    Clinical potential of necitumumab in non-small cell lung carcinoma

    Despite significant progress, new therapeutic approaches for advanced non-small cell lung cancer (NSCLC) are highly needed, particularly for the treatment of patients with squamous cell carcinoma. The epidermal growth factor receptor (EGFR) is often overexpressed in NSCLC and represents a relevant target for specific treatments. Although EGFR mutations are more frequent in non-squamous histology, the receptor itself is more often overexpressed in squamous NSCLC. Necitumumab is a human monoclonal antibody that is able to inhibit the EGFR pathway and cause antibody-dependent cell cytotoxicity. This drug has been studied in combination with first-line chemotherapy for advanced NSCLC in two Phase III trials, and a significant survival benefit was reported in squamous NSCLC (SQUIRE trial); by contrast, necitumumab did not prove itself beneficial in non-squamous histotype (INSPIRE trial). On the basis of the SQUIRE results, necitumumab was approved in combination with cisplatin and gemcitabine as a first-line treatment for advanced squamous NSCLC, both in the US and Europe, where its availability is limited to patients with EGFRexpressing tumors. The aim of this review is to describe the tolerability and the efficacy of necitumumab by searching the available published data and define its potential role in the current landscape of NSCLC treatment

    The Effect of Hot Gas in WMAP's First Year Data

    By cross-correlating templates constructed from the 2 Micron All Sky Survey (2MASS) Extended Source (XSC) catalogue with WMAP's first year data, we search for the thermal Sunyaev-Zel'dovich signature induced by hot gas in the local Universe. Assuming that galaxies trace the distribution of hot gas, we select regions on the sky with the largest projected density of galaxies. Under conservative assumptions on the amplitude of foreground residuals, we find a temperature decrement of -35 ±\pm 7 μ\muK (∼5σ\sim 5\sigma detection level, the highest reported so far) in the ∼\sim 26 square degrees of the sky containing the largest number of galaxies per solid angle. We show that most of the reported signal is caused by known galaxy clusters which, when convolved with the average beam of the WMAP W band channel, subtend a typical angular size of 20--30 arcmins. Finally, after removing from our analyses all pixels associated with known optical and X-ray galaxy clusters, we still find a tSZ decrement of -96 ±\pm 37 μ\muK in pixels subtending about ∼\sim 0.8 square degrees on the sky. Most of this signal is coming from five different cluster candidates in the Zone of Avoidance (ZoA), present in the Clusters In the ZoA (CIZA) catalogue. We found no evidence that structures less bound than clusters contribute to the tSZ signal present in the WMAP data.Comment: 10 pages, 4 figures, matches accepted version in ApJ Letter

    Detection of Anomalous Microwave Emission in the Pleiades Reflection Nebula with WMAP and the COSMOSOMAS Experiment

    We present evidence for anomalous microwave emission (AME) in the Pleiades reflection nebula, using data from the seven-year release of the Wilkinson Microwave Anisotropy Probe (WMAP) and from the COSMOSOMAS experiment. The flux integrated in a 1-degree radius around R.A.=56.24^{\circ}, Dec.=23.78^{\circ} (J2000) is 2.15 +/- 0.12 Jy at 22.8 GHz, where AME is dominant. COSMOSOMAS data show no significant emission, but allow to set upper limits of 0.94 and 1.58 Jy (99.7% C.L.) respectively at 10.9 and 14.7 GHz, which are crucial to pin down the AME spectrum at these frequencies, and to discard any other emission mechanisms which could have an important contribution to the signal detected at 22.8 GHz. We estimate the expected level of free-free emission from an extinction-corrected H-alpha template, while the thermal dust emission is characterized from infrared DIRBE data and extrapolated to microwave frequencies. When we deduct the contribution from these two components at 22.8 GHz the residual flux, associated with AME, is 2.12 +/- 0.12 Jy (17.7-sigma). The spectral energy distribution from 10 to 60 GHz can be accurately fitted with a model of electric dipole emission from small spinning dust grains distributed in two separated phases of molecular and atomic gas, respectively. The dust emissivity, calculated by correlating the 22.8 GHz data with 100-micron data, is found to be 4.36+/-0.17 muK/MJy/sr, a value that is rather low compared with typical values in dust clouds. The physical properties of the Pleiades nebula indicate that this is indeed a much less opaque object than others were AME has usually been detected. This fact, together with the broad knowledge of the stellar content of this region, provides an excellent testbed for AME characterization in physical conditions different from those generally explored up to now.Comment: Accepted for publication in ApJ. 12 pages, 8 figure

