    Digital solar system geology

    All available synoptic maps of the solid-surface bodies of the Solar System were digitized for presentation in the planned Atlas of the Solar System by Greeley and Batson. Since the last report (Batson et al., 1990), preliminary Uranian satellite maps were replaced with improved versions, Galilean satellite geology was simplified and digitized, structure was added to many maps, and the maps were converted to a standard format, with corresponding standing colors for the mapped units. Following these changes, the maps were re-reviewed by their authors and are now undergoing final editing before preparation for publication. In some cases (for Mercury, Venus, and Mars), more detailed maps were digitized and then simplified for the Atlas. Other detailed maps are planned to be digitized in the coming year for the Moon and the Galilean satellites. For most of the remaining bodies such as the Uranian satellites, the current digitized versions contain virtually all the detail that can be mapped given the available data; those versions will be unchanged for the Atlas. These digital geologic maps are archived at the digital scale of 1/16 degree/ pixel, in sinusoidal format. The availability of geology of the Solar System in a digital database will facilitate comparisons and integration with other data: digitized lunar geologic maps have already been used in a comparison with Galileo SSI observations of the Moon

    Impact cratering and the surface age of Venus: The Pre-Magellan controversy

    The average surface age of a planet is a major indicator of the level of its geologic activity and thus of the dynamics of its interior. Radar images obtained by Venera 15/16 from the northern quarter of the Venus (lat 30 to 90 degs) reveal about 150 features that resemble impact craters, and they were so interpreted by Soviet investigators B. A. Ivanov, A. T. Basilevsky, and their colleagues. These features range in diameter from about 10 to 145 km. Their areal density is remarkably similar to the density of impact structures found on the American and European continental shields. The basic difference between the Soviet and American estimates of the average surface age of Venus's northern quarter is due to which crater-production rate is used for the Venusian environment. Cratering rates based on the lunar and terrestrial cratering records, as well as statistical calculations based on observed and predicted Venus-crossing asteroids and comets, have been used in both the Soviet and American calculations. The single largest uncertainty in estimating the actual cratering rates near Venus involves the shielding effect of the atmosphere

    Expression of the chromatin-associated protein HMGA2 correlates with progression, metastasis and therapy resistance in pancreatic ductal adenocarcinoma (PDAC). Hmga2 has also been identified as a marker of a transient subpopulation of PDAC cells that has increased metastatic ability. Here, we characterize the requirement for Hmga2 during growth, dissemination, and metastasis of PDAC in vivo using conditional inactivation of Hmga2 in well-established autochthonous mouse models of PDAC. Overall survival, primary tumour burden, presence of disseminated tumour cells in the peritoneal cavity or circulating tumour cells in the blood, and presence and number of metastases were not significantly different between mice with Hmga2-wildtype or Hmga2-deficient tumours. Treatment of mice with Hmga2-wildtype and Hmga2-deficient tumours with gemcitabine did not uncover a significant impact of Hmga2-deficiency on gemcitabine sensitivity. Hmga1 and Hmga2 overlap in their expression in both human and murine PDAC, however knockdown of Hmga1 in Hmga2-deficient cancer cells also did not decrease metastatic ability. Thus, Hmga2 remains a prognostic marker which identifies a metastatic cancer cell state in primary PDAC, however Hmga2 has limited if any direct functional impact on PDAC progression and therapy resistance

    Use of tunable nanopore blockade rates to investigate colloidal dispersions

    Tunable nanopores in elastomeric membranes have been used to study the dependence of ionic current blockade rate on the concentration and electrophoretic mobility of particles in aqueous suspensions. A range of nanoparticle sizes, materials and surface functionalities has been tested. Using pressure-driven flow through a pore, the blockade rate for 100 nm carboxylated polystyrene particles was found to be linearly proportional to both transmembrane pressure (controlled between 0 and 1.8 kPa) and particle concentration (between 7 x 10^8 and 4.5 x 10^10 mL^-1). This result can be accurately modelled using Nernst-Planck transport theory. Using only an applied potential across a pore, the blockade rates for carboxylic acid and amine coated 500 nm and 200 nm silica particles were found to correspond to changes in their mobility as a function of the solution pH. Scanning electron microscopy and confocal microscopy have been used to visualise changes in the tunable nanopore geometry in three dimensions as a function of applied mechanical strain. The pores observed were conical in shape, and changes in pore size were consistent with ionic current measurements. A zone of inelastic deformation adjacent to the pore has been identified as critical in the tuning process

    A Novel Splice-Site Variant in CACNA1F Causes a Phenotype Synonymous with Ă…land Island Eye Disease and Incomplete Congenital Stationary Night Blindness

