    Detecting Evasive Malicious URL Using Graph Algorithm

    The present disclosure describes systems and methods to enable inspecting of byte streams using a bipartite match algorithm to detect use of evasive techniques to defeat Uniform Resource (URL) filtering. According to an aspect of the present disclosure, a two stage dynamic programming algorithm is provided that matches signatures once and only once just like DFA in the first stage, and uses polynomial runtime to select a matching pattern in the second stage. According to an example implementation of the present disclosure, the patterns are compiled into an n*m binary matrix plus one additional column to describe the modifiers, where n is the number of patterns and m is the number of unique signatures used in the patterns. In the first stage of runtime, an n*m binary matrix is built where n is the number of tokens matched against the DFA and m is the number of signatures such that e(i,j)=true if and only if the ith token matches the jth signature. In the second stage of runtime, the sub runtime n*m matrix is obtained for a particular pattern such that e(i,j)=true if and only if the ith token matches the jth signatures in the pattern. A bipartite graph can be built where there are two sets of vertex, n and m, where n is token and m is signature. There is a weight 1 edge between an ith vertex in n and an nth vertex in m if e(i,j) in the matrix is true

    Low-cycle Fatigue and Fracture Behaviour of 9%Ni Steel Flux Cored Arc Welding joint at Cryogenic Temperature

    In the present work, low-cycle fatigue (LCF) test and crack tip opening displacement (CTOD) test were performed for 9% Ni steel flux cored arc welding (FCAW) joint at room temperature (296 K) and cryogenic temperature (80 K). At cryogenic temperature, the strain amplitude had a far greater impact on fatigue life of 9%Ni steel welded joint and it decreased dramatically lead to a significant increase in fatigue life. It was found that most fracture initiation of joints located in fusion area at room temperature, while it occurred in weld seam at low temperature. The fracture toughness of weld seam was higher than that of fusion zone no matter the testing temperature. The effect of precipitated phase was the true reason. The fatigue cracks propagated in transgranular mode at room temperature, ultimately, and intergranular mode at low temperature in both LCF specimens and CTOD specimens

    Clinicopathological characteristics and prognostic analysis of Lauren classification in gastric adenocarcinoma in China

    BACKGROUND: According to the Lauren classification, gastric adenocarcinomas are divided into diffuse and intestinal types. The causative attribution explaining the dismal prognosis of diffuse-type remains unknown. METHODS: We examined the archive of 1000 patients with gastric adenocarcinomas who received radical gastrectomy in our center and assessed the effect of the Lauren classification on survival in a multivariate approach. Moreover we compared the variation of clinical features between the diffuse-type and intestinal-type and explored the contributing factors for the prognostic difference. RESULTS: There were 805 resectable patients for the final analysis. Diffuse-type comprised of 48.7% in the gastric carcinoma in our group and showed poorer prognosis than intestinal-type (P=0.013). Multivariate analysis revealed that independent prognostic factors for gastric carcinoma patients were T stage (P<0.001), N stage (P<0.001) tumor size (P<0.001) and Lauren classification (P=0.003). For the clinical features, diffuse-type was significantly associated with younger age (p<0.001), female preponderance (p <0.001), distal location (P<0.001), advanced pT (p < 0.001), advanced pN (p < 0.001) and advanced TNM stage (p = 0.027). CONCLUSIONS: Diffuse type adenocarcinoma carries a worse prognosis that may be partially explained by the tendency of this subtype to present at more advanced T and N stage. However, Lauren classification has prognostic significance that is independent of T and N stage as well as other prognostic variables based on the multivariate cox analysis

    High expression of transcriptional coactivator p300 correlates with aggressive features and poor prognosis of hepatocellular carcinoma

