    Determinants of residents’ support for sustainable tourism development: an empirical study in Midyat, Turkey

    The purpose of this study is to examine the extent to which residents’ perceptions of their place image can predict their perceptions of the impacts of tourism, and ultimately, to elucidate their support for sustainable tourism development in Midyat, Turkey. This city currently faces a range of negative impacts associated with tourism, such as inflation, high leakage, threats to family structures, environmental degradation, pollution, and crowding. At the same time, Midyat seeks to maximize the positive impacts of tourism, including job creation and employment, environmental protection, cultural and educational enrichment, and recreational opportunities. Therefore, there is an urgent need for a strategic shift in development. The study population consists of residents residing in Midyat, Turkey, from whom three hundred and fifty-six questionnaires were collected. PLS (Partial Least Squares) path analysis was utilized to analyze the research model constructed based on the literature. The results demonstrated that residents’ place image significantly predicted two out of three impacts of tourism, namely, socio-cultural and economic impacts. Additionally, residents’ perceptions of environmental and socio-cultural impacts were significant predictors of their support or opposition to sustainable tourism. While perceived environmental impacts have a positive effect on support for sustainable tourism, perceived economic impacts have a negative effect. This finding can guide tourism planners and professionals to make more informed decisions and take stronger steps toward sustainable tourism development. This study revealed that the quality of life, job opportunities, infrastructure, and economic income of Midyat’s residents are insufficient. However, the city is characterized by social attributes, such as friendliness, trust, and supportiveness. The results are limited to local residents, and using only one tourist stakeholder to assess sustainable tourism development is insufficient.     Modeling of an AC-electro-osmosis based microfluidic mixer

    The purpose of this study is presenting an active micromixer, which is based on AC electro-osmotic flow driven on 3D micro wires. In order to solve governing equations of AC electroosmosis, a commercial software COMSOL Multiphysics® is implemented. Different wire configurations with various imposed electric fields and flow rates are tested for evaluating mixing efficiencies. The analyses show that mixing performance is significantly improved by number of the wires as well as wire orientation. It is also revealed that the degree of mixing can also be controlled by the tuning of the applied voltage for a given flow rate. © 2017 ASME The American Society of Mechanical Engineers

    Deep reinforcement learning for subpixel neural tracking

    Automatically tracing elongated structures, such as axons and blood vessels, is a challenging problem in the field of biomedical imaging, but one with many downstream applications. Real, labelled data is sparse, and existing algorithms either lack robustness to different datasets, or otherwise require significant manual tuning. Here, we instead learn a tracking algorithm in a synthetic environment, and apply it to tracing axons. To do so, we formulate tracking as a reinforcement learning problem, and apply deep reinforcement learning techniques with a continuous action space to learn how to track at the subpixel level. We train our model on simple synthetic data and test it on mouse cortical two-photon microscopy images. Despite the domain gap, our model approaches the performance of a heavily engineered tracker from a standard analysis suite for neuronal microscopy. We show that fine-tuning on real data improves performance, allowing better transfer when real labelled data is available. Finally, we demonstrate that our model's uncertainty measure-a feature lacking in hand-engineered trackers-corresponds with how well it tracks the structure

    The effect of racemic gossypol and AT-101 on angiogenic profile of OVCAR-3 cells: a preliminary molecular framework for gossypol enantiomers

    To compare the effect of racemic gossypol with its (–)/(–) enantiomer (AT-101) on expression profiles of angiogenic molecules by mRNA levels in human ovarian cancer cell line OVCAR-3. Methods: Cell viability assay (2,3-bis (2-methoxy-4-nitro-5- sulfophenyl)-5-[(phenylamino) carbonyl]-2H-tetrazolium hydroxide) was used to detect cytotoxicity of gossypol enantiomers. DNA fragmentation by an enzyme-linked immunosorbent (ELISA) assay was used to evaluate the rate of apoptosis. The mRNA expression levels of angiogenic molecules were investigated by Human Angiogenesis RT2 ProfilerTM PCR Array (SuperArray, Frederick, MD). Results: Both racemic form and AT-101 resulted in a significant cytotoxicity and induced apoptosis. This effect was observed in a dose- and time dependent manner. However, AT-101 was much more potent. In addition, the treatment of 10 μM of racemic gossypol alone and 3 μM of AT-101 alone resulted in significant down-regulation (≥ 3 fold) in mRNA levels of some pivotal angiogenic molecules in OVCAR-3, but altered gene profiles were different by the treatment of each enantiomer. Conclusion: The efficacy of two gossypol enantiomers in OVCAR-3 cells showed distinction. AT-101 was much more potent than racemic gossypol, not only by means of cell death and apoptosis, but also by modulation of angiogenic molecules released from OVCAR-3 cells. Further studies with endothelial cells should be done to verify the anti-angiogenic effect of gossypol enantiomers in cancer treatment

    Improving the Performance of Thinning Algorithms with Directed Rooted Acyclic Graphs

    In this paper we propose a strategy to optimize the performance of thinning algorithms. This solution is obtained by combining three proven strategies for binary images neighborhood exploration, namely modeling the problem with an optimal decision tree, reusing pixels from the previous step of the algorithm, and reducing the code footprint by means of Directed Rooted Acyclic Graphs. A complete and open-source benchmarking suite is also provided. Experimental results confirm that the proposed algorithms clearly outperform classical implementations

