    Patterns of urediniospores release and development of rose apple rust

    O objetivo deste trabalho foi avaliar padrões epidemiológicos de campo do fungo (Puccinia psidii Winter) que causa a ferrugem em jambeiro (Syzygium jambos). Entre 29/3 e 31/12/1988, levantou-se a incidência da doença e a população aérea de urediniósporos, na Universidade de Brasília, DF. Avaliaram-se dez plantas, sendo que, entre duas delas, instalou-se uma armadilha Burkard caça-esporos de sete dias. A maior quantidade de urediniósporos capturada foi de 23 de junho a 6 de julho. A principal carga de esporos ocorreu de maio a julho, entre as 10h e as 13 horas. Os maiores números de brotos de jambeiro com pústulas esporulantes ocorreram entre 28 de abril e 20 de julho. O número de brotos terminais infectados foi positivamente correlacionado com a quantidade de esporos. A temperatura (às 12h) foi negativamente correlacionada com a quantidade de urediniósporos dispersos no ar. O número de dias com umidade relativa (UR) do ar (às 24h) ³80% foi positivamente correlacionado com a quantidade de brotos deformados. A quantidade de urediniósporos foi positivamente correlacionada com o número de dias com temperatura £20oC e UR(24h) ³80%. Ao maior pico de urediniósporos capturados precedeu um período sem precipitação, porém com vários dias de UR(24h) ³80% e temperatura £20oC. Estes dados mostraram que a doença foi favorecida por temperaturas £20oC e por UR noturna ³80%.This research was conducted to evaluate some epidemiological features of the rose apple (Syzygium jambos) rust (Puccinia psidii Winter) under field conditions at the Universidade de Brasília, Brasília, DF, Brazil, from March 29 through December 31, 1988. Data for the amount of airborne urediniospores were collected in ten plants, using a Burkard's seven-day volumetric spore trap set between two rose apple trees. Most of the airborne urediniospores were collected between June 23 and July 6, from 10 a.m. to 1 p.m. The major quantity of S. jambos affected shoots occurred between April 28 and July 20. There was a positive correlation between the amount of trapped urediniospores and infected young shoots. A negative correlation was shown between temperature (12h) and number of airborne urediniospores. The number of days with relative humidity (RH) ³80% was positively correlated to the amount of deformed shoots. Also, the amount of urediniospores and infected young shoots were positively correlated to the number of days with night (24h) temperature £20oC or night RH³80%. These data show that this disease was favored by low temperatures (£20oC), high night RH (³80%) and high levels of airborne urediniospores

    Non-stationary wave relaxation methods for general linear systems of Volterra equations: convergence and parallel GPU implementation

    Producción CientíficaIn the present paper, a parallel-in-time discretization of linear systems of Volterra equations of type u¯(t)=u¯0+∫t0K(t−s)u¯(s) d s+f¯(t),0<t≤T, is addressed. Related to the analytical solution, a general enough functional setting is firstly stated. Related to the numerical solution, a parallel numerical scheme based on the Non-Stationary Wave Relaxation (NSWR) method for the time discretization is proposed, and its convergence is studied as well. A CUDA parallel implementation of the method is carried out in order to exploit Graphics Processing Units (GPUs), which are nowadays widely employed for reducing the computational time of several general purpose applications. The performance of these methods is compared to some sequential implementation.     Sequence Analysis of the UCP1 Gene in a Severe Obese Population from Southern Italy

    Brown adipose tissue, where Uncoupling Protein 1 (UCP1) activity uncouples mitochondrial respiration, is an important site of facultative energy expenditure. This tissue may normally function to prevent obesity. Our aim was to investigate by sequence analysis the presence of UCP1 gene variations that may be associated with obesity. We studied 100 severe obese adults (BMI > 40 kg/m2) and 100 normal-weight control subjects (BMI range = 19–24.9 kg/m2). We identified 7 variations in the promoter region, 4 in the intronic region and 4 in the exonic region. Globally, 72% of obese patients bore UCP1 polymorphisms. Among UCP1 variants, g.IVS4−208T>G SNP was associated with obesity (OR: 1.77; 95% CI = 1.26–2.50; P = .001). Further, obese patients bearing the g.−451C>T (CT+TT) or the g.940G>A (GA+AA) genotypes showed a higher BMI than not polymorphic obese patients (P = .008 and P = .043, resp.). In conclusion, UCP1 SNPs could represent “thrifty” factors that promote energy storage in prone subjects

    Detailed peptide profiling of “Scotta”: from a dairy waste to a source of potential health-promoting compound

    “Scotta” is a liquid waste deriving from Ricotta cheese production, which is wrongly considered only a dairy by-product. In this work, with the aim to elucidate the presence of valuable bioactive compounds in Buffalo’s Scotta, a peptide fraction under 3000 Da was isolated by ultra-filtration, purified by solid-phase extraction, and,subsequently, characterized in detail by liquid chromatography coupled to Orbitrap mass spectrometry. Analytical results revealed a complex profile, leading to the identification of 226 peptides, belonging to alpha, beta, and kappa caseins. A database-driven search approach was used to assess the biological effects of some of the identified peptides. A wide range of healthy properties was ascribed to the encrypted peptides, comprising antihypertensive, antimicrobial, immunomodulating, opioid, antioxidant, and antithrombotic. The peptidomic profile of Scotta was highlighted in depth for the first time, and the results revealed that this matrix should not be considered only a mere by-product, but a source of potential health-promoting peptides, which can be recovered and employed in nutraceuticals and functional foods

