    Impact of pre-treatments on properties of lignocelluloses and their accessibility for a subsequent carboxymethylation

    In this issue, different chemical (alkaline and sulfite pulping, ozonolysis) and mechanical (vibratory ball milling) pre-treatments were utilized for activating wheat straw and beech sawdust prior to carboxymethylation. Detailed analysis by a range of methods, including Klason-lignin, cellulose and hemicellulose quantification, Powder-X-ray diffraction (PXRD) and attenuated total reflection (ATR) IR spectroscopy, enabled the investigation of material alterations. Subsequently, carboxymethylation was carried out with both untreated and activated materials, allowing the evaluation of activation steps by determining degrees of substitution with carboxymethyl groups (DSCM). Moreover, carboxymethylation conditions were optimized, realizing high DSCM of up to 1.05. Results further revealed that ball milling enhanced the subsequent conversion; whereas chemical pre-treatments did not effectively increase material accessibilities. Further studies on chemically untreated material s emphasized that a highly reactive surface was already generated in the course of the carboxymethylation, inter alia through the concomitant dissolution of matrix components

    Faunal diversity of Fagus sylvatica forests: A regional and European perspective based on three indicator groups

    <p>While the postglacial history of European beech (Fagus sylvatica) and the plant species composition of beech forests in  Central Europe are fairly well understood, the faunal biodiversity has been less well investigated. We studied three groups of  mostly sedentary organisms in beech forest at regional and European scales by combining field studies with a compilation of existing literature and expert knowledge. Specifically, we examined the relationship between host tree genera and saproxylic  beetles, and the diversity and composition of forest ground-dwelling molluscs and ground beetles in relation to the abundance  of beech. At a west central European scale (Germany), where beech has a “young” ecological and biogeographical history,  we found 48 primeval forest relict species of saproxylic beetles associated with beech, 124 ground beetles and 91 molluscs  inhabiting beech forest, yet none exclusive of west central European beech forests. High levels of faunal similarity between beech and other woodland trees suggested that many of the beech forest dwelling species are euryoecious and likely to  originate from mid-Holocene mixed broadleaf forests. Beech forests of the mountain ranges in southern and east central  Europe, which are ecologically and biogeographically “old”, were found to harbour distinct species assemblages, including  beech forest specialists (such as 10 carabid species in the Carpathians) and narrow-range endemics of broadleaf forest. The  observed biodiversity patterns suggest differentiated conservation priorities in “young” and “old” European beech forest  regions.</p

    Screening for trbB- and traG-like sequences by PCR for the detection of conjugative plasmids in bacterial soil isolates

MICROBIOLOGICAL RESEARCH. 2001;156(2):159-168.The transfer regions of different conjugative plasmids show significant similarities in the genetic organization and in the amino acid sequence of some gene products, especially of proteins from the traG or trbB family. These similarities are also evident on the level of the nucleotide sequences. On the basis of conserved DNA regions we designed degenerate PCR primer pairs to detect specifically tra regions within a collection of bacterial clones isolated from an agricultural soil. Most of the potential transfer-proficient indigenous bacterial isolates were able to mobilize a derivative of the nonconjugative IncQ plasmid RSF1010 into recipient strains. With the help of the primers it should be possible to evaluate the genetic potential for horizontal gene transfer carried out by conjugative plasmids