    <p>Abstract</p> <p>Background</p> <p>It has been suggested that p300 participates in the regulation of a wide range of cell biological processes and mutation of p300 has been identified in certain types of human cancers. However, the expression dynamics of p300 in hepatocellular carcinoma (HCC) and its clinical/prognostic significance are unclear.</p> <p>Methods</p> <p>In this study, the methods of reverse transcription-polymerase chain reaction (RT-PCR), Western blotting and immunohistochemistry (IHC) were utilized to investigate protein/mRNA expression of p300 in HCCs. Receiver operating characteristic (ROC) curve analysis, spearman's rank correlation, Kaplan-Meier plots and Cox proportional hazards regression model were used to analyze the data.</p> <p>Results</p> <p>Up-regulated expression of p300 mRNA and protein was observed in the majority of HCCs by RT-PCR and Western blotting, when compared with their adjacent non-malignant liver tissues. According to the ROC curves, the cutoff score for p300 high expression was defined when more than 60% of the tumor cells were positively stained. High expression of p300 was examined in 60/123 (48.8%) of HCCs and in 8/123 (6.5%) of adjacent non-malignant liver tissues. High expression of p300 was correlated with higher AFP level, larger tumor size, multiplicity, poorer differentiation and later stage (<it>P </it>< 0.05). In univariate survival analysis, a significant association between overexpression of p300 and shortened patients' survival was found (<it>P </it>= 0.001). In different subsets of HCC patients, p300 expression was also a prognostic indicator in patients with stage II (<it>P </it>= 0.007) and stage III (<it>P </it>= 0.011). Importantly, p300 expression was evaluated as an independent prognostic factor in multivariate analysis (<it>P </it>= 0.021). Consequently, a new clinicopathologic prognostic model with three poor prognostic factors (p300 expression, AFP level and vascular invasion) was constructed. The model could significantly stratify risk (low, intermediate and high) for overall survival (<it>P </it>< 0.0001).</p> <p>Conclusions</p> <p>Our findings provide a basis for the concept that high expression of p300 in HCC may be important in the acquisition of an aggressive phenotype, suggesting that p300 overexpression, as examined by IHC, is an independent biomarker for poor prognosis of patients with HCC. The combined clinicopathologic prognostic model may become a useful tool for identifying HCC patients with different clinical outcomes.</p

    Arbuscular mycorrhizal fungi improve selenium uptake by modulating root transcriptome of rice (Oryza sativa L.)

    Although selenium (Se) is an essential trace element in humans, the intake of Se from food is still generally inadequate throughout the world. Inoculation with arbuscular mycorrhizal fungi (AMF) improves the uptake of Se in rice (Oryza sativa L.). However, the mechanism by which AMF improves the uptake of Se in rice at the transcriptome level is unknown. Only a few studies have evaluated the effects of uptake of other elements in rice under the combined effects of Se and AMF. In this study, Se combined with the AMF Funneliformis mosseae (Fm) increased the biomass and Se concentration of rice plants, altered the pattern of ionomics of the rice roots and shoots, and reduced the antagonistic uptake of Se with nickel, molybdenum, phosphorus, and copper compared with the treatment of Se alone, indicating that Fm can enhance the effect of fertilizers rich in Se. Furthermore, a weighted gene co-expression network analysis (WGCNA) showed that the hub genes in modules significantly associated with the genes that contained Se and were related to protein phosphorylation, protein serine/threonine kinase activity, membrane translocation, and metal ion binding, suggesting that the uptake of Se by the rice roots may be associated with these genes when Fm and Se act in concert. This study provides a reference for the further exploration of genes related to Se uptake in rice under Fm treatment

    Ophiopogonin D Attenuates Doxorubicin-Induced Autophagic Cell Death by Relieving Mitochondrial Damage In Vitro and In Vivo

    ABSTRACT It has been reported that ophiopogonin D (OP-D), a steroidal glycoside and an active component extracted from Ophiopogon japonicas, promotes antioxidative protection of the cardiovascular system. However, it is unknown whether OP-D exerts protective effects against doxorubicin (DOX)-induced autophagic cardiomyocyte injury. Here, we demonstrate that DOX induced excessive autophagy through the generation of reactive oxygen species (ROS) in H9c2 cells and in mouse hearts, which was indicated by a significant increase in the number of autophagic vacuoles, LC3-II/LC3-I ratio, and upregulation of the expression of GFP-LC3. Pretreatment with OP-D partially attenuated the above phenomena, similar to the effects of treatment with 3-methyladenine. In addition, OP-D treatment significantly relieved the disruption of the mitochondrial membrane potential by antioxidative effects through downregulating the expression of both phosphorylated c-Jun N-terminal kinase and extracellular signal-regulated kinase. The ability of OP-D to reduce the generation of ROS due to mitochondrial damage and, consequently, to inhibit autophagic activity partially accounts for its protective effects in the hearts against DOX-induced toxicity
