    Biology of human hair: Know your hair to control it

    Hair can be engineered at different levels—its structure and surface—through modification of its constituent molecules, in particular proteins, but also the hair follicle (HF) can be genetically altered, in particular with the advent of siRNA-based applications. General aspects of hair biology are reviewed, as well as the most recent contributions to understanding hair pigmentation and the regulation of hair development. Focus will also be placed on the techniques developed specifically for delivering compounds of varying chemical nature to the HF, indicating methods for genetic/biochemical modulation of HF components for the treatment of hair diseases. Finally, hair fiber structure and chemical characteristics will be discussed as targets for keratin surface functionalization

    Conservation of Olfactory Avoidance in Drosophila Species and Identification of Repellents for Drosophila suzukii

    Flying insects use olfaction to navigate towards fruits in complex odor environments with remarkable accuracy. Some fruits change odor profiles substantially during ripening and related species can prefer different stages. In Drosophila species attractive odorants have been studied extensively, but little is understood about the role of avoidance pathways. In order to examine the role of the avoidance cue CO(2) emitted from fruit on behavior of two species with different ripening stage preferences, we investigated the CO(2)-detection pathway in Drosophila melanogaster and Drosophila suzukii, a harmful pest of fruits. Avoidance to CO(2) is not conserved in D. suzukii suggesting a behavioral adaptation that could facilitate attraction to younger fruit with higher CO(2) emission levels. We investigated known innate avoidance pathways from five species at different evolutionary distances: D. melanogaster, D. yakuba, D. suzukii, D. pseudoobscura and D. virilis. Surprisingly, only DEET shows strong repellency across all species, whereas CO(2), citronellal and ethyl 3-hydroxybutyrate show only limited conservation. These findings guide us to test recently discovered safe DEET substitutes, and we identify one that protects fruits from D. suzukii thus providing a new behavioral strategy for controlling agricultural pests

    Carbon dioxide is produced by many organic processes, and is a convenient volatile cue for insects searching for blood hosts, flowers, communal nests, fruit, and wildfires. Curiously, although Drosophila melanogaster feed on yeast that produce CO2 and ethanol during fermentation, laboratory experiments suggest that walking flies avoid CO2. Here, we resolve this paradox by showing that both flying and walking Drosophila find CO2 attractive, but only when in an active state associated with foraging. Aversion at low activity levels may be an adaptation to avoid CO2-seeking-parasites, or succumbing to respiratory acidosis in the presence of high concentrations of CO2 that exist in nature. In contrast to CO2, flies are attracted to ethanol in all behavioral states, and invest twice the time searching near ethanol compared to CO2. These behavioral differences reflect the fact that whereas CO2 is generated by many natural processes, ethanol is a unique signature of yeast fermentation. Using genetic tools, we determined that the evolutionarily ancient ionotropic co-receptor IR25a is required for CO2 attraction, and that the receptors necessary for CO2 avoidance are not involved.     Carbon Dioxide and Fruit Odor Transduction in Drosophila Olfactory Neurons. What Controls their Dynamic Properties?

    We measured frequency response functions between odorants and action potentials in two types of neurons in Drosophila antennal basiconic sensilla. CO(2) was used to stimulate ab1C neurons, and the fruit odor ethyl butyrate was used to stimulate ab3A neurons. We also measured frequency response functions for light-induced action potential responses from transgenic flies expressing H134R-channelrhodopsin-2 (ChR2) in the ab1C and ab3A neurons. Frequency response functions for all stimulation methods were well-fitted by a band-pass filter function with two time constants that determined the lower and upper frequency limits of the response. Low frequency time constants were the same in each type of neuron, independent of stimulus method, but varied between neuron types. High frequency time constants were significantly slower with ethyl butyrate stimulation than light or CO(2) stimulation. In spite of these quantitative differences, there were strong similarities in the form and frequency ranges of all responses. Since light-activated ChR2 depolarizes neurons directly, rather than through a chemoreceptor mechanism, these data suggest that low frequency dynamic properties of Drosophila olfactory sensilla are dominated by neuron-specific ionic processes during action potential production. In contrast, high frequency dynamics are limited by processes associated with earlier steps in odor transduction, and CO(2) is detected more rapidly than fruit odor
