    By integrating, on a wafer plane, GaAs semiconductor optoelectronic modulators and detectors with computer-generated holograms between then, the potential for in-plane interconnections is proposed. We report the fabrication and characterisation of a binary-phase relief hologram etched in a GaAs wafer using an averaged Fresnel zone plate design to focus light to 2 x 2 spots for array interconnection. Efficiencies of 28% for this design of binary CGH etched in GaAs have been achieved, close to the theoretical maximum

    Spatiotemporal variation of Diptera changes how we evaluate High Nature Value (HNV) wet grasslands

    Proposed strategies to protect biodiversity within agricultural systems are often based on botanical criteria with plant species richness generally considered the prime indicator of conservation potential. While wet grasslands dominated by rushes (Juncus spp.) are commonly considered to be of lesser ecological value than those which are more botanically diverse (e.g. Carex dominated wet grasslands), their value for invertebrates such as Diptera has not yet been fully explored. Data from two Diptera families (Sciomyzidae and Syrphidae) were examined at spatial, temporal and spatiotemporal scales to determine the contribution of two different (Juncus and Carex dominated) wet grassland habitats towards the maintenance of Diptera diversity. The two habitats were significantly different in terms of community structure for both families and temporal variation was a significant component of dipteran diversity. Spatiotemporal analysis showed that species turnover between habitats at different times made the most significant contribution to overall Diptera diversity. Temporal variation of both families suggests that the relative importance of each habitat type to overall diversity fluctuates depending on sampling period, with both habitats supporting diversity at different times. Our results indicate that lowland wet grasslands characterised by Juncus cover need to be recognised as ecologically important for the maintenance of dipteran diversity. We discuss the possible implications for the diversity of Diptera in wet grasslands if these commonly perceived marginal areas (both agriculturally and ecologically) are ignored in conservation strategies. The necessity of recognising spatiotemporal variation when evaluating habitats using invertebrates as indicators is also discussed. © 2017 Springer Science+Business Media Dordrech

    Indicators of Diptera diversity in wet grassland habitats are influenced by environmental variability, scale of observation, and habitat type

    In low intensity agri-ecosystems such as wet grassland habitats, the inclusion of invertebrates in conservation assessments and monitoring is usually limited to charismatic groups such as bees or butterflies. However, wet grasslands support a wide range of inveterate groups, some of which may exhibit limited movement not generally represented by more mobile groups such as those typically examined. The use of surrogate species which exemplify broader invertebrate diversity has been suggested as a possible means of including these overlooked invertebrates (such as Diptera) in conservation planning within these habitats. Based on collections made by Malaise trap, we utilized two families of Diptera (Sciomyzidae and Syrphidae) as indicators of a wider range of dipteran diversity (nine Diptera families identified to parataxonomic unit level [PUs]) in wet grassland habitats. We examined the role of environmental variability, spatial scale, and habitat type on patterns of cross-taxon congruence for all three assemblages. Both environmental correlation and community congruence were significantly stronger among assemblages when examined at low spatial scales, highlighting the need to examine dipteran groups at scales untypical of current agri-environmental assessments; namely field and farm level. Furthermore, when wet grasslands were differentiated into two habitat categories (sedge and rush dominated grasslands), the significance of the community congruence increased markedly. This correlation was particularly strong between Sciomyzidae and PUs which demonstrated similar differentiation based on habitat type, implying that assemblages which exhibit comparable ecological partitioning are more likely to be useful surrogates of one another. Correlations between richness, abundance and Shannon's diversity were highly variable among groups, suggesting compositional analysis as the most appropriate examination of dipteran diversity for surrogacy studies. The results indicate that cross-assemblage congruence of Diptera is influenced by similarity of response to environmental variability, scale of observation, and examination of assemblages differentiated into appropriate habitat categories. The results illustrate the need to investigate invertebrate biodiversity surrogates at scales appropriate to the indicator groups and examine congruence among assemblages within specific habitat categories. Such an approach has the potential to maximise gamma diversity in areas where wet grasslands are under threat of intensification or abandonment. © 2017 Elsevier Lt

    The presence of Tetanocera punctifrons Rondani, 1868 in Ireland is reconfirmed. The species is distinguished from the very similar T. latifibula Frey, 1924. The female abdomens of both species are described for the first time. The distributions of both species are summarised. The history of type examinations and of taxonomists’ conceptions of the two species is tracked, especially the relative reliability of published figures of diagnostic features. We emphasise the need for such analyses of rare and closely related species, even if apparently disjunct in distribution. Habitats of T. punctifrons and T. latifibula are described, and the biology and morphology of the immature stages are compared

