    Resistance training and changes to plasma lipoproteins in postmenopausal women

    Objectives. The main purpose of this study was to assess the effect of progressive resistance training on the blood lipid profile in postmenopausal women. Methods. Twenty-six female participants aged 50 - 75 years were selected from the population of Grahamstown, South Africa. All participants were previously sedentary and possessed at least one lipid profile abnormality but were otherwise healthy. Pretests included a sub-maximal stress test, stature, mass, central and limb girths as well as an oral glucose tolerance test (OGTT) and a total blood lipid profile. Participants took part in a 24-week progressive resistance training programme, consisting of three supervised sessions per week, each lasting 45 minutes. Participants were not permitted to lose more than 10% of initial body mass during the 24-week study. All pre-test measures, excluding the stress test and the OGTT, were repeated every 4 weeks for the duration of the study. Results. Body mass, body mass index and waist-to-hip ratio did not change. Girth measures at mid-humerus, chest, waist, hip, mid-quadricep and mid-gastrocnemius all decreased significantly (p<0.05). LDL-cholesterol increased significantly over the course of 24 weeks (3.61 mmol.l-1 to 4.07 mmol.l-1), as did total cholesterol (5.81 mmol.l-1- 6.24 mmol.l-1). Triglyceride concentration remained unchanged and HDL-cholesterol decreased significantly between the pre-test measure (1.55 mmol.l-1) and the measure after 6 months (1.42 mmol.l-1). Conclusion. The blood lipid profile in a sample of postmenopausal women was not positively affected by a progressive resistance training programme over a 24-week period

    Investigating the Impact of Cerium Oxide Nanoparticles Upon the Ecologically Significant Marine Cyanobacterium Prochlorococcus

    Cerium oxide nanoparticles (nCeO_{2}) are used at an ever-increasing rate, however, their impact within the aquatic environment remains uncertain. Here, we expose the ecologically significant marine cyanobacterium Prochlorococcus sp. MED4 to nCeO_{2} at a wide range of concentrations (1 ÎŒg L^{–1} to 100 mg L^{–1}) under simulated natural and nutrient rich growth conditions. Flow cytometric analysis of cyanobacterial populations displays the potential of nCeO_{2} (100 ÎŒg L^{–1}) to significantly reduce Prochlorococcus cell density in the short-term (72 h) by up to 68.8% under environmentally relevant conditions. However, following longer exposure (240 h) cyanobacterial populations are observed to recover under simulated natural conditions. In contrast, cell-dense cultures grown under optimal conditions appear more sensitive to exposure during extended incubation, likely as a result of increased rate of encounter between cyanobacteria and nanoparticles at high cell densities. Exposure to supra-environmental nCeO_{2} concentrations (i.e., 100 mg L^{–1}) resulted in significant declines in cell density up to 95.7 and 82.7% in natural oligotrophic seawater and nutrient enriched media, respectively. Observed cell decline is associated with extensive aggregation behaviour of nCeO_{2} upon entry into natural seawater, as observed by dynamic light scattering (DLS), and hetero-aggregation with cyanobacteria, confirmed by fluorescent microscopy. Hence, the reduction of planktonic cells is believed to result from physical removal due to co-aggregation and co-sedimentation with nCeO_{2} rather than by a toxicological and cell death effect. The observed recovery of the cyanobacterial population under simulated natural conditions, and likely reduction in nCeO_{2} bioavailability as nanoparticles aggregate and undergo sedimentation in saline media, means that the likely environmental risk of nCeO_{2} in the marine environment appears low

    Impact of training status on maximal oxygen uptake criteria attainment during running