    A metabolic imbalance between anabolic drive and catabolic forces is commonly observed in chronic heart failure (CHF) patients, with the latter prevailing over anabolic hormones. Moreover, anabolic deficiencies are independent markers of poor prognosis. This finding represents a solid background for the implementation of therapeutic trials based on replacement therapy. The somatotropic axis (GH/IGF-1) is the most powerful anabolic axis of the body and its decline is related with a poor outcome and a worse clinical status. Growth hormone (GH) administration may enter the therapeutic arena as adjunctive treatment in patients affected by CHF and GH/IGF-1 deficiency.     Impact of safety-related dose reductions or discontinuations on sustained virologic response in HCV-infected patients: Results from the GUARD-C Cohort

    BACKGROUND: Despite the introduction of direct-acting antiviral agents for chronic hepatitis C virus (HCV) infection, peginterferon alfa/ribavirin remains relevant in many resource-constrained settings. The non-randomized GUARD-C cohort investigated baseline predictors of safety-related dose reductions or discontinuations (sr-RD) and their impact on sustained virologic response (SVR) in patients receiving peginterferon alfa/ribavirin in routine practice. METHODS: A total of 3181 HCV-mono-infected treatment-naive patients were assigned to 24 or 48 weeks of peginterferon alfa/ribavirin by their physician. Patients were categorized by time-to-first sr-RD (Week 4/12). Detailed analyses of the impact of sr-RD on SVR24 (HCV RNA <50 IU/mL) were conducted in 951 Caucasian, noncirrhotic genotype (G)1 patients assigned to peginterferon alfa-2a/ribavirin for 48 weeks. The probability of SVR24 was identified by a baseline scoring system (range: 0-9 points) on which scores of 5 to 9 and <5 represent high and low probability of SVR24, respectively. RESULTS: SVR24 rates were 46.1% (754/1634), 77.1% (279/362), 68.0% (514/756), and 51.3% (203/396), respectively, in G1, 2, 3, and 4 patients. Overall, 16.9% and 21.8% patients experienced 651 sr-RD for peginterferon alfa and ribavirin, respectively. Among Caucasian noncirrhotic G1 patients: female sex, lower body mass index, pre-existing cardiovascular/pulmonary disease, and low hematological indices were prognostic factors of sr-RD; SVR24 was lower in patients with 651 vs. no sr-RD by Week 4 (37.9% vs. 54.4%; P = 0.0046) and Week 12 (41.7% vs. 55.3%; P = 0.0016); sr-RD by Week 4/12 significantly reduced SVR24 in patients with scores <5 but not 655. CONCLUSIONS: In conclusion, sr-RD to peginterferon alfa-2a/ribavirin significantly impacts on SVR24 rates in treatment-naive G1 noncirrhotic Caucasian patients. Baseline characteristics can help select patients with a high probability of SVR24 and a low probability of sr-RD with peginterferon alfa-2a/ribavirin

    Inhibition of the Soluble Epoxide Hydrolase Promotes Albuminuria in Mice with Progressive Renal Disease

    Epoxyeicotrienoic acids (EETs) are cytochrome P450-dependent anti-hypertensive and anti-inflammatory derivatives of arachidonic acid, which are highly abundant in the kidney and considered reno-protective. EETs are degraded by the enzyme soluble epoxide hydrolase (sEH) and sEH inhibitors are considered treatment for chronic renal failure (CRF). We determined whether sEH inhibition attenuates the progression of CRF in the 5/6-nephrectomy model (5/6-Nx) in mice. 5/6-Nx mice were treated with a placebo, an ACE-inhibitor (Ramipril, 40 mg/kg), the sEH-inhibitor cAUCB or the CYP-inhibitor fenbendazole for 8 weeks. 5/6-Nx induced hypertension, albuminuria, glomerulosclerosis and tubulo-interstitial damage and these effects were attenuated by Ramipril. In contrast, cAUCB failed to lower the blood pressure and albuminuria was more severe as compared to placebo. Plasma EET-levels were doubled in 5/6 Nx-mice as compared to sham mice receiving placebo. Renal sEH expression was attenuated in 5/6-Nx mice but cAUCB in these animals still further increased the EET-level. cAUCB also increased 5-HETE and 15-HETE, which derive from peroxidation or lipoxygenases. Similar to cAUCB, CYP450 inhibition increased HETEs and promoted albuminuria. Thus, sEH-inhibition failed to elicit protective effects in the 5/6-Nx model and showed a tendency to aggravate the disease. These effects might be consequence of a shift of arachidonic acid metabolism into the lipoxygenase pathway
