    TBL1 is required for the mesenchymal phenotype of transformed breast cancer cells

    The epithelial-to-mesenchymal transition (EMT) and its reversion (MET) are related to tumor cell dissemination and migration, tumor circulating cell generation, cancer stem cells, chemoresistance, and metastasis formation. To identify chromatin and epigenetic factors possibly involved in the process of EMT, we compare the levels of expression of epigenetic genes in a transformed human breast epithelial cell line (HMEC-RAS) versus a stable clone of the same cell line expressing the EMT master regulator ZEB1 (HMEC-RAS-ZEB1). One of the factors strongly induced in the HMEC-RAS-ZEB1 cells was Transducin beta-like 1 (TBL1), a component of the NCoR complex, which has both corepressor and coactivator activities. We show that TBL1 interacts with ZEB1 and that both factors cooperate to repress the promoter of the epithelial gene E-cadherin (CDH1) and to autoactivate the ZEB1 promoter. Consistent with its central role, TBL1 is required for mesenchymal phenotypes of transformed breast epithelial and breast cancer cell lines of the claudin-low subtype. Importantly, a high expression of the TBL1 gene correlates with poor prognosis and increased proportion of metastasis in breast cancer patients, indicating that the level of TBL1 expression can be used as a prognostic marker.Ministerio de Economía y Competitividad BFU2014-53543-P, BFU2017-85420-RJunta de Andalucía BIO-32

    This final project relates to the field of business administration and technology, as it seeks to analyze aspects of the market potential of an online shopping application in the gift segment and the formulation of a marketing plan. It is part of a double diploma program between Instituto Politécnico de Bragança and Federal University of Technology - Paraná. It is critical to recognize the market potential in order to understand a company's external environment and identify how the business can be inserted in the market to expand and consolidate. We are currently inserted in a globalized, international and connected context, resulting in the emerging of new market needs which leads to new opportunities. A technological startup, based in Portugal – Techwelf – identified an opportunity by noticing that offering someone a present can be enhanced by a pleasant, innovative and user-friendly experience, making it more dynamic, innovative and creative. The act of giving, although millennial, still remains contemporary and important in society. This practice can be understood as a form of self-expression, materialistic exchange and also as a form of building social networks and communities[1]. Along these lines, the market potential will be analyzed thought two methods of investigation. The first being of exploratory nature through direct and unstructured online interviews, with the purpose of formulating hypotheses. The second method is of descriptive nature through field survey using an online questionnaire, in order to identify market segmentation. The sampling technique used in both cases will be non-probabilistic for convenience and also for quotas. The investigation will be conducted in the city of Curitiba, capital of Paraná, Brazil, as it reflects a consumption pattern aligned with the project proposal. Secondary data will also be used given its contribution as a complementary and comparative source that adds to the delimitation of the problem. As the project is still under development, the results are still preliminary and need to be matured before being presented.     Mycobiota and mycotoxins present in finished fish feeds from farms in the Rio de Janeiro State, Brazil

    The aim of the present study was to determine species of the fungal genera Aspergillus, Fusarium, and Penicillium and fumonisin B1 (FB1), aflatoxin B1 (AFB1), and ochratoxin A (OTA) contamination from feed intended for fish farms. A total of 60 samples were sampled from tilapia farms in the Rio de Janeiro State, Brazil. The quantitative enumeration of fungi as colony-forming units per gram of feed (CFU/g) was performed using the surface spread method in different culture media. The results were expressed as fungal isolation frequency and relative density. Fungal total counts ranged from <1 × 102 to 4.7 × 104 CFU/g. Fusarium counts were not observed. Among toxigenic genera, Aspergillus (68%) was the most prevalent, followed by Penicillium species (60%). Aspergillus niger aggregate (36%), Aspergillus flavus (35%), and Penicillium citrinum (71%) were the most prevalent species. A high percentage of samples (98%) were contaminated with FB1 levels, while 55% and 3.3% were contaminated with AFB1 and OTA, respectively. The simultaneous occurrence of these mycotoxins emphasizes the need for further research in the area to better assess the risk to the health of fish farms and their implications for the health of consumers of this meat.Fil: Barbosa, Tatiana S.. In rocky shores, desiccation is triggered by daily tide changes, and experimental evidence suggests that local distribution of algal species across the intertidal rocky zone is related to their capacity to tolerate desiccation. In this context, the permanence of Pyropia columbina in the high intertidal rocky zone is explained by its exceptional physiological tolerance to desiccation. This study explored the metabolic pathways involved in tolerance to desiccation in the Chilean P. columbina, by characterizing its transcriptome under contrasting conditions of hydration. We obtained 1,410 ESTs from two subtracted cDNA libraries in naturally hydrated and desiccated fronds. Results indicate that transcriptome from both libraries contain transcripts from diverse metabolic pathways related to tolerance. Among the transcripts differentially expressed, 15% appears involved in protein synthesis, processing and degradation, 14.4% are related to photosynthesis and chloroplast, 13.1% to respiration and mitochondrial function (NADH dehydrogenase and cytochrome c oxidase proteins), 10.6% to cell wall metabolism, and 7.5% are involved in antioxidant activity, chaperone and defense factors (catalase, thioredoxin, heat shock proteins, cytochrome P450). Both libraries highlight the presence of genes/proteins never described before in algae. This information provides the first molecular work regarding desiccation tolerance in P. columbina, and helps, to some extent, explaining the classical patterns of ecological distribution described for algae across the intertidal zone.http://ref.scielo.org/jm5rc

    Designed to promote students' learning self-regulation, "Testas' (Mis)adventures" is an intervention project consisting of five narrative books and one manual with the theoretical framework and working guidelines for teachers and parents. Testas' narratives introduce an embedded self-regulated learning model, which provides an opportunity to discuss a learning strategies repertoire. The ability of students to relate to Testas' study problems, enables them to understand the leading role of learning strategies in the achievement of academic goals.