    Evaluation of the nasal mucociliary transport rate by rhinoscintigraphy before and after surgery in patients with deviated nasal septum

    In this study, we have investigated the effect of nasal septal deviation (NSD) on nasal mucociliary activity and how does a septoplasty operation affecs the nasal mucociliary transport rate in the first and third months during the post-operative period. Twenty-two patients who were diagnosed with NSD and 22 healthy controls were studied using rhinoscintigraphy with Tc-99m-macroaggregated albumin (Tc-99m-MAA). On each case, the nasal mucociliary transport rate (NMTR) was measured pre-operatively only on five cases, on the first and third months of post-operative period. The NMTRs of patients with a deviated septum were significantly lower than the NMTRs of the healthy controls on both the convex and concave sides. Significant improvement was observed in the first post-operative month. On the concave and convex sides, the average postop third month post-operative NMTR value was higher than the first month post-operative NMTR values. It was concluded that the septoplasty operation improves reduced NMTRs after surgery. The effect of nasal surgery on nasal mucociliary activity may be more accurately evaluated in the third month than the first month of post-operative period

    Anisomycin activates JNK and sensitises DU 145 prostate carcinoma cells to Fas mediated apoptosis

    Treatment of the hormone refractory prostate cancer cell line DU 145 with sublethal concentrations of chemotherapeutic drugs has been reported to sensitise these cells to Fas mediated apoptosis. However, the mechanism by which this occurs has not been determined. Our group has shown that inhibition of JNK activity completely abrogates the effects of chemotherapeutic drugs. Using anisomycin, a potent JNK agonist, we have demonstrated a role for JNK in Fas mediated apoptosis in DU 145 cells. Inhibition of Caspase 8 and Caspase 9 completely inhibits this process which suggests that DU 145 cells require mitochondrial amplification of the Fas apoptotic signal. Furthermore, we have shown that inhibition of Fas mediated apoptosis is an early event in DU 145 cells, occurring upstream of Caspase 8 cleavage. It is hoped that identifying the target of JNK will allow novel therapies to be developed for the treatment of hormone refractory prostate cancer. Such therapies are especially important because no single or combined treatment to date has significantly prolonged survival in patients with hormone refractory prostate cancer

    The Role of Lactic Acid Adsorption by Ion Exchange Chromatography

    Background: The polyacrylic resin Amberlite IRA-67 is a promising adsorbent for lactic acid extraction from aqueous solution, but little systematic research has been devoted to the separation efficiency of lactic acid under different operating conditions. Methodology/Principal Findings: In this paper, we investigated the effects of temperature, resin dose and lactic acid loading concentration on the adsorption of lactic acid by Amberlite IRA-67 in batch kinetic experiments. The obtained kinetic data followed the pseudo-second order model well and both the equilibrium and ultimate adsorption slightly decreased with the increase of the temperature at 293–323K and 42.5 g/liter lactic acid loading concentration. The adsorption was a chemically heterogeneous process with a mean free energy value of 12.18 kJ/mol. According to the Boyd _ plot, the lactic acid uptake process was primarily found to be an intraparticle diffusion at a lower concentration (,50 g/liter) but a film diffusion at a higher concentration (.70 g/liter). The values of effective diffusion coefficient D i increased with temperature. By using our Equation (21), the negative values of DGu and DHu revealed that the adsorption process was spontaneous and exothermic. Moreover, the negative value of DSu reflected the decrease of solid-liquid interface randomness at the solid-liquid interface when adsorbing lactic acid on IRA-67.     Dexamethasone potentiates the antiangiogenic activity of docetaxel in castration-resistant prostate cancer

    We sought to characterise whether dexamethasone (DEX) may enhance tumour response to docetaxel in in vitro and in vivo models of metastatic prostate cancer (CaP). In vitro experiments conducted on PC3 and human bone marrow endothelial cells (hBMECs) determined that administration of DEX (10 nM) reduced constitutive nuclear factor-κB (NF-κB) activity, decreasing interleukin (IL)-8, CXCL1 and VEGF gene expression in PC3 cells. Dexamethasone also attenuated docetaxel-induced NF-κB and activator protein-1 transcription and reduced docetaxel-promoted expression/secretion of IL-8 and CXCL1 in PC3 and hBMECs. Although DEX failed to enhance docetaxel cytotoxicity on PC3 cells, DEX potentiated the antiangiogenic activity of docetaxel in vitro, further reducing vessel area and vessel length in developing endothelial tubes (P<0.05). Docetaxel had a potent antiangiogenic activity in the dorsal skin flap-implanted PC3 tumours in vivo. Small blood vessel formation was further suppressed in tumours co-treated with docetaxel and DEX, substantiated by an increased average vessel diameter and segment length and a decreased number of branch points in the residual tumour vasculature (P<0.001). Our data show that DEX potentiates the antiangiogenic activity of docetaxel, suggesting a putative mechanism for the palliative and survival benefits of these agents in metastatic CaP
