    Efeito de um programa de fisioterapia funcional em crianças com paralisia cerebral associado a orientações aos cuidadores: estudo preliminar

    O objetivo foi verificar o efeito de um programa de fisioterapia funcional para crianças com paralisia cerebral, associado a orientações aos pais e/ou cuidadores; e verificar a correlação entre as habilidades funcionais e a assistência do cuidador, utilizando o Inventário de Avaliação Pediátrica de Incapacidade (PEDI). Participaram quatro crianças entre 24 e 43 meses de idade, hemiplégicas, espásticas e nível I no sistema de classificação da função motora ampla (GMFCS). Foram realizadas quatro avaliações - uma antes do início do programa, as demais aos 30, 60 e 90 dias após a primeira -, empregando-se as partes I (Habilidades funcionais) e II (Assistência do cuidador) do PEDI. As crianças foram submetidas a sessões de uma hora de fisioterapia funcional três vezes por semana, durante três meses: duas vezes a sessão era de fisioterapia com base no conceito neuroevolutivo Bobath e uma vez, treino de atividades da vida diária. Também foram dadas orientações por escrito aos pais e/ou cuidadores quanto à assistência à criança, incentivando-os a praticá-la em casa. A análise dos resultados mostrou que, na última avaliação, as crianças obtiveram escores significativamente maiores que na primeira. Foi verificada correlação altamente significativa (r=1,0; p=0,083) entre as partes I e II. O programa de fisioterapia funcional associado às orientações aos pais e/ou cuidadores foi efetivo em melhorar o desempenho funcional de crianças nível I com hemiplegia espástica.The purpose was to verify the effect of a functional physical therapy program on children with cerebral palsy, associated to guidance to parents and/or caregivers; and to search for correlations between the child's functional abilities and caregivers' assistance, by means of the Pediatric Evaluation Disability Inventory (PEDI). Four hemiplegic, spastic children between 24 and 43 months old, classified at the Gross Motor Function Classification System level I, were submitted to four evaluations, the first prior to program onset, and the others 30, 60, and 90 days after the first one. PEDI parts I (Functional abilities) and II (Caregivers' assistance) were used. Caregivers were provided with written instructions on how to best deliver care at home. The physical therapy program consisted of three weekly 1-hour sessions for three months; two sessions were of physical therapy based on Bobath concept, and one, of daily activities training. The analysis of results showed children obtained a significantly higher score at the last assessment as compared to the first. A high, significant correlation was found between PEDI parts I and II (r=1.0; p=0.083). The functional physical therapy program associated to instructions to caregivers proved thus effective to improve the functional performance of level-I children with spastic hemiplegia

    Analysis of an N-terminal deletion in subunit a of the Escherichia coli ATP synthase

    Subunit a is a membrane-bound stator subunit of the ATP synthase and is essential for proton translocation. The N-terminus of subunit a in E. coli is localized to the periplasm, and contains a sequence motif that is conserved among some bacteria. Previous work has identified mutations in this region that impair enzyme activity. Here, an internal deletion was constructed in subunit a in which residues 6-20 were replaced by a single lysine residue, and this mutant was unable to grow on succinate minimal medium. Membrane vesicles prepared from this mutant lacked ATP synthesis and ATP-driven proton translocation, even though immunoblots showed a significant level of subunit a. Similar results were obtained after purification and reconstitution of the mutant ATP synthase into liposomes. The location of subunit a with respect to its neighboring subunits b and c was probed by introducing cysteine substitutions that were known to promote cross-linking: a_L207C + c_I55C, a_L121C + b_N4C, and a_T107C + b_V18C. The last pair was unable to form cross-links in the background of the deletion mutant. The results indicate that loss of the N-terminal region of subunit a does not generally disrupt its structure, but does alter interactions with subunit b

    Atomic model for the membrane-embedded VO motor of a eukaryotic V-ATPase

    Vacuolar-type ATPases (V-ATPases) are ATP-powered proton pumps involved in processes such as endocytosis, lysosomal degradation, secondary transport, TOR signalling, and osteoclast and kidney function. ATP hydrolysis in the soluble catalytic V1 region drives proton translocation through the membrane-embedded VO region via rotation of a rotor subcomplex. Variability in the structure of the intact enzyme has prevented construction of an atomic model for the membrane-embedded motor of any rotary ATPase. We induced dissociation and auto-inhibition of the V1 and VO regions of the V-ATPase by starving the yeast Saccharomyces cerevisiae, allowing us to obtain a ~3.9-Å resolution electron cryomicroscopy map of the VO complex and build atomic models for the majority of its subunits. The analysis reveals the structures of subunits ac8c'c"de and a protein that we identify and propose to be a new subunit (subunit f). A large cavity between subunit a and the c-ring creates a cytoplasmic half-channel for protons. The c-ring has an asymmetric distribution of proton-carrying Glu residues, with the Glu residue of subunit c" interacting with Arg735 of subunit a. The structure suggests sequential protonation and deprotonation of the c-ring, with ATP-hydrolysis-driven rotation causing protonation of a Glu residue at the cytoplasmic half-channel and subsequent deprotonation of a Glu residue at a luminal half-channel

    Control of wood thermal treatment and its effects on decay resistance: a review

    Key message. An efficient use of thermal treatment of wood requires a depth understanding of the chemical modifications induced. This is a prerequisite to avoid problems of process control, and to provide high quality treated wood with accurately assessed properties to the market. Properties and structural anatomy of thermally modified woods are slightly different than un-processed woods from a same wood species. So it is necessary to create or adapt new analytical methods to control their quality. Context. Heat treatment as a wood modification process is based on chemical degradation of wood polymer by heat transfer. It improves mainly the resistance of wood to decay and provides dimensional stability. These improvements, which come at the expense of a weakening of mechanical properties, have been extensively studied. Since a decade, researches focused mainly on the understanding of wood thermal degradation, on modelling, on quality prediction and quality control. Aims. We aimed at reviewing the recent advances about (i) the analytical methods used to control thermal treatment; (ii) the effects on wood decay resistance and (iii) the advantages and drawbacks of a potential industrial use of wood heating. Methods. We carried out a literature review of the main industrial methods used to evaluate the conferred wood properties, by thermal treatment. We used papers and reports published between 1970 and 2015, identified in the web of science data base.. Result.s Approximately 100 papers mostly published after 2000 were retrieved. They concentrated on: (i) wood mass loss due to thermal degradation determination, (ii) spectroscopic analyses of wood properties, (iii) colour measurements, (iv) chemical composition, (v) non-destructive mechanical assessments and (vi) use of industrial data. Conclusions. One of most interesting property of heat-treated wood remains its decay resistance. Durability test with modified wood in laboratory are expensive and time-consuming. This review displays data from different analytical methods, such as spectroscopy, thermogravimetry, chemical analyses or mechanical tests that have the potential to be valuable indicators to assess the durability of heat treated wood at industrial scale. However, each method has its limits and drawbacks, such as the required investment for the equipment, reliability and accuracy of the results and ease of use at industrial scale. (Résumé d'auteur
