    A preliminary analysis of spatiotemporal patterns in swordfish habitat distributions.

    A species distribution model (SDM) for swordfish that was in the development stage has been finalized. The model used detailed biological and oceanographic data to define the spatial distribution of Swordfish. The SDM adequately predicted Swordfish habitat (and thus fish) distributions such that it was found suitable for investigations into the spatiotemporal distribution of habitat. Results of this preliminary investigation supports the current hypothesized stock boundaries between the north and south Atlantic stocks used for management. Both the north and south Atlantic may be experiencing an expansion of habitat. This could result in decreased density of swordfish into a larger area and/or change MSY production metrics.     A Step Forward in Breast Cancer Research: Gold Nanoparticles as Photothermal Therapy Enhancers

    Gold nanoparticles (AuNPs) have been widely used and characterized for multiple biomedical applications, including the enhancement of photothermal therapy (PTT). AuNPs present a particular plasmon resonance band and are able to convert the absorbed optical radiation into heat, which validates their use in PTT. Several production methods have already been proposed for the synthesis of AuNPs, allowing to optimize the particles' morphology, size and optical properties. However, the production methods commonly used are frequently associated with the use of toxic reagents such as Cetyltrimethylammonium bromide, which presents some concerns for clinical applications. Herein, it is proposed a novel AuNPs' core synthesis method using tetrachloroauric acid and a mixture of reducing agents, later on coated with a combination of hyaluronic and oleic acids. The coating here represents a potential improvement of AuNPs biocompatibility, biodegradability and lifetime, while simultaneously potentiating the attachment towards specific ligands, such as the CD44 receptor, to develop more localized and highly selective tools. The produced functionalized nanoparticles were characterized by Dynamic Light Scattering, Microscopy Techniques and Spectroscopy, showing diameter sizes under 350 nm, polydispersity index smaller than 0.4 and enhanced absorbance in the Near Infrared (NIR, 650 to 900 nm) range. Moreover, the AuNPs safety and efficacy were preliminarily assessed in vitro using breast cancer cell lines. No toxicity was observed by MTT assay, both in breast cancer cell lines, and red blood cells. The irradiation process was proved to be safe; however, when combined with the AuNPs administration, it resulted in a significant reduction of cell viability for some of the breast cell lines tested. Thus, the results highlight the potential of the proposed system for some type of tumors, even though further tests are required to better understand the mechanisms behind the obtained results

    The Role of Rosmarinic Acid on the Bioproduction of Gold Nanoparticles as Part of a Photothermal Approach for Breast Cancer Treatment.

    Breast cancer is a high-burden malignancy for society, whose impact boosts a continuous search for novel diagnostic and therapeutic tools. Among the recent therapeutic approaches, photothermal therapy (PTT), which causes tumor cell death by hyperthermia after being irradiated with a light source, represents a high-potential strategy. Furthermore, the effectiveness of PTT can be improved by combining near infrared (NIR) irradiation with gold nanoparticles (AuNPs) as photothermal enhancers. Herein, an alternative synthetic method using rosmarinic acid (RA) for synthesizing AuNPs is reported. The RA concentration was varied and its impact on the AuNPs physicochemical and optical features was assessed. Results showed that RA concentration plays an active role on AuNPs features, allowing the optimization of mean size and maximum absorbance peak. Moreover, the synthetic method explored here allowed us to obtain negatively charged AuNPs with sizes favoring the local particle accumulation at tumor site and maximum absorbance peaks within the NIR region. In addition, AuNPs were safe both in vitro and in vivo. In conclusion, the synthesized AuNPs present favorable properties to be applied as part of a PTT system combining AuNPs with a NIR laser for the treatment of breast cancer

    Does ICCAT need ecosystem plans? a pilot ecosystem plan for the Atlantic tropical ecoregion.

