    Morphology and Anatomy of Foliar Nectaries and Associated Leaves in Mallotus (Euphorbiaceae)

    The morphology and anatomy of the foliar nectaries and associated leaves offour species of Mallotus (Euphorbiaceae) were studied. Light microscopic observations of paraffin- and plastic-embedded specimens were complemented with scanning electron micrographs. Leaf anatomy of the four species is typical of large mesophytic plants. Aattened foliar nectaries are shown to be composed of specialized epidermal cells. The nonvascularized nectaries consist of narrow columnar cells each with a large nucleus, numerous vacuoles, and dense cytoplasm. Subglandular parenchyma cells have more pronounced nuclei, more vacuoles and denser cytoplasm than do typical laminar parenchyma. Structurally, these nectaries are similar to those found in other taxa of Euphorbiaceae and in other families of flowering plants. Brief field observations confirmed that ants are readily attracted to the nectar and probably function in a mutualistic relationship with the plants. The actual mechanism of nectar secretion was not studied

    Effect of Different Steeping Conditions on the Peroxidase Activities of Some Improved Sorghum Varieties

    In order to evaluate the impact of duration and the incorporation or  otherwise of air rest during steeping on the peroxidase activities of  sorghum grains, five improved varieties were studied. Steeping durations used ranged from 0 (raw grains) to 72 hours, in two formats: with air rest and without air rest. Results obtained showed that among the raw grains which were the control experiment, variety SK5912 gave the highest  peroxidase activity, followed by variety KSV 8 while variety CSRO2 had the least peroxidase activity. During steeping, we observed that in almost all cases, higher peroxidase activities were obtained with air rested sorghum grains than those without air rest. In terms of how the different varieties compared during steeping, we found that variety KSV 8 gave the highest peroxidase activity at both with and without air rest regimes in all the steeping periods used. Also, we observed that in all cases, the highest increase in peroxidase expression was obtained during the first 6 hours of steeping, shown by a steep rise of over 50% increase in activity. The other steeping times following the first 6 hours however showed just gradual  increases in peroxidase over the period of sorghum. The major significance of this result is that although increasing duration showed increased  peroxidase activities, such increases were less jumpy than those obtained during the first 6 hours. Therefore, long steeping times may not  necessarily be very beneficial, with the added benefit that costs associated with long steeping durations may be avoided.Key words: Sorghum, cereals, peroxidases, enzymes, steeping, germination, air rest

    Design principles for riboswitch function

    Scientific and technological advances that enable the tuning of integrated regulatory components to match network and system requirements are critical to reliably control the function of biological systems. RNA provides a promising building block for the construction of tunable regulatory components based on its rich regulatory capacity and our current understanding of the sequence–function relationship. One prominent example of RNA-based regulatory components is riboswitches, genetic elements that mediate ligand control of gene expression through diverse regulatory mechanisms. While characterization of natural and synthetic riboswitches has revealed that riboswitch function can be modulated through sequence alteration, no quantitative frameworks exist to investigate or guide riboswitch tuning. Here, we combined mathematical modeling and experimental approaches to investigate the relationship between riboswitch function and performance. Model results demonstrated that the competition between reversible and irreversible rate constants dictates performance for different regulatory mechanisms. We also found that practical system restrictions, such as an upper limit on ligand concentration, can significantly alter the requirements for riboswitch performance, necessitating alternative tuning strategies. Previous experimental data for natural and synthetic riboswitches as well as experiments conducted in this work support model predictions. From our results, we developed a set of general design principles for synthetic riboswitches. Our results also provide a foundation from which to investigate how natural riboswitches are tuned to meet systems-level regulatory demands

    The statistical neuroanatomy of frontal networks in the macaque

    We were interested in gaining insight into the functional properties of frontal networks based upon their anatomical inputs. We took a neuroinformatics approach, carrying out maximum likelihood hierarchical cluster analysis on 25 frontal cortical areas based upon their anatomical connections, with 68 input areas representing exterosensory, chemosensory, motor, limbic, and other frontal inputs. The analysis revealed a set of statistically robust clusters. We used these clusters to divide the frontal areas into 5 groups, including ventral-lateral, ventral-medial, dorsal-medial, dorsal-lateral, and caudal-orbital groups. Each of these groups was defined by a unique set of inputs. This organization provides insight into the differential roles of each group of areas and suggests a gradient by which orbital and ventral-medial areas may be responsible for decision-making processes based on emotion and primary reinforcers, and lateral frontal areas are more involved in integrating affective and rational information into a common framework

