XRD. The experiments show that bio-oil, gas, and coke yields were 4.38, 30.56, and 35.67% (wt.), respectively. The bio-oil characterized by AOCS, ASTM, and ABNT/NBR methods for density, kinematic viscosity, and acid value. The bio-oil density, viscosity, and acid value were 1.0468 g/cm3, 68.34 mm2/s, and 70.26 KOH/g, respectively. The chemical composition and chemical functions of bio-oil are determined by GC-MS and FT-IR. The GC-MS identified in bio-oil 21.52% (wt.) hydrocarbons and 78.48% (wt.) oxygenates (4.06% esters, 8.52% carboxylic acids, 3.53% ketones, 35.16% phenols, 20.52% cresols, 5.75% furans, and 0.91% (wt.) aldehydes), making it possible to apply fractional distillation to obtain fossil fuel-like fractions rich in hydrocarbons. The distillation of bio-oil is carried out in a laboratory-scale column, according to the boiling temperature of fossil fuels. The distillation of bio-oil yielded fossil fuel-like fractions (gasoline, kerosene, and light diesel) of 4.70, 28.21, and 22.35% (wt.), respectively
Preparation of a Nanoemulsion with Carapa guianensis
Andiroba (Carapa guianensis) seeds are the source of an oil with a wide range of biological activities and ethnopharmacological uses. However, few studies have devoted attention to innovative formulations, including nanoemulsions. The present study aimed to obtain a colloidal system with the andiroba oil using a low-energy and organic-solvent-free method. Moreover, the preliminary residual larvicidal activity of the nanoemulsion against Aedes aegypti was evaluated. Oleic and palmitic acids were the major fatty acids, in addition to the phytosterol β-sitosterol and limonoids (tetranortriterpenoids). The required hydrophile-lipophile was around 11.0 and the optimal nanoemulsion was obtained using polysorbate 85. The particle size distribution suggested the presence of small droplets (mean diameter around 150 nm) and low polydispersity index (around 0.150). The effect of temperature on particle size distribution revealed that no major droplet size increase occurred. The preliminary residual larvicidal assay suggested that the mortality increased as a function of time. The present study allowed achievement of a potential bioactive oil in water nanoemulsion that may be a promising controlled release system. Moreover, the ecofriendly approach involved in the preparation associated with the great bioactive potential of C. guianensis makes this nanoemulsion very promising for valorization of this Amazon raw material
Chemical Structure and Localization of Levan, the Predominant Fructan Type in Underground Systems of Gomphrena marginata (Amaranthaceae)
Gomphrena marginata Seub. (Amaranthaceae) is an endemic species from Brazilian campos rupestres with a fructan accumulating underground reserve system. Analyses of high performance anion exchange chromatography (HPAEC–PAD) revealed the presence of the soluble carbohydrates glucose, fructose, sucrose, 1-kestose, 6-kestose, nystose and fructans with degree of polymerization (DP) up to approximately 40 fructose units. Data of 1H and 13C Nuclear Magnetic Resonance (NMR) spectroscopy, including Heteronuclear Single-Quantum Correlation (HSQC) and Heteronuclear Multiple-Bonds Correlation (HMBC) showed the presence of β (2,6) linkages, characteristic of the linear molecule of levan-type fructan(2,6). These results confirmed previous studies suggesting that the reserve carbohydrate in the underground system of this species was levan-type fructans, similar to that of G. macrocephala. Structural analyses of the thickened underground system using light microscopy revealed a mixed origin system consisting mainly of a gemmiferous tuberous root with the upper region formed by short branched stems, both presenting vascular cylinders with unusual growth patterns. Fructan spherocrystals were visualized under polarized light and scanning electron microscopy (SEM) mostly in the cortex and vascular cylinder in both thickened stem and root. In addition to data reported in the literature concerning the occurrence of fructans in the Amaranthaceae, the results presented here suggest that fructans are a trait in this family while the levan-type fructan prevail in Gomphrena species
Pathogen-Specific Epitopes as Epidemiological Tools for Defining the Magnitude of Mycobacterium leprae Transmission in Areas Endemic for Leprosy
During recent years, comparative genomic analysis has allowed the identification of Mycobacterium leprae-specific genes with potential application for the diagnosis of leprosy. In a previous study, 58 synthetic peptides derived from these sequences were tested for their ability to induce production of IFN-γ in PBMC from endemic controls (EC) with unknown exposure to M. leprae, household contacts of leprosy patients and patients, indicating the potential of these synthetic peptides for the diagnosis of sub- or preclinical forms of leprosy. In the present study, the patterns of IFN-γ release of the individuals exposed or non-exposed to M. leprae were compared using an Artificial Neural Network algorithm, and the most promising M. leprae peptides for the identification of exposed people were selected. This subset of M. leprae-specific peptides allowed the differentiation of groups of individuals from sites hyperendemic for leprosy versus those from areas with lower level detection rates. A progressive reduction in the IFN-γ levels in response to the peptides was seen when contacts of multibacillary (MB) patients were compared to other less exposed groups, suggesting a down modulation of IFN-γ production with an increase in bacillary load or exposure to M. leprae. The data generated indicate that an IFN-γ assay based on these peptides applied individually or as a pool can be used as a new tool for predicting the magnitude of M. leprae transmission in a given population