    Objectives. The aims of this study were to assess whether training status influenced maximal physiological and perceptual responses and whether certain maximal criteria were more sensitive for individuals with different levels of training. Methods. Males who were either trained (N=8) or untrained (N=9) underwent a maximal treadmill test to assess whether thecriteria to indicate VO2 max were training-specific.Results. VO2 max was significantly higher in the trained (70.0mlO2.kg-1.min-1) compared with the untrained group (54.5 mlO2.kg-1.min-1). Only 11% of the trained and 56% of the untrained individuals achieved a plateau in the oxygen uptake curve. Peak treadmill running speed was significantly faster and total test time significantly longer in the trained group. In contrast, peak lactate, although maximal for both groups, was significantly higher in the untrained group (13.5 mmol.l-1 compared with 10.3 mmol.l-1). The other responses were not different between the groups. Noteworthy is that none of the subjects achieved all of the criteria indicating a maximal effort. The criteria most achieved in both sampleswere HRmax, RPEmax and Lamax with the latter being the mostattained in the untrained group and RPEmax being mostly attainedby the trained group.Conclusions. The criteria used to indicate attainment of VO2 max may be limited and may differ when comparing a heterogeneous training sample. Although VO2 max was significantly higher in the trained group, responses were different for O2 plateau attainment and Lamax but similar for the other British Association of Sports Sciences criteria. It may be concluded that the physiological variables coinciding with maximal effort may differ in individuals with different levels of training

    Effects of continuing professional development on group work practices in Scottish primary schools

    This study investigated the effects of a continuing professional development (CPD) initiative that provided collaborative group work skills training for primary school teachers. The study collected data from 24 primary school classrooms in different schools in a variety of urban and rural settings. The sample was composed of 332 pupils aged 9-12 years old, and 24 primary school teachers. Results indicated the CPD initiative had a significant impact on the attainment of pupils in science. In addition data indicated that the CPD promoted effective discourse and pupil dialogue during science lessons. Pre-post test observation scores were significantly different in terms of children giving of suggestions or courses of actions, offering of explanations, and telling someone to say something or carry out an action . Increases in effective dialogue were significantly correlated to increased science attainment and teacher evaluations of the impact of the CPD were positive. Significant correlations were found between teacher evaluation of impact upon pupil learning and increased attainment in science. The design and structure of CPD initiatives and the implications for practice, policy and future research are explored

    Environmentally relevant concentrations of titanium dioxide nanoparticles pose negligible risk to marine microbes

    Nano-sized titanium dioxide (nTiO2) represents the highest produced nanomaterial by mass worldwide and, due to its prevalent industrial and commercial use, it inevitably reaches the natural environment. Previous work has revealed a negative impact of nTiO2 upon marine phytoplankton growth, however, studies are typically carried out at concentrations far exceeding those measured and predicted to occur in the environment currently. Here, a series of experiments were carried out to assess the effects of both research-grade nTiO2 and nTiO2 extracted from consumer products upon the marine dominant cyanobacterium, Prochlorococcus, and natural marine communities at environmentally relevant and supra-environmental concentrations (i.e., 1 ÎŒg L−1 to 100 mg L−1). Cell declines observed in Prochlorococcus cultures were associated with the extensive aggregation behaviour of nTiO2 in saline media and the subsequent entrapment of microbial cells. Hence, higher concentrations of nTiO2 particles exerted a stronger decline of cyanobacterial populations. However, within natural oligotrophic seawater, cultures were able to recover over time as the nanoparticles aggregated out of solution after 72 h. Subsequent shotgun proteomic analysis of Prochlorococcus cultures exposed to environmentally relevant concentrations confirmed minimal molecular features of toxicity, suggesting that direct physical effects are responsible for short-term microbial population decline. In an additional experiment, the diversity and structure of natural marine microbial communities showed negligible variations when exposed to environmentally relevant nTiO2 concentrations (i.e., 25 ÎŒg L−1). As such, the environmental risk of nTiO2 towards marine microbial species appears low, however the potential for adverse effects in hotspots of contamination exists. In future, research must be extended to consider any effect of other components of nano-enabled product formulations upon nanomaterial fate and impact within the natural environment

    Gad65 is recognized by t-cells, but not by antibodies from nod-mice

    Since the 64kDa-protein glutamic acid decarboxylase (GAD) is one of the major autoantigens in T-cell mediated Type 1 diabetes, its relevance as a T-cell antigen needs to be clarified. After isolation of splenic T-cells from non-obese diabetic (NOD) mice, a useful model for human Type 1 diabetes, we found that these T-cells proliferate spontaneously when incubated with human GAD65, but only marginally after incubation with GAD67, both recombinated in the baculovirus system. No effect was observed with non-diabetic NOD mice or with T-cells from H-2 identical NON-NOD-H-2g7 control mice. It has been published previously that NOD mice develop autoantibodies against a 64kDa protein detected with mouse beta cells. In immunoprecipitation experiments with sera from the same NOD mice and 33S-methionine-labelled GAD, no autoantibody binding could be detected. We conclude firstly that GAD65 is an important T-cell antigen which is relevant early in the development of Type 1 diabetes and secondly that there is an antigenic epitope in the human GAD65 molecule recognized by NOD T-cells, but not by NOD autoantibodies precipitating conformational epitopes. Our results therefore provide further evidence that GAD65 is a T-cell antigen in NOD mice, being possibly also involved in very early processes leading to the development of human Type 1 diabetes

