    Snow spectral albedo at Summit, Greenland: measurements and numerical simulations based on physical and chemical properties of the snowpack

    The broadband albedo of surface snow is determined both by the near-surface profile of the physical and chemical properties of the snowpack and by the spectral and angular characteristics of the incident solar radiation. Simultaneous measurements of the physical and chemical properties of snow were carried out at Summit Camp, Greenland (72°36´ N, 38°25´ W, 3210 m a.s.l.) in May and June 2011, along with spectral albedo measurements. One of the main objectives of the field campaign was to test our ability to predict snow spectral albedo by comparing the measured albedo to the albedo calculated with a radiative transfer model, using measured snow physical and chemical properties. To achieve this goal, we made daily measurements of the snow spectral albedo in the range 350–2200 nm and recorded snow stratigraphic information down to roughly 80 cm. The snow specific surface area (SSA) was measured using the DUFISSS instrument (DUal Frequency Integrating Sphere for Snow SSA measurement, Gallet et al., 2009). Samples were also collected for chemical analyses including black carbon (BC) and dust, to evaluate the impact of light absorbing particulate matter in snow. This is one of the most comprehensive albedo-related data sets combining chemical analysis, snow physical properties and spectral albedo measurements obtained in a polar environment. The surface albedo was calculated from density, SSA, BC and dust profiles using the DISORT model (DIScrete Ordinate Radiative Transfer, Stamnes et al., 1988) and compared to the measured values. Results indicate that the energy absorbed by the snowpack through the whole spectrum considered can be inferred within 1.10%. This accuracy is only slightly better than that which can be obtained considering pure snow, meaning that the impact of impurities on the snow albedo is small at Summit. In the near infrared, minor deviations in albedo up to 0.014 can be due to the accuracy of radiation and SSA measurements and to the surface roughness, whereas deviations up to 0.05 can be explained by the spatial heterogeneity of the snowpack at small scales, the assumption of spherical snow grains made for DISORT simulations and the vertical resolution of measurements of surface layer physical properties. At 1430 and around 1800 nm the discrepancies are larger and independent of the snow properties; we propose that they are due to errors in the ice refractive index at these wavelengths. This work contributes to the development of physically based albedo schemes in detailed snowpack models, and to the improvement of retrieval algorithms for estimating snow properties from remote sensing data

    Spermidine Associated to Non-Surgical Treatment of Periodontal Disease: Split Mouth Randomized Controlled Clinical Trial

    Objectives. Spermidine is an endogenous polyamine whose increase induces the autocrine remodeling into targeted cells. The aim of the present study was to evaluate the adjunctive beneficial effects of the local delivery of spermidine to periodontal non-surgical therapy on reduction of periodontal pocket. Methods. In this split mouth, blind, randomized controlled clinical trial, 20 patients with severe chronic generalized periodontal disease scheduled for cause related non-surgical periodontal treatment were enrolled. In three quadrants for each patients, three experimental teeth with probing pocket depth (PPD) 65 6mm were selected and randomly assigned to one of the following treatments: scaling and root planning (SRP)+ spermidine gel (spermidine + alginate) (group a), SRP+ placebo gel (only alginate) (positive control, group b) or SRP (negative control, group c). PPD at baseline, 3 and 6 months after treatment was assessed. Descriptive and inferential statistics was done. Results. After periodontal treatment, a reduction of PPD was observed in all sites. No differences emerged between groups at each time point. In all groups, a significant PPD reduction was observed at T1 and T2 compared to baseline (p<0.05 and p<0.001). PPD reduction from T1 to T2 was significant only in group a (p<0.001). Conclusions. After non-surgical treatment, local delivery of spermidine may induce prolonged improvement of clinical outcome

    In situ Forming Hyperbranched PEG—Thiolated Hyaluronic Acid Hydrogels With Honey-Mimetic Antibacterial Properties

