    Antimicrobial and Photoantimicrobial Activities of Chitosan/CNPPV Nanocomposites

    Multidrug-resistant bacteria represent a global health and economic burden that urgently calls for new technologies to combat bacterial antimicrobial resistance. Here, we developed novel nanocomposites (NCPs) based on chitosan that display different degrees of acetylation (DAs), and conjugated polymer cyano-substituted poly(p-phenylene vinylene) (CNPPV) as an alternative approach to inactivate Gram-negative (E. coli) and Gram-positive (S. aureus) bacteria. Chitosan's structure was confirmed through FT-Raman spectroscopy. Bactericidal and photobactericidal activities of NCPs were tested under dark and blue-light irradiation conditions, respectively. Hydrodynamic size and aqueous stability were determined by DLS, zeta potential (ZP) and time-domain NMR. TEM micrographs of NCPs were obtained, and their capacity of generating reactive oxygen species (ROS) under blue illumination was also characterized. Meaningful variations on ZP and relaxation time T2 confirmed successful physical attachment of chitosan/CNPPV. All NCPs exhibited a similar and shrunken spherical shape according to TEM. A lower DA is responsible for driving higher bactericidal performance alongside the synergistic effect from CNPPV, lower nanosized distribution profile and higher positive charged surface. ROS production was proportionally found in NCPs with and without CNPPV by decreasing the DA, leading to a remarkable photobactericidal effect under blue-light irradiation. Overall, our findings indicate that chitosan/CNPPV NCPs may constitute a valuable asset for the development of innovative strategies for inactivation and/or photoinactivation of bacteria.     In Vitro and In Vivo Activity of a Palladacycle Complex on Leishmania (Leishmania) amazonensis

    Leishmaniasis is an important public health problem with an estimated annual incidence of 1.5 million of new human cases of cutaneous leishmaniasis and 500,000 of visceral leishmaniasis. Treatment of the diseases is limited by toxicity and parasite resistance to the drugs currently in use, validating the need to develop new leishmanicidal compounds. We evaluated the killing by the palladacycle complex DPPE 1.2 of Leishmania (Leishmania) amazonensis, an agent of human cutaneous leishmaniasis in the Amazon region, Brazil. DPPE 1.2 destroyed promastigotes of L. (L.) amazonensis in vitro at nanomolar concentrations, whereas intracellular amastigotes were killed at drug concentrations 10-fold less toxic than those displayed to macrophages. L. (L.) amazonensis-infected BALB/c mice treated by intralesional injection of DPPE 1.2 exhibited a significant decrease of foot lesion sizes and a 97% reduction of parasite burdens when compared to untreated controls. Additional experiments indicated the inhibition of the cathepsin B activity of L. (L.) amazonensis amastigotes by DPPE 1.2. Further studies are needed to explore the potential of DPPE 1.2 as an additional option for the chemotherapy of leishmaniasis

    Hymenoptera “parasitica” no estado do Mato Grosso do Sul, Brasil

    A checklist composed of 105 species of parasitic Hymenoptera, which includes the non-aculeate Apocrita, recorded in the state of Mato Grosso do Sul (MS), Brazil, is presented. A new list, containing 153 genera obtained in recent surveys is also presented; out of these 131 are new records.     A new miniature Pristella (actinopterygii: Characiformes: Characidae) with reversed sexual dimorphism from the rio tocantins and rio São Francisco basins, Brazil

    A new species of the genus Pristella Eigenmann, 1908 (Pristella crinogi sp. nov.) is described from the middle rio Tocantins and middle rio São Francisco basins, Brazil. The new species can be diagnosed from its two congeners, Pristella ariporo Conde-Saldaña, Albornoz-Garzón, García-Melo, Villa-Navarro, Mirande, and Lima, 2019 and Pristella maxillaris (Ulrey, 1894), by a combination of color pattern and teeth morphology characters. A phylogenetic analysis of the genus recovered P. crinogi as the sister taxa of P. ariporo. Pristella crinogi, along with P. ariporo, are the first characiform fishes, and one of the first bony fishes, to be reported as presenting a reversed sexual dimorphism, with females presenting a more developed color pattern than males. Comments on theminiaturization of the species, as well as remarks on the biogeography of the genus Pristella, are presented.Une nouvelle espèce de genre Pristella Eigenmann, 1908 (Pristella crinogi sp. nov.) est décrite des bassins versants du rio Tocantins moyen et du rio São Francisco moyen (Brésil). La nouvelle espèce se distingue de ses deux congénères, Pris- tella ariporo Conde-Saldaña, Albornoz-Garzón, García-Melo, Villa-Navarro, Mirande et Lima, 2019 et Pristella maxillaris (Ulrey, 1894), par une combinaison de caractères associés à la coloration et à la morphologie des dents. Une analyse phylogénétique du genre fait ressortir P. crinogi comme taxon-frère de P. ariporo. Pristella crinogi, ainsi que P. ariporo sont les premiers pois- sons characiformes et parmi les premiers poissons osseux pour lesquels un dimorphisme sexuel inversé est signalé, les femelles présentant une coloration plus développée que les mâles.     Evaluation of Eosin-Methylene Blue as a Photosensitizer for Larval Control of Aedes aegypti by a Photodynamic Process

    Aedes aegypti (Ae. aegypti) is a competent vector for transmitting important viral diseases such as yellow fever, dengue, chikungunya, and Zika. Several strategies have been applied to avoid Ae. aegypti proliferation by using environmental management, biological, and chemical approaches. However, the development of new methods for effective control of the insect vector population is still needed. Photodynamic control is an alternative way to control the vector population by using a physical approach based on the larval phototoxicity of a photosensitizer. In this context, the present study evaluated the use of eosin-methylene blue (EMB) as a new photosensitizer for photodynamic control of Ae. aegypti larval populations. The photodynamic assays were performed submitting Ae. aegypti third-instar larvae to different EMB concentrations (0.0, 0.5, 1.0, 5.0, 10.0, 50.0, and 100.0 µg mL−1) in combination of three different light doses (24.3, 48.6, and 97.2 J cm−2) under either white-light radiation from RGB LEDs or sunlight. The results demonstrated that EMB presented a rapid internalization into the larvae and was phototoxic. The photodynamic action induced 100% of larval mortality after about 40 min of sunlight irradiation even using low EMB concentration (0.5 µg mL−1). The findings reveal EMB as an effective photoactive compound to control larval populations of Ae. aegypti by photodynamic process induced by either sunlight or white-light from RGB LEDs