    Adoption, yield and profitability of tomato grafting technique in Vietnam

    Theme: Families, Farms, FoodThis paper assesses the impact of AVRDC’s tomato grafting approach on yield and farm profitability in Lam Dong province and Red River Delta, Vietnam. Tomato grafting is advantageous to farmers suffering from soil-borne disease and abiotic stresses. However, there is scanty information on the extent of knowledge on adoption studies of tomato grafting technology in Vietnam. Based on a farm household survey conducted in August 2012, this paper provides detailed assessment of the adoption and profitability of introducing tomato grafting in the two study areas. Results indicate a 100% (n=225) adoption in Lam Dong province, and a 48% (n=36) adoption in the Red River Delta. The use of rootstock varieties differs in both locations to address location-specific agronomic challenges: tomato variety ‘Vimina’ (or HW7996) to address bacterial wilt (BW) problem, and eggplant EG203 variety in the Red River Delta to address both BW and waterlogging problem. Estimates from a Cobb-Douglas production function show that tomato grafting increases yield by 30% based. Marketable yield of grafted tomato was significantly larger (71.3 t/ha in Lam Dong Province and 75.0 t/ha in Red River Delta) than nongrafted (48.0 t/ha in Red River Delta). The benefit-cost ratio of grafted tomato production was higher compared to non-grafted due to increased yield and higher premium price. Nonetheless, further validation studies are required, considering the relatively small sample size in the Red River Delta and the high variability of some parameters.C.A. Genova, P.     Text-based Editing of Talking-head Video

    Editing talking-head video to change the speech content or to remove filler words is challenging. We propose a novel method to edit talking-head video based on its transcript to produce a realistic output video in which the dialogue of the speaker has been modified, while maintaining a seamless audio-visual flow (i.e. no jump cuts). Our method automatically annotates an input talking-head video with phonemes, visemes, 3D face pose and geometry, reflectance, expression and scene illumination per frame. To edit a video, the user has to only edit the transcript, and an optimization strategy then chooses segments of the input corpus as base material. The annotated parameters corresponding to the selected segments are seamlessly stitched together and used to produce an intermediate video representation in which the lower half of the face is rendered with a parametric face model. Finally, a recurrent video generation network transforms this representation to a photorealistic video that matches the edited transcript. We demonstrate a large variety of edits, such as the addition, removal, and alteration of words, as well as convincing language translation and full sentence synthesis

    We have been recently faced with the problem of cross--identifying stars recorded in historical catalogues with those extracted from recent fully digitized surveys (such as DENIS and 2MASS). Positions mentioned in the old catalogues are frequently of poor precision, but are generally accompanied by finding charts where the interesting objects are flagged. Those finding charts are sometimes our only link with the accumulated knowledge of past literature. While checking the identification of some of these objects in several catalogues, we had the surprise to discover a number of discrepancies in recent works.The main reason for these discrepancies was generally the blind application of the smallest difference in position as the criterion to identify sources from one historical catalogue to those in more recent surveys. In this paper we give examples of such misidentifications, and show how we were able to find and correct them.We present modern procedures to discover and solve cross--identification problems, such as loading digitized images of the sky through the Aladin service at CDS, and overlaying entries from historical catalogues and modern surveys. We conclude that the use of good finding charts still remains the ultimate (though time--consuming) tool to ascertain cross--identifications in difficult cases.Comment: 4 pages, 1 figure, accepted by A&

    Global gene expression analysis provides insight into local adaptation to geothermal streams in tadpoles of the Andean toad Rhinella spinulosa

    Indexación: Web of Science; Scopus.The anuran Rhinella spinulosa is distributed along the Andes Range at altitudes that undergo wide daily and seasonal variation in temperature. One of the populations inhabits geothermal streams, a stable environment that influences life history traits such as the timing of metamorphosis. To investigate whether this population has undergone local adaptation to this unique habitat, we carried out transcriptome analyses in animals from two localities in two developmental stages (prometamorphic and metamorphic) and exposed them to two temperatures (20 and 25 degrees C). RNA-Seq, de novo assembly and annotation defined a transcriptome revealing 194,469 high quality SNPs, with 1,507 genes under positive selection. Comparisons among the experimental conditions yielded 1,593 differentially expressed genes. A bioinformatics search for candidates revealed a total of 70 genes that are highly likely to be implicated in the adaptive response of the population living in a stable environment, compared to those living in an environment with variable temperatures. Most importantly, the population inhabiting the geothermal environment showed decreased transcriptional plasticity and reduced genetic variation compared to its counterpart from the non-stable environment.