    The rapidly increasing resistance of bacteria to currently approved antibiotic drugs makes surgical interventions and the treatment of bacterial infections increasingly difficult. In recent years, complementary strategies to classical antibiotic therapy have, therefore, gained importance. One of these strategies is the use of medicinal honey in the treatment of bacterially colonized wounds. One of the several bactericidal effects of honey is based on the in situ generation of hydrogen peroxide through the activity of the enzyme glucose oxidase. The strategy underlying this work is to mimic this antibacterial redox effect of honey in an injectable, biocompatible, and rapidly forming hydrogel. The hydrogel was obtained by thiol–ene click reaction between hyperbranched polyethylene glycol diacrylate (HB PEGDA), synthesized using reversible addition-fragmentation chain transfer (RAFT) polymerization, and thiolated hyaluronic acid (HA-SH). After mixing 500 µL HB PEGDA (10%, w/w) and 500 µL HA-SH (1%, w/w) solutions, hydrogels formed in ∼60 s (HB PEGDA/HA-SH 10.0–1.0), as assessed by the tube inverting test. The HB PEGDA/HA-SH 10.0–1.0 hydrogel (200 µL) was resistant to in vitro dissolution in water for at least 64 days, absorbing up to 130 wt% of water. Varying glucose oxidase (GO) amounts (0–500 U/L) and constant glucose content (2.5 wt%) were loaded into HB PEGDA and HA-SH solutions, respectively, before hydrogel formation. Then, the release of H2O2 was evaluated through a colorimetric pertitanic acid assay. The GO content of 250 U/L was selected, allowing the formation of 10.8 ± 1.4 mmol H2O2/L hydrogel in 24 h, under static conditions. The cytocompatibility of HB PEGDA/HA-SH 10.0–1.0 hydrogels loaded with different GO activities (≤ 500 U/L) at a constant glucose amount (2.5 wt%) was investigated by in vitro assays at 24 h with L929 and HaCaT cell lines, according to DIN EN ISO 10993-5. The tests showed cytocompatibility for GO enzyme activity up to 250 U/L for both cell lines. The antibacterial activity of HB PEGDA/HA-SH 10.0–1.0 hydrogels loaded with increasing amounts of GO was demonstrated against various gram-positive bacteria (S. aureus and S. epidermidis), antibiotic-resistant gram-positive bacteria (MRSA and MRSE), gram-negative bacteria (P. aeruginosa, E. coli, and A. baumanii), and antibiotic-resistant gram-negative strains (P. aeruginosa and E. coli) using agar diffusion tests. For all gram-positive bacterial strains, increasing efficacy was measured with increasing GO activity. For the two P. aeruginosa strains, efficacy was shown only from an enzyme activity of 125 U/L and for E. coli and A. baumanii, efficacy was shown only from 250 U/L enzyme activity. HB PEGDA/HA-SH 10.0–1.0 hydrogels loaded with ≤250 U/L GO and 2.5 wt% glucose are promising formulations due to their fast-forming properties, cytocompatibility, and ability to produce antibacterial H2O2, warranting future investigations for bacterial infection treatment, such as wound care

    Medical-grade silicone coated with rhamnolipid R89 is effective against Staphylococcus spp. Biofilms

    Staphylococcus aureus and Staphylococcus epidermidis are considered two of the most important pathogens, and their biofilms frequently cause device-associated infections. Microbial biosurfactants recently emerged as a new generation of anti-adhesive and anti-biofilm agents for coating implantable devices to preserve biocompatibility. In this study, R89 biosurfactant (R89BS) was evaluated as an anti-biofilm coating on medical-grade silicone. R89BS is composed of homologues of the mono- (75%) and di-rhamnolipid (25%) families, as evidenced by mass spectrometry analysis. The antimicrobial activity against Staphylococcus spp. planktonic and sessile cells was evaluated by microdilution and metabolic activity assays. R89BS inhibited S. aureus and S. epidermidis growth with minimal inhibitory concentrations (MIC99) of 0.06 and 0.12 mg/mL, respectively and dispersed their pre-formed biofilms up to 93%. Silicone elastomeric discs (SEDs) coated by R89BS simple adsorption significantly counteracted Staphylococcus spp. biofilm formation, in terms of both built-up biomass (up to 60% inhibition at 72 h) and cell metabolic activity (up to 68% inhibition at 72 h). SEM analysis revealed significant inhibition of the amount of biofilm-covered surface. No cytotoxic effect on eukaryotic cells was detected at concentrations up to 0.2 mg/mL. R89BS-coated SEDs satisfy biocompatibility requirements for leaching products. Results indicate that rhamnolipid coatings are effective anti-biofilm treatments and represent a promising strategy for the prevention of infection associated with implantable devices

