    In Vitro and In Vivo Activity of a Palladacycle Complex on Leishmania (Leishmania) amazonensis

    Leishmaniasis is an important public health problem with an estimated annual incidence of 1.5 million of new human cases of cutaneous leishmaniasis and 500,000 of visceral leishmaniasis. Treatment of the diseases is limited by toxicity and parasite resistance to the drugs currently in use, validating the need to develop new leishmanicidal compounds. We evaluated the killing by the palladacycle complex DPPE 1.2 of Leishmania (Leishmania) amazonensis, an agent of human cutaneous leishmaniasis in the Amazon region, Brazil. DPPE 1.2 destroyed promastigotes of L. (L.) amazonensis in vitro at nanomolar concentrations, whereas intracellular amastigotes were killed at drug concentrations 10-fold less toxic than those displayed to macrophages. L. (L.) amazonensis-infected BALB/c mice treated by intralesional injection of DPPE 1.2 exhibited a significant decrease of foot lesion sizes and a 97% reduction of parasite burdens when compared to untreated controls. Additional experiments indicated the inhibition of the cathepsin B activity of L. (L.) amazonensis amastigotes by DPPE 1.2. Further studies are needed to explore the potential of DPPE 1.2 as an additional option for the chemotherapy of leishmaniasis

    The three organometallic complexes [(Cis-PtII (DDH) (2,5-Dihidroxibenzensulfonic)2, RhI (CO)2 Cl(2-Aminobenzothiazole) and RhI (CO)2 Cl(5-Cl-2-Methilbenzothiazole)] used in this study had been previously found to have a high in vitro activity against promastigote and amastigote like forms of Leishmania donovani. Here, the cytotoxic effect of these new organometallic complexes on the J-774 macrophages were studied. Only the RhI(CO)2 Cl (2-Aminobenzothiazole) complex induced substantial toxicity in the cells. Also, we assayed the effect of this complex on the parasite's biosynthesis of macromolecules. The RhI(CO)2Cl (5-Cl-2-Methylbenzothiazole) complex inhibited DNA, RNA, and protein synthesis. On the other hand, the two other compounds tested did not inhibit the incorporation of radioactive precursors. Finally important ultrastructural alterations in the parasites treated with the two non-cytotoxic complexes were observed

    Manipulation of Agricultural Habitats to Improve Conservation Biological Control in South America

    International audienceStable and diversified agroecosystems provide farmers with important ecosystem services, which are unfortunately being lost at an alarming rate under the current conventional agriculture framework. Nevertheless, this concern can be tackled by using ecological intensification as an alternative strategy to recuperate ecosystem services (e.g., biological control of pests). To this end, the manipulation of agricultural habitats to enhance natural enemy conservation has been widely explored and reported in Western Europe and North America, whereas in other parts of the world, the investigation of such topic is lagging behind (e.g., South America). In this forum, we gathered published and unpublished information on the different ecological habitat management strategies that have been implemented in South America and their effects on pest control. Additionally, we identify the various challenges and analyze the outlook for the science of conservation biological control in South America. More specifically, we reviewed how different agricultural practices and habitat manipulation in South America have influenced pest management through natural enemy conservation. The main habitat manipulations reported include plant diversification (intercropping, insectary plants, agroforestry), conservation and management of non-crop vegetation, and application of artificial foods. Overall, we noticed that there is a significant discrepancy in the amount of research on conservation biological control among South American countries, and we found that, although intercropping, polycultures, and crop rotation have been reported in agroecosystems since pre-Inca times, more systematic studies are required to evaluate the true effects of habitat management to implement conservation biological control for pest control in South America
