    Regulation of nitrogen uptake and assimilation: Effects of nitrogen source, root-zone pH, and aerial CO2 concentration on growth and productivity of soybeans

    An important feature of controlled-environment crop production systems such as those to be used for life support of crews during space exploration is the efficient utilization of nitrogen supplies. Making decisions about the best sources of these supplies requires research into the relationship between nitrogen source and the physiological processes which regulate vegetative and reproductive plant growth. Work done in four areas within this research objective is reported: (1) experiments on the effects of root-zone pH on preferential utilization of NO3(-) versus NH4(+) nitrogen; (2) investigation of processes at the whole-plant level that regulate nitrogen uptake; (3) studies of the effects of atmospheric CO2 and NO3(-) supply on the growth of soybeans; and (4) examination of the role of NO3(-) uptake in enhancement of root respiration

    Simulation model for plant growth in controlled environment systems

    The role of the mathematical model is to relate the individual processes to environmental conditions and the behavior of the whole plant. Using the controlled-environment facilities of the phytotron at North Carolina State University for experimentation at the whole-plant level and methods for handling complex models, researchers developed a plant growth model to describe the relationships between hierarchial levels of the crop production system. The fundamental processes that are considered are: (1) interception of photosynthetically active radiation by leaves, (2) absorption of photosynthetically active radiation, (3) photosynthetic transformation of absorbed radiation into chemical energy of carbon bonding in solube carbohydrates in the leaves, (4) translocation between carbohydrate pools in leaves, stems, and roots, (5) flow of energy from carbohydrate pools for respiration, (6) flow from carbohydrate pools for growth, and (7) aging of tissues. These processes are described at the level of organ structure and of elementary function processes. The driving variables of incident photosynthetically active radiation and ambient temperature as inputs pertain to characterization at the whole-plant level. The output of the model is accumulated dry matter partitioned among leaves, stems, and roots; thus, the elementary processes clearly operate under the constraints of the plant structure which is itself the output of the model

    Nitrogen uptake and utilization by intact plants

    The results of experiments support the proposed conceptual model that relates nitrogen uptake activity by plants as a balanced interdependence between the carbon-supplying function of the shoot and the nitrogen-supplying function of the roots. The data are being used to modify a dynamic simulation of plant growth, which presently describes carbon flows through the plant, to describe nitrogen uptake and assimilation within the plant system. Although several models have been proposed to predict nitrogen uptake and partitioning, they emphasize root characteristics affecting nutrient uptake and relay on empirical methods to describe the relationship between nitrogen and carbon flows within the plant. Researchers, on the other hand, propose to continue to attempt a mechanistic solution in which the effects of environment on nitrogen (as well as carbon) assimilation are incorporated through their direct effects on photosynthesis, respiration, and aging processes

    A weighting method to improve habitat association analysis: tested on British carabids

    Analysis of species’ habitat associations is important for biodiversity conservation and spatial ecology. The original phi coefficient of association is a simple method that gives both positive and negative associations of individual species with habitats. The method originates in assessing the association of plant species with habitats, sampled by quadrats. Using this method for mobile animals creates problems as records often have imprecise locations, and would require either using only records related to a single habitat or arbitrarily choosing a single habitat to assign. We propose and test a new weighted version of the index that retains more records, which improves association estimates and allows assessment of more species. It weights habitats that lie within the area covered by the species record with their certainty level, in our case study, the proportion of the grid cell covered by that habitat. We used carabid beetle data from the National Biodiversity Network atlas and CEH Land Cover Map 2015 across Great Britain to compare the original method with the weighted version. We used presence‐only data, assigning species absences using a threshold based on the number of other species found at a location, and conducted a sensitivity analysis of this threshold. Qualitative descriptions of habitat associations were used as independent validation data. The weighted index allowed the analysis of 52 additional species (19% more) and gave results with as few as 50 records. For the species we could analyse using both indices, the weighted index explained 70% of the qualitative validation data compared to 68% for the original, indicating no accuracy loss. The weighted phi coefficient of association provides an improved method for habitat analysis giving information on preferred and avoided habitats for mobile species that have limited records, and can be used in modelling and analysis that directs conservation policy and practice