    The implementation of an Ecosystem Approach Fisheries Management (EAFM) in ICCAT has been slow and patchy, as it lacks a long-term plan, vision and guidance on how to operationalize it. Ecosystem plans are needed to formalize the process of operationalizing the EAFM by identifying and formalizing ecosystem goals and objectives, planning actions based on priorities, measuring performance of the whole fishery system, addressing trade-offs, and incorporating them in fisheries management. The Specific Contract N0 2 under the Framework Contract - EASME/EMFF/2016/008 provisions of Scientific Advice for Fisheries Beyond EU Waters has developed a pilot ecosystem plan for the tropical ecoregion of the Atlantic Ocean. In this document, we highlight the main potential benefits of developing ecosystem plans in ICCAT. Second, we briefly describe the main core elements developed in the pilot ecosystem plan for the Tropical ecoregion of the Atlantic Ocean. Third, we summarize our main thoughts and lessons learned in the development of this pilot ecosystem plan for one ecoregion within ICCAT. Last, we propose a list of actions, research activities and capacity building activities to foster the development, use and implementation of ecosystem plans in ICCAT.Versión del edito

    Gold nanoparticles as a part of a photothermal therapy system.

    Introduction Photothermal therapy (PTT) is attracting increased attention for the treatment of superficial localized tumors, relying on the induction of local hyperthermia of tumor cells upon their irradiation with light beams1. PTT efficacy depends, however, on the heat generated and, on the depth reached by the light. Some strategies to improve PTT efficacy includes the use of the near infrared (NIR, 650 to 900 nm) radiation to enhance the penetration depth of the light, combined with gold nanoparticles (AuNPs) to enhance the photothermal effect2. Experimental Methods Core AuNPs were synthesized by a novel method using tetrachloroauric acid and a mixture of reducing agents, and subsequently coated with a combination of hyaluronic and oleic acids, for improving the NPs biocompatibility, biodegradability, and lifetime. This coating also promotes the binding of specific cell receptors of the tumor cells. The particles were physico-chemically characterized, and in vitro and in vivo tests were carried out in breast cancer models to assess their safety and efficacy, when applied alone or combined with NIR irradiation3. Results and Discussion AuNPs presented a predominant spherical morphology with sizes under 350 nm, polydispersity index lower than 0.4 and enhanced absorbance in the NIR. The particles showed no toxicity in vitro and promising efficacy in vivo when administering the NPs in situ and later irradiating them externally. Histopathological analysis of tumors treated with both AuNPs and laser irradiation showed the presence of necrosis in most of the tumors and no effect or practically absence in healthy surrounding cells, which are very encouraging outcomes. Conclusion The results are promising, however, there is still room for improving the system, namely by reducing even more the invasiveness of the treatment through the combined use of aerogels structures. Aerogel’s unique properties4 make them ideal candidates to minimize the exposure of healthy tissues to laser radiation, acting as light and thermal insulators, as well as to incorporate the nanoparticles into their skeletal structure and thus potentiating a topical application of the particles. For these reasons, some exploratory methods were carried to produce and design aerogels structures for PTT applications

    On tuning passive black-box macromodels of LTI systems via adaptive weighting

    This paper discusses various approaches for tuning the accuracy of rational macromodels obtained via black-box identification or approximation of sampled frequency responses of some unknown Linear and Time-Invariant system. Main emphasis is on embedding into the model extraction process some information on the nominal terminations that will be connected to the model during normal operation, so that the corresponding accuracy is optimized. This goal is achieved through an optimization based on a suitably defined cost function, which embeds frequency-dependent weights that are adaptively refined during the model construction. A similar procedure is applied in a postprocessing step for enforcing model passivity. The advantages of proposed algorithm are illustrated on a few application examples related to power distribution networks in electronic system

    Selecting ecosystem indicators for fisheries targeting highly migratory species: An EU project to advance the operationalization of the EAFM in ICCAT and IOTC