    Rudimentary G-Quadruplex-Based Telomere Capping In Saccharomyces Cerevisiae

    Telomere capping conceals chromosome ends from exonucleases and checkpoints, but the full range of capping mechanisms is not well defined. Telomeres have the potential to form G-quadruplex (G4) DNA, although evidence for telomere G4 DNA function in vivo is limited. In budding yeast, capping requires the Cdc13 protein and is lost at nonpermissive temperatures in cdc13-1 mutants. Here, we use several independent G4 DNA-stabilizing treatments to suppress cdc13-1 capping defects. These include overexpression of three different G4 DNA binding proteins, loss of the G4 DNA unwinding helicase Sgs1, or treatment with small molecule G4 DNA ligands. In vitro, we show that protein-bound G4 DNA at a 3\u27 overhang inhibits 5\u27-\u3e 3\u27 resection of a paired strand by exonuclease I. These findings demonstrate that, at least in the absence of full natural capping, G4 DNA can play a positive role at telomeres in vivo

    Mobile Offloading in Residential Wireless Access Markets

    The growth of mobile data traffic has been increasing at a tremendous pace. Currently, mobile broadband is mostly served by macro base stations. Strong attenuation occurs when the signals penetrate through buildings affecting the quality of service. Hence, mobile operators need to enhance the capacity to minimise congestion problems and improve the coverage on their macro networks for better network operation. As most of the mobile traffic occurs indoors, there is a need for indoor network deployments. A qualitative analysis employing the combination of various research methods (value network configuration, system dynamics, expert interviews) has been carried out to investigate various factors besides mobile offloading, which could lead to a large-scale deployment of femtocells in homes with the focus on Finland. This study also discusses several options to cope with the mobile traffic growth and examines different indoor network deployment scenarios. Furthermore, the scenarios of Femtocell-as-a-Service are highlighted and its possible impacts on mobile operators’ business are covered as well. The analysis is also conducted to illustrate how different actors are interrelated in the home network business. The analysis reveals that the primary factor that will contribute to the success of femtocell deployment in homes is the need for future mobile offloading. However, many other factors have important implications which require careful consideration by mobile operators. With the presence of good macro network capacity like in Finland, new services and partnership with third party providers has been identified as the most important factor. Mobile operators need to offer an incentive to attract end-users to adopt a femtocell service due to the strong positioning of WiFi in homes. Therefore, new services would also enable the penetration of femtocells into homes to compete with WiFi access points. Three factors (interference mitigation and interoperability, femtocell management system, backhaul) are also important as these are needed to ensure the femtocell networks operate reliably. As lowering the cost is essential for building scale, the model of Femtocell-as-a-Service is useful as it reduces capital investments for mobile operators. Interviews with the experts found that major mobile operators are less likely to adopt this model. This is better suited for smaller mobile operators that want to quickly enter the femtocell business

    Large introns in relation to alternative splicing and gene evolution: a case study of Drosophila bruno-3

    Background: Alternative splicing (AS) of maturing mRNA can generate structurally and functionally distinct transcripts from the same gene. Recent bioinformatic analyses of available genome databases inferred a positive correlation between intron length and AS. To study the interplay between intron length and AS empirically and in more detail, we analyzed the diversity of alternatively spliced transcripts (ASTs) in the Drosophila RNA-binding Bruno-3 (Bru-3) gene. This gene was known to encode thirteen exons separated by introns of diverse sizes, ranging from 71 to 41,973 nucleotides in D. melanogaster. Although Bru-3's structure is expected to be conducive to AS, only two ASTs of this gene were previously described. Results: Cloning of RT-PCR products of the entire ORF from four species representing three diverged Drosophila lineages provided an evolutionary perspective, high sensitivity, and long-range contiguity of splice choices currently unattainable by high-throughput methods. Consequently, we identified three new exons, a new exon fragment and thirty-three previously unknown ASTs of Bru-3. All exon-skipping events in the gene were mapped to the exons surrounded by introns of at least 800 nucleotides, whereas exons split by introns of less than 250 nucleotides were always spliced contiguously in mRNA. Cases of exon loss and creation during Bru-3 evolution in Drosophila were also localized within large introns. Notably, we identified a true de novo exon gain: exon 8 was created along the lineage of the obscura group from intronic sequence between cryptic splice sites conserved among all Drosophila species surveyed. Exon 8 was included in mature mRNA by the species representing all the major branches of the obscura group. To our knowledge, the origin of exon 8 is the first documented case of exonization of intronic sequence outside vertebrates. Conclusion: We found that large introns can promote AS via exon-skipping and exon turnover during evolution likely due to frequent errors in their removal from maturing mRNA. Large introns could be a reservoir of genetic diversity, because they have a greater number of mutable sites than short introns. Taken together, gene structure can constrain and/or promote gene evolution

