    Estudio y caracterización de los áridos reciclados procedentes de piezas prefabricadas para la elaboración de hormigón estructural

    Los procesos de fabricación deben ser pensados y diseñados sabiendo que el material va a ser, en algún momento de su vida, reciclado y usado posteriormente como materia prima, y es por ello que debe fabricarse desde el inicio pensando en esta cualidad nueva, y si no es posible hacerlo quedará relegado por otros que si la posean. Debemos llegar al extremo de considerar los residuos como materia prima, con cualidades perfectamente válidas para poder ser empleados como tales, y cumplir con el concepto “cradle to cradle”, ya planteado en 2010 por Michael Braugant y Willian Mcdonough [3]. En este trabajo se plantea el uso de un árido más específico que el usado procedente de la demolición de estructuras de hormigón estudiado en contadas ocasiones [1], se plantea el uso del árido procedente de piezas prefabricadas de hormigón que pasan a ser desechadas [2]. En las plantas de prefabricación, el residuo generado a partir de las piezas que no pasan el control de calidad llega a ser aproximadamente el 5% de la producción. Haciendo la consideración de que la producción de piezas prefabricadas en España está en torno al 22% de la producción total de hormigón, es indudable que merece la pena realizar el estudio que permita determinar la posibilidad de reutilizarlo como materia prima en la realización de nuevas piezas dentro de la misma fábrica. Siguiendo los pasos del trabajo expuesto en el “Estudio Experimental sobre Propiedades Mecánicas del Hormigón Reciclado con Áridos Procedentes de la no Calidad en Prefabricación , desarrollado por D. José Ángel Pérez Benedicto. En la presente comunicación se expone como se ha procedido a la caracterización del árido procedente del machaqueo de piezas prefabricadas que no han superado controles de calidad. Cómo árido de referencia se procede a ensayar un árido calizo, como árido más comúnmente utilizado en la fabricación de hormigones. Los dos tipos de árido se analizan frente a las diferentes exigencias marcadas en la normativa vigente en España, la “Instrucción de hormigón estructural (EHE-08) según Real Decreto 1247/2008, de 18 de julio.     血液脳関門開放術による遺伝子治療法の開発 --身体を傷つけない脳疾患の治療を目指して--. 京都大学プレスリリース. 2023-04-20.Intracerebral vector delivery in nonhuman primates has been a major challenge. We report successful blood-brain barrier opening and focal delivery of adeno-associated virus serotype 9 vectors into brain regions involved in Parkinson’s disease using low-intensity focus ultrasound in adult macaque monkeys. Openings were well tolerated with generally no associated abnormal magnetic resonance imaging signals. Neuronal green fluorescent protein expression was observed specifically in regions with confirmed blood-brain barrier opening. Similar blood-brain barrier openings were safely demonstrated in three patients with Parkinson’s disease. In these patients and in one monkey, blood-brain barrier opening was followed by 18F-Choline uptake in the putamen and midbrain regions based on positron emission tomography. This indicates focal and cellular binding of molecules that otherwise would not enter the brain parenchyma. The less-invasive nature of this methodology could facilitate focal viral vector delivery for gene therapy and might allow early and repeated interventions to treat neurodegenerative disorders

    Evaluation of the humoral and mucosal immune response of a multiepitope vaccine against COVID-19 in pigs

    IntroductionThis study evaluated the immune response to a multiepitope recombinant chimeric protein (CHIVAX) containing B- and T-cell epitopes of the SARS-CoV-2 spike’s receptor binding domain (RBD) in a translational porcine model for pre-clinical studies.MethodsWe generated a multiepitope recombinant protein engineered to include six coding conserved epitopes from the RBD domain of the SARS-CoV-2 S protein. Pigs were divided into groups and immunized with different doses of the protein, with serum samples collected over time to determine antibody responses by indirect ELISA and antibody titration. Peptide recognition was also analyzed by Western blotting. A surrogate neutralization assay with recombinant ACE2 and RBDs was performed. Intranasal doses of the immunogen were also prepared and tested on Vietnamese minipigs.ResultsWhen the immunogen was administered subcutaneously, it induced specific IgG antibodies in pigs, and higher doses correlated with higher antibody levels. Antibodies from immunized pigs recognized individual peptides in the multiepitope vaccine and inhibited RBD-ACE2 binding for five variants of concern (VOC). Comparative antigen delivery methods showed that both, subcutaneous and combined subcutaneous/intranasal approaches, induced specific IgG and IgA antibodies, with the subcutaneous approach having superior neutralizing activity. CHIVAX elicited systemic immunity, evidenced by specific IgG antibodies in the serum, and local mucosal immunity, indicated by IgA antibodies in saliva, nasal, and bronchoalveolar lavage secretions. Importantly, these antibodies demonstrated neutralizing activity against SARS-CoV-2 in vitro.DiscussionThe elicited antibodies recognized individual epitopes on the chimeric protein and demonstrated the capacity to block RBD-ACE2 binding of the ancestral SARS-CoV-2 strain and four VOCs. The findings provide proof of concept for using multiepitope recombinant antigens and a combined immunization protocol to induce a neutralizing immune response against SARS-CoV-2 in the pig translational model for preclinical studies

Dendritic cells (DCs) play a key role in the defense against viral infections, for instance plasmacytoid DCs (pDCs), have the capacity to produce vast amounts of interferon-alpha (IFN-α). In COVID-19 there is a deficit in DC numbers and IFN-α production, which has been associated with disease severity. In this work, we described that in addition to the DC deficiency, several DC activation and homing markers were altered in acute COVID-19 patients, which were associated with multiple inflammatory markers.     Deciphering the quality of SARS-CoV-2 specific T-cell response associated with disease severity, immune memory and heterologous response

    SARS-CoV-2 specific T-cell response has been associated with disease severity, immune memory and heterologous response to endemic coronaviruses. However, an integrative approach combining a comprehensive analysis of the quality of SARS-CoV-2 specific T-cell response with antibody levels in these three scenarios is needed. In the present study, we found that, in acute infection, while mild disease was associated with high T-cell polyfunctionality biased to IL-2 production and inversely correlated with anti-S IgG levels, combinations only including IFN-γ with the absence of perforin production predominated in severe disease. Seven months after infection, both non-hospitalised and previously hospitalised patients presented robust anti-S IgG levels and SARS-CoV-2 specific T-cell response. In addition, only previously hospitalised patients showed a T-cell exhaustion profile. Finally, combinations including IL-2 in response to S protein of endemic coronaviruses were the ones associated with SARS-CoV-2 S-specific T-cell response in pre-COVID-19 healthy donors’ samples.