    Several international legal agreements and guidelines have set the minimum standards and key principles to guide the implementation of an ecosystem approach to fisheries management (EAFM). However, the implementation of an EAFM in tuna Regional Fisheries Management Organizations (RFMOs) has been patchy and lack a long-term plan, vision and guidance on how to operationalize it. The Specific Contract N0 2 “selecting ecosystem indicators for fisheries targeting highly migratory species-” (SC02 project) under the Framework Contract - EASME/ EMFF/2016/008 provisions of Scientific Advice for Fisheries Beyond EU Waters- addresses several scientific challenges and provides insights to support the implementation of an EAFM through collaboration and consultation with the International Commission for the Conservation of Atlantic Tunas (ICCAT) and the Indian Ocean Tuna Commission (IOTC). Specifically, this project first highlights properties of success and best practices from other regions of the world in operationalizing the ecosystem approach that potentially could be transferred to ICCAT and IOTC. Second, it delivered a list of potential ecosystem indicators of relevance to tuna RFMOs (ICCAT and IOTC) that are suitable to track the impacts of fisheries targeting tuna and tuna-like species on the broader pelagic ecosystem. Third, it designed a general framework based on a rule-based decision tree to provide guidance on how reference points could be set and used for diverse types of ecosystem indicators. Fourth, it proposed candidate ecoregions within the Atlantic and Indian Oceans which could be used to guide region-based ecosystem plans, assessments and research to ultimately provide better ecosystem-based advice to inform fisheries management. Fifth, it developed two pilot ecosystem plans for two case study regions, the tropical ecoregion within the ICCAT convention area, and the temperate ecoregion within the IOTC convention area. At this stage, these pilot ecosystem plans aim to create awareness about the need for ecosystem planning, start a discussion about the elements that need to be part of a planning process, and initiate a discussion in ICCAT and IOTC about the potential needs of ecosystem plans and their function. Finally, this project provided recommendations to foster the potential development, use, and implementation of ecosystem plans in ICCAT and IOTC

    Functional and evolutionary significance of unknown genes from uncultivated taxa

    Most microbes on our planet remain uncultured and poorly studied. Recent efforts to catalog their genetic diversity have revealed that a significant fraction of the observed microbial genes are functional and evolutionary untraceable, lacking homologs in reference databases. Despite their potential biological value, these apparently unrelated orphan genes from uncultivated taxa have been routinely discarded in metagenomics surveys. Here, we analyzed a global multi-habitat dataset covering 151,697 medium and high-quality metagenome assembled genomes (MAGs), 5,969 single-amplified genomes (SAGs), and 19,642 reference genomes, and identified 413,335 highly curated novel protein families under strong purifying selection out of previously considered orphan genes. These new protein families, representing a three-fold increase over the total number of prokaryotic orthologous groups described to date, spread out across the prokaryote phylogeny, can span multiple habitats, and are notably overrepresented in recently discovered taxa. By genomic context analysis, we pinpointed thousands of unknown protein families to phylogenetically conserved operons linked to energy production, xenobiotic metabolism and microbial resistance. Most remarkably, we found 980 previously neglected protein families that can accurately distinguish entire uncultivated phyla, classes, and orders, likely representing synapomorphic traits that fostered their divergence. The systematic curation and evolutionary analysis of the unique genetic repertoire of uncultivated taxa opens new avenues for understanding the biology and ecological roles of poorly explored lineages at a global scale

    Is upper gastrointestinal radiography a cost-effective alternative to a Helicobacter pylori “Test and Treat” strategy for patients with suspected peptic ulcer disease?

    Current clinical consensus supports an initial Helicobacter pylori (HP) “test and treat” approach when compared to immediate endoscopy for patients with suspected peptic ulcer disease. Alternative diagnostic approaches that incorporate upper GI radiography (UGI) have not been previously evaluated. We sought to determine the cost effectiveness of UGI compared to a HP test and treat strategy, incorporating recent data addressing the reduced prevalence of HP, lower cost of diagnostic interventions, and reduced attribution of PUD to HP. METHODS : Using decision analysis, three diagnostic and treatment strategies were evaluated: 1) Test and Treat —initial HP serology, treat patients who test positive with HP eradication and antiulcer therapy; 2) Initial UGI series —treat all patients with documented ulcer disease with HP eradication and antiulcer therapy; and 3) Initial UGI series, HP serology if ulcer present — treat ulcer and HP based on diagnostic test results. RESULTS : The estimated cost per ulcer cured for each strategy were as follows: test and treat, 3,025;initialUGI,3,025; initial UGI, 3,690; and UGI with serology, 3,790.Theestimatedcostperpatienttreatmentwere:testandtreat,3,790. The estimated cost per patient treatment were: test and treat, 498; initial UGI, 610;andUGIwithserology,610; and UGI with serology, 620. When UGI reimbursement was decreased to less than $50, the UGI strategies yielded a lower cost per patient treated than the test and treat strategy.