    Marker-Assisted Pyramiding of Blast-Resistance Genes in a japonica Elite Rice Cultivar through Forward and Background Selection

    Rice blast, caused by Pyricularia oryzae, is one of the main rice diseases worldwide. The pyramiding of blast-resistance (Pi) genes, coupled to Marker-Assisted BackCrossing (MABC), provides broad-spectrum and potentially durable resistance while limiting the donor genome in the background of an elite cultivar. In this work, MABC coupled to foreground and background selections based on KASP marker assays has been applied to introgress four Pi genes (Piz, Pib, Pita, and Pik) in a renowned japonica Italian rice variety, highly susceptible to blast. Molecular analyses on the backcross (BC) lines highlighted the presence of an additional blast-resistance gene, the Pita-linked Pita2/Ptr gene, therefore increasing the number of blast-resistance introgressed genes to five. The recurrent genome was recovered up to 95.65%. Several lines carrying four (including Pita2) Pi genes with high recovery percentage levels were also obtained. Phenotypic evaluations confirmed the effectiveness of the pyramided lines against multivirulent strains, which also had broad patterns of resistance in comparison to those expected based on the pyramided Pi genes. The developed blast-resistant japonica lines represent useful donors of multiple blast-resistance genes for future rice-breeding programs related to the japonica group

    Property-based interest propagation in ontology-based user model

    We present an approach for propagation of user interests in ontology-based user models taking into account the properties declared for the concepts in the ontology. Starting from initial user feedback on an object, we calculate user interest in this particular object and its properties and further propagate user interest to other objects in the ontology, similar or related to the initial object. The similarity and relatedness of objects depends on the number of properties they have in common and their corresponding values. The approach we propose can support finer recommendation modalities, considering the user interest in the objects, as well as in singular properties of objects in the recommendation process. We tested our approach for interest propagation with a real adaptive application and obtained an improvement with respect to IS-A-propagation of interest values

    Propagating user interests in ontology-based user model

    In this paper we address the problem of propagating user interests in ontology-based user models. Our ontology-based user model (OBUM) is devised as an overlay over the domain ontology. Using ontologies as the basis of the user profile allows the initial user behavior to be matched with existing concepts in the domain ontology. Such ontological approach to user profiling has been proven successful in addressing the cold-start problem in recommender systems, since it allows for propagation from a small number of initial concepts to other related domain concepts by exploiting the ontological structure of the domain. The main contribution of the paper is the novel algorithm for propagation of user interests which takes into account i) the ontological structure of the domain and, in particular, the level at which each domain item is found in the ontology; ii) the type of feedback provided by the user, and iii) the amount of past feedback provided for a certain domain object

    Extrinsic primary afferent signalling in the gut

    Visceral sensory neurons activate reflex pathways that control gut function and also give rise to important sensations, such as fullness, bloating, nausea, discomfort, urgency and pain. Sensory neurons are organised into three distinct anatomical pathways to the central nervous system (vagal, thoracolumbar and lumbosacral). Although remarkable progress has been made in characterizing the roles of many ion channels, receptors and second messengers in visceral sensory neurons, the basic aim of understanding how many classes there are, and how they differ, has proven difficult to achieve. We suggest that just five structurally distinct types of sensory endings are present in the gut wall that account for essentially all of the primary afferent neurons in the three pathways. Each of these five major structural types of endings seems to show distinctive combinations of physiological responses. These types are: 'intraganglionic laminar' endings in myenteric ganglia; 'mucosal' endings located in the subepithelial layer; 'muscular–mucosal' afferents, with mechanosensitive endings close to the muscularis mucosae; 'intramuscular' endings, with endings within the smooth muscle layers; and 'vascular' afferents, with sensitive endings primarily on blood vessels. 'Silent' afferents might be a subset of inexcitable 'vascular' afferents, which can be switched on by inflammatory mediators.