    Modularity and Intrinsic Evolvability of Hsp90-Buffered Change

    Hsp90 controls dramatic phenotypic transitions in a wide array of morphological features of many organisms. The genetic-background dependence of specific abnormalities and their response to laboratory selection suggested Hsp90 could be an ‘evolutionary capacitor’, allowing developmental variation to accumulate as neutral alleles under normal conditions and manifest selectable morphological differences during environmental stress. The relevance of Hsp90-buffered variation for evolution has been most often challenged by the idea that large morphological changes controlled by Hsp90 are unconditionally deleterious. To address this issue, we tested an Hsp90-buffered abnormality in Drosophila for unselected pleiotropic effects and correlated fitness costs. Up to 120-fold differences in penetrance among six highly related selection lines, started from an initially small number of flies and rapidly selected for and against a deformed eye trait (dfe), did not translate into measurable differences in any of several tests of viability, lifespan or competitive fitness. Nor were 17 dfe Quantitative Trait Loci (QTL) associated with fitness effects in over 1,400 recombinant lines. Our ability to detect measurable effects of inbreeding, media environment and the white mutation in the selection line backgrounds independent of dfe penetrance suggests that, within the limitations of laboratory tests of fitness, this large morphological change controlled by Hsp90 was selectable independent of strong, correlated and unconditionally deleterious effects—abundant, polygenic variation hidden by Hsp90 allows potentially deleterious alleles to be readily replaced during selection by less deleterious alleles with similar phenotypic effects. Hsp90 links environmental stress with the expression of developmental variation controlling unprecedented morphological plasticity. As outlined here and in the companion paper of this issue, the complex genetic architecture of Hsp90-buffered variation supports a remarkable modularity of Hsp90 effects on quantitative and qualitative phenotypes, consistent with the ‘Hsp90 capacitor hypothesis’ and standard quantitative genetic models of threshold traits

    Tumor innate immunity primed by specific interferon-stimulated endogenous retroviruses.

    Mesenchymal tumor subpopulations secrete pro-tumorigenic cytokines and promote treatment resistance1-4. This phenomenon has been implicated in chemorefractory small cell lung cancer and resistance to targeted therapies5-8, but remains incompletely defined. Here, we identify a subclass of endogenous retroviruses (ERVs) that engages innate immune signaling in these cells. Stimulated 3 prime antisense retroviral coding sequences (SPARCS) are oriented inversely in 3' untranslated regions of specific genes enriched for regulation by STAT1 and EZH2. Derepression of these loci results in double-stranded RNA generation following IFN-γ exposure due to bi-directional transcription from the STAT1-activated gene promoter and the 5' long terminal repeat of the antisense ERV. Engagement of MAVS and STING activates downstream TBK1, IRF3, and STAT1 signaling, sustaining a positive feedback loop. SPARCS induction in human tumors is tightly associated with major histocompatibility complex class 1 expression, mesenchymal markers, and downregulation of chromatin modifying enzymes, including EZH2. Analysis of cell lines with high inducible SPARCS expression reveals strong association with an AXL/MET-positive mesenchymal cell state. While SPARCS-high tumors are immune infiltrated, they also exhibit multiple features of an immune-suppressed microenviroment. Together, these data unveil a subclass of ERVs whose derepression triggers pathologic innate immune signaling in cancer, with important implications for cancer immunotherapy

    Readability estimates for commonly used health-related quality of life surveys

    To estimate readability of seven commonly used health-related quality of life instruments: SF-36, HUI, EQ-5D, QWB-SA, HALex, Minnesota Living with Heart Failure Questionnaire (MLHFQ), and the NEI-VFQ-25. The Flesch–Kincaid (F–K) and Flesch Reading Ease (FRE) formulae were used to estimate readability for every item in each measure. The percentage of items that require more than 5 years of formal schooling according to F–K was 50 for the EQ-5D, 53 for the SF-36, 80 for the VFQ-25, 85 for the QWB-SA, 100 for the HUI, HALex, and the MLHFQ. The percentage of items deemed harder than “easy” according to FRE was 50 for the SF-36, 67 for the EQ-5D, 79 for the QWB-SA, 80 for the VFQ-25, 100 for the HUI, HALex, and the MLHFQ. All seven surveys have a substantial number of items with high readability levels that may not be appropriate for the general population

    Estimating health-adjusted life expectancy conditional on risk factors: results for smoking and obesity

    BACKGROUND: Smoking and obesity are risk factors causing a large burden of disease. To help formulate and prioritize among smoking and obesity prevention activities, estimations of health-adjusted life expectancy (HALE) for cohorts that differ solely in their lifestyle (e.g. smoking vs. non smoking) can provide valuable information. Furthermore, in combination with estimates of life expectancy (LE), it can be tested whether prevention of obesity and smoking results in compression of morbidity. METHODS: Using a dynamic population model that calculates the incidence of chronic disease conditional on epidemiological risk factors, we estimated LE and HALE at age 20 for a cohort of smokers with a normal weight (BMI < 25), a cohort of non-smoking obese people (BMI>30) and a cohort of 'healthy living' people (i.e. non smoking with a BMI < 25). Health state valuations for the different cohorts were calculated using the estimated disease prevalence rates in combination with data from the Dutch Burden of Disease study. Health state valuations are multiplied with life years to estimate HALE. Absolute compression of morbidity is defined as a reduction in unhealthy life expectancy (LE-HALE) and relative compression as a reduction in the proportion of life lived in good health (LE-HALE)/LE. RESULTS: Estimates of HALE are highest for a 'healthy living' cohort (54.8 years for men and 55.4 years for women at age 20). Differences in HALE compared to 'healthy living' men at age 20 are 7.8 and 4.6 for respectively smoking and obese men. Differences in HALE compared to 'healthy living' women at age 20 are 6.0 and 4.5 for respectively smoking and obese women. Unhealthy life expectancy is about equal for all cohorts, meaning that successful prevention would not result in absolute compression of morbidity. Sensitivity analyses demonstrate that although estimates of LE and HALE are sensitive to changes in disease epidemiology, differences in LE and HALE between the different cohorts are fairly robust. In most cases, elimination of smoking or obesity does not result in absolute compression of morbidity but slightly increases the part of life lived in good health. CONCLUSION: Differences in HALE between smoking, obese and 'healthy living' cohorts are substantial and similar to differences in LE. However, our results do not indicate that substantial compression of morbidity is to be expected as a result of successful smoking or obesity prevention