    Background: CACNA1F-related disorders encompass progressive and non-progressive disorders, including Ă…land island eye disease and incomplete congenital stationary night blindness. These two X-linked disorders are characterized by nystagmus, color vision defect, myopia, and electroretinography (ERG) abnormalities. Ocular hypopigmentation and iris transillumination are reported only in patients with Ă…land island eye disease. Around 260 variants were reported to be associated with these two non-progressive disorders, with 19 specific to Ă…land island eye disease and 14 associated with both Ă…land island eye disease and incomplete congenital stationary night blindness. CACNA1F variants spread on the gene and further analysis are needed to reveal phenotype-genotype correlation. Case Report: A complete ocular exam and genetic testing were performed on a 13-year-old boy. A novel splice-site variant, c.4294-11C>G in intron 36 in CACNA1F, was identified at hemizygous state in the patient and at heterozygous state in his asymptomatic mother and explained the phenotype synonymous with Ă…land island eye disease and incomplete congenital stationary night blindness observed in the patient. Conclusion: We present a novel variant in the CACNA1F gene causing phenotypic and electrophysiologic findings indistinguishable from those of AIED/CSNB2A disease. This finding further expands the mutational spectrum and our knowledge of CACNA1F-related disease

    Development of a Mobile App for Self-Care Against COVID-19 Using the Analysis, Design, Development, Implementation, and Evaluation (ADDIE) Model: Methodological Study

    Background: Mobile apps have been shown to play an important role in the management, care, and prevention of infectious diseases. Thus, skills for self-care—one of the most effective ways to prevent illness—can be improved through mobile health apps. Objective: This study aimed to design, develop, and evaluate an educational mobile-based self-care app in order to help the self-prevention of COVID-19 in underdeveloped countries. We intended the app to be easy to use, quick, and inexpensive. Methods: In 2020 and 2021, we conducted a methodological study. Using the ADDIE (analysis, design, development, implementation, and evaluation) educational model, we developed a self-care management mobile app. According to the ADDIE model, an effective training and performance support tool is built through the 5 phases that comprise its name. There were 27 participants who conducted 2 evaluations of the mobile app’s usability and impact using the mobile health app usability and self-care inventory scales. The study design included pre- and posttesting. Results: An Android app called MyShield was developed. The results of pre- and posttests showed that on a scale from 0 to 5, MyShield scored a performance average of 4.17 in the physical health dimension and an average of 3.88 in the mental well-being dimension, thereby showing positive effects on self-care skills. MyShield scored highly on the “interface and satisfaction,” “ease of use,” and “usefulness” components. Conclusions: MyShield facilitates learning self-care skills at home, even during quarantine, increasing acquisition of information. Given its low development cost and the ADDIE educational design on which it is based, the app can be helpful in underdeveloped countries. Thus, low-income countries—often lacking other tools—can use the app as an effective tool for fighting COVID-19, if it becomes a standard mobile app recommended by the government

    TISs-ST: a web server to evaluate polymorphic translation initiation sites and their reflections on the secretory targets

    <p>Abstract</p> <p>Background</p> <p>The nucleotide sequence flanking the translation initiation codon (start codon context) affects the translational efficiency of eukaryotic mRNAs, and may indicate the presence of an alternative translation initiation site (TIS) to produce proteins with different properties. Multi-targeting may reflect the translational variability of these other protein forms. In this paper we present a web server that performs computations to investigate the usage of alternative translation initiation sites for the synthesis of new protein variants that might have different functions.</p> <p>Results</p> <p>An efficient web-based tool entitled TISs-ST (Translation Initiation Sites and Secretory Targets) evaluates putative translation initiation sites and indicates the prediction of a signal peptide of the protein encoded from this site. The TISs-ST web server is freely available to both academic and commercial users and can be accessed at <url>http://ipe.cbmeg.unicamp.br/pub/TISs-ST</url>.</p> <p>Conclusion</p> <p>The program can be used to evaluate alternative translation initiation site consensus with user-specified sequences, based on their composition or on many position weight matrix models. TISs-ST provides analytical and visualization tools for evaluating the periodic frequency, the consensus pattern and the total information content of a sequence data set. A search option allows for the identification of signal peptides from predicted proteins using the PrediSi software.</p

    The +4G Site in Kozak Consensus Is Not Related to the Efficiency of Translation Initiation

    The optimal context for translation initiation in mammalian species is GCCRCCaugG (where R = purine and “aug” is the initiation codon), with the -3R and +4G being particularly important. The presence of +4G has been interpreted as necessary for efficient translation initiation. Accumulated experimental and bioinformatic evidence has suggested an alternative explanation based on amino acid constraint on the second codon, i.e., amino acid Ala or Gly are needed as the second amino acid in the nascent peptide for the cleavage of the initiator Met, and the consequent overuse of Ala and Gly codons (GCN and GGN) leads to the +4G consensus. I performed a critical test of these alternative hypotheses on +4G based on 34169 human protein-coding genes and published gene expression data. The result shows that the prevalence of +4G is not related to translation initiation. Among the five G-starting codons, only alanine codons (GCN), and glycine codons (GGN) to a much smaller extent, are overrepresented at the second codon, whereas the other three codons are not overrepresented. While highly expressed genes have more +4G than lowly expressed genes, the difference is caused by GCN and GGN codons at the second codon. These results are inconsistent with +4G being needed for efficient translation initiation, but consistent with the proposal of amino acid constraint hypothesis