    Treatment of malignant tumors of the skull base with multi-session radiosurgery

    <p>Abstract</p> <p>Objective</p> <p>Malignant tumors that involve the skull base pose significant challenges to the clinician because of the proximity of critical neurovascular structures and limited effectiveness of surgical resection without major morbidity. The purpose of this study was to evaluate the efficacy and safety of multi-session radiosurgery in patients with malignancies of the skull base.</p> <p>Methods</p> <p>Clinical and radiographic data for 37 patients treated with image-guided, multi-session radiosurgery between January 2002 and December 2007 were reviewed retrospectively. Lesions were classified according to involvement with the bones of the base of the skull and proximity to the cranial nerves.</p> <p>Results</p> <p>Our cohort consisted of 37 patients. Six patients with follow-up periods less than four weeks were eliminated from statistical consideration, thus leaving the data from 31 patients to be analyzed. The median follow-up was 37 weeks. Ten patients (32%) were alive at the end of the follow-up period. At last follow-up, or the time of death from systemic disease, tumor regression or stable local disease was observed in 23 lesions, representing an overall tumor control rate of 74%. For the remainder of lesions, the median time to progression was 24 weeks. The median progression-free survival was 230 weeks. The median overall survival was 39 weeks. In the absence of tumor progression, there were no cranial nerve, brainstem or vascular complications referable specifically to CyberKnife<sup>® </sup>radiosurgery.</p> <p>Conclusion</p> <p>Our experience suggests that multi-session radiosurgery for the treatment of malignant skull base tumors is comparable to other radiosurgical techniques in progression-free survival, local tumor control, and adverse effects.</p

    Differences between <i>Trypanosoma brucei gambiense</i> groups 1 and 2 in their resistance to killing by Trypanolytic factor 1

    &lt;p&gt;&lt;b&gt;Background:&lt;/b&gt; The three sub-species of &lt;i&gt;Trypanosoma brucei&lt;/i&gt; are important pathogens of sub-Saharan Africa. &lt;i&gt;T. b. brucei&lt;/i&gt; is unable to infect humans due to sensitivity to trypanosome lytic factors (TLF) 1 and 2 found in human serum. &lt;i&gt;T. b. rhodesiense&lt;/i&gt; and &lt;i&gt;T. b. gambiense&lt;/i&gt; are able to resist lysis by TLF. There are two distinct sub-groups of &lt;i&gt;T. b. gambiense&lt;/i&gt; that differ genetically and by human serum resistance phenotypes. Group 1 &lt;i&gt;T. b. gambiense&lt;/i&gt; have an invariant phenotype whereas group 2 show variable resistance. Previous data indicated that group 1 &lt;i&gt;T. b. gambiense&lt;/i&gt; are resistant to TLF-1 due in-part to reduced uptake of TLF-1 mediated by reduced expression of the TLF-1 receptor (the haptoglobin-hemoglobin receptor (&lt;i&gt;HpHbR&lt;/i&gt;)) gene. Here we investigate if this is also true in group 2 parasites.&lt;/p&gt; &lt;p&gt;&lt;b&gt;Methodology:&lt;/b&gt; Isogenic resistant and sensitive group 2 &lt;i&gt;T. b. gambiense&lt;/i&gt; were derived and compared to other T. brucei parasites. Both resistant and sensitive lines express the &lt;i&gt;HpHbR&lt;/i&gt; gene at similar levels and internalized fluorescently labeled TLF-1 similar fashion to &lt;i&gt;T. b. brucei&lt;/i&gt;. Both resistant and sensitive group 2, as well as group 1 &lt;i&gt;T. b. gambiense&lt;/i&gt;, internalize recombinant APOL1, but only sensitive group 2 parasites are lysed.&lt;/p&gt; &lt;p&gt;&lt;b&gt;Conclusions:&lt;/b&gt; Our data indicate that, despite group 1 &lt;i&gt;T. b. gambiense&lt;/i&gt; avoiding TLF-1, it is resistant to the main lytic component, APOL1. Similarly group 2 &lt;i&gt;T. b. gambiense&lt;/i&gt; is innately resistant to APOL1, which could be based on the same mechanism. However, group 2 &lt;i&gt;T. b. gambiense&lt;/i&gt; variably displays this phenotype and expression does not appear to correlate with a change in expression site or expression of &lt;i&gt;HpHbR&lt;/i&gt;. Thus there are differences in the mechanism of human serum resistance between &lt;i&gt;T. b. gambiense&lt;/i&gt; groups 1 and 2.&lt;/p&gt

    A CATT Negative Result after Treatment for Human African Trypanosomiasis Is No Indication for Cure

    The 2 year follow-up period required after treatment of human African trypanosomiasis (HAT) patients is a major challenge for patients and control programmes alike. The patient should return every 6 months for lumbar puncture and cerebrospinal fluid examination since, so far, no markers for cure have been identified in blood. The Card Agglutination Test for Trypanosomiasis (CATT) is a simple, rapid test for trypanosome-specific antibody detection in blood that is extensively used in endemic areas to screen for HAT. We examined the value of a normalising CATT as a marker for treatment outcome. We observed that CATT titres decreased after treatment both in patients who experienced treatment failure as well as in cured patients. We conclude that CATT, though a good screening test, is unreliable for monitoring treatment outcome. We also showed that the sensitivity of CATT in relapse cases was as low as 78%, and as a consequence some relapse cases might be missed in screening programs if they have no clinical signs yet