    The prevalence and bacteriology of a symptomatic bacteriuria among antenatal patients in Nnamdi Azikiwe University Teaching Hospital Nnewi; South Eastern Nigeria

    Urinary tract infection in pregnancy leads to poor pregnancy outcome. Diagnosis and treatment of asymptomatic bacteriuria markedly improves pregnancy outcome as well as reduce the incidence of acutepyelonephritis. To determine the prevalence and bacteriology of asymptomatic bacteriuria among Antenatal patients in our centre, and to know if routine screening will be justifiable. This was a prospective study carried out between April and August 2008. Sample size was statistically determined.Women who consented were interviewed and mid stream urine samples were collected and processed in the microbiology laboratory, using standard microbiological methods. Out of 357 women studied, 65(18.21%) had significant bacteriuria. was the commonest isolate (25.6%), while was the least frequent isolate (3.66%). Women in third trimester had the highest prevalence (25.68%) while those in the first trimester had the least (15.79%).Women that had only primary education had the highest prevalence (27.50%) while those that had tertiary educationhad the least prevalence (21.10%). The prevalence of significant asymptomatic bacteriuria among the women studied was high.Screening of all the pregnant women and treatment will reduce the incidence and complications of overt urinary tract infection in pregnancy among these women

    Influence of mine drainage on water quality along River Nyaba in Enugu South-Eastern Nigeria

    Major and Trace elements concentration were measured in water samples collected in and around Okpara coal mine in Enugu southeastern Nigeria to investigate the influence of mine drainage on the quality of water. The cations and trace elements were determined by ICP- MS while the anions were measured by spectrophotometer and titration methods. Field parameters such as pH, temperature and conductivity were determined in the field using standard equipment. The results show that the water is acidic to moderately acidic (pH 2.84 to 6.69) with pH increasing along the flow direction. The mean values of pH (4.66 (dry), 4.22 (wet), Color (334.34 TCU (dry), 153.11 TCU (wet) and turbidity (53,67 NTU (dry), 17.43 NTU (wet) as well as iron (6.35 mg/L(dry), 5.14 mg/L(wet), aluminum(1.14 mg/L(dry), 4.30 mg/L(wet), manganese (1.43 mg/L(dry), 5.36 mg/L (wet) and nickel, 0.053 mg/L (wet) recorded in the dry and wet seasons are above levels recommended by WHO for drinking water and other domestic purposes. Mean levels of fluoride (5.4 mg/L) with ranged of 0.00 to 45 mg/L, potassium (12 mg/L) with ranged of 1.17 to 27.85 mg/L and nickel (53.10 ìg/L) with ranged of 1.50 to 309.30 ìg/L, as well as maximum levels of chromium (100 ìg/L) with ranged of 0.05 to 100 ìg/L , chloride(400 mg/L) with ranged of 40 to 400 mg/L, nitrate(1012 mg/L) with ranged of 158 to 1012 mg/L and sulphate (517 mg/L) with ranged of 10 to 512 mg/L obtained in the wet season are above the WHO maximum permissible level. Generally, the levels of the elements decrease with distance away from the mine waste except for nitrate and fluoride. Thus the quality of the water is most probably influenced by acidic mine drainage and it impact on human health and the environment could be severe. Microbial assessment and element speciation are recommended for further quality assessment in the study area