    Dietary elimination of children with food protein induced gastrointestinal allergy – micronutrient adequacy with and without a hypoallergenic formula?

    Background: The cornerstone for management of Food protein-induced gastrointestinal allergy (FPGIA) is dietary exclusion; however the micronutrient intake of this population has been poorly studied. We set out to determine the dietary intake of children on an elimination diet for this food allergy and hypothesised that the type of elimination diet and the presence of a hypoallergenic formula (HF) significantly impacts on micronutrient intake. Method: A prospective observational study was conducted on children diagnosed with FPIGA on an exclusion diet who completed a 3 day semi-quantitative food diary 4 weeks after commencing the diet. Nutritional intake where HF was used was compared to those without HF, with or without a vitamin and mineral supplement (VMS). Results: One-hundred-and-five food diaries were included in the data analysis: 70 boys (66.7%) with median age of 21.8 months [IQR: 10 - 67.7]. Fifty-three children (50.5%) consumed a HF and the volume of consumption was correlated to micronutrient intake. Significantly (p <0.05) more children reached their micronutrient requirements if a HF was consumed. In those without a HF, some continued not to achieve requirements in particular for vitamin D and zinc, in spite of VMS. Conclusion: This study points towards the important micronutrient contribution of a HF in children with FPIGA. Children, who are not on a HF and without a VMS, are at increased risk of low intakes in particular vitamin D and zinc. Further studies need to be performed, to assess whether dietary intake translates into actual biological deficiencies

    The aim of this article is to highlight the importance of 'spatiality' in understanding the materialization of risk society and cultivation of risk sensibilities. More specifically it provides a cultural analysis of pathogen virulence (as a social phenomenon) by means of tracing and mapping the spatial flows that operate in the uncharted zones between the microphysics of infection and the macrophysics of epidemics. It will be argued that epidemic space consists of three types of forces: the vector, the index and the vortex. It will draw on Latour's Actor Network Theory to argue that epidemic space is geared towards instability when the vortex (of expanding associations and concerns) displaces the index (of finding a single cause)

    Predictors of Recovery from Prenatal Depressive Symptoms from Pregnancy Through Postpartum

    Abstract Background: Identifying predictors of the course of depressive symptoms from pregnancy through postpartum is important to inform clinical interventions. Methods: This longitudinal study investigated predictors of recovery from prenatal elevated depressive symptoms in the postpartum period. Forty-one pregnant women completed demographic, interpersonal, and psychosocial self-report assessment measures at 32 weeks of gestation and again 12 weeks postpartum. Results: Of those with elevated depressive symptoms, defined as a Beck Depression Inventory-II (BDI-II) score ≄10, at the prenatal baseline, 39% (n=16) recovered to nonelevated symptom levels postpartum, whereas 61% (n=25) experienced sustained elevated symptoms. Women who recovered evidenced significantly lower baseline depression severity and more frequent engagement in physical activity and cohabitated with a romantic partner. In multiparous women (n=25), history of past postpartum depression (PPD) differentiated between those with transient and those with persisting symptoms, although history of lifetime depression did not. None of the additional demographic, interpersonal, or psychosocial variables investigated differentiated between groups. Logistic regression analysis showed prenatal depression severity and exercise frequency as predictors of recovery postpartum. Conclusions: Results suggest most women will not experience spontaneous recovery. Women with prenatal heightened symptom severity and previous experiences with PPD are acutely vulnerable to experience sustained symptoms. In contrast, having a cohabitating partner and engagement in prenatal exercise predicted symptom improvement. Physical exercise may be an important clinical recommendation, as it may improve mood. Given the small sample size, these results are preliminary.
