The PDZ domain of the SpoIVB serine peptidase facilitates multiple functions
During spore formation in Bacillus subtilis, the SpoIVB protein is a critical component of the sigma (K) regulatory checkpoint. SpoIVB has been shown to be a serine peptidase that is synthesized in the spore chamber and which self-cleaves, releasing active forms. These forms can signal proteolytic processing of the transcription factor sigma (K) in the outer mother cell chamber of the sporulating cell. This forms the basis of the sigma (K) checkpoint and ensures accurate sigma (K)-controlled gene expression. SpoIVB has also been shown to activate a second distinct process, termed the second function, which is essential for the formation of heat-resistant spores. In addition to the serine peptidase domain, SpoIVB contains a PDZ domain. We have altered a number of conserved residues in the PDZ domain by site-directed mutagenesis and assayed the sporulation phenotype and signaling properties of mutant SpoIVB proteins. Our work has revealed that the SpoIVB PDZ domain could be used for up to four distinct processes, (i) targeting of itself for trans proteolysis, (11) binding to the protease inhibitor BofC, (iii) signaling of pro-sigma (K) processing, and (iv) signaling of the second function of SpoIVB
Timing of Cattle Graze as a Management Tool for Plant and Invertebrate Communities in the Centennial Valley, Montana
Grazing is an important natural disturbance in the western US. While many studies have focused on the effects of grazing intensity and frequency, little is known about the importance of graze timing and how it can affect plant and invertebrate community structure and productivity. Timing may be a critical factor for promoting ecological heterogeneity, biological diversity, and abundance of food resources for native birds. Our primary goal is to understand how timing of graze and climate variability interact to influence plant and invertebrate communities throughout the growing season in wet meadows and grasslands in the Centennial Valley of southwest Montana. In 2011, control (no graze), early graze (beginning June 15), and traditional late graze (beginning July 15) treatments were established and replicated in a heterogeneous, irrigated, wet meadow habitat. From June-September, plant and invertebrate communities were quantitatively sampled to examine dynamics of biomass and diversity. In 2012, a similar experimental design will be implemented in a less-disturbed and drier native grassland. A new component will examine interactions between climate variability and timing by incorporating a water-addition treatment to simulate the extra moisture resulting from a wet year. We predict that increased climate variability will likely impact wet meadow and grassland habitat plant and insect phenology, with timing of graze becoming particularly critical during dry years. Location, Location, Location Should Be Environment, Environment, Environment : A Market-Based Tool to Simplify Environmental Considerations in Residential Real Estate
The most important rule of real estate (location, location, location) should be upgraded to the three E’s: environment, environment, environment. What we value in real estate is the natural and human environment of a site and its structures. A home is typically an American’s most significant asset; thus, environmental issues should be of interest, primarily because the effects of environmental degradation can cause devaluation while simultaneously imposing substantial expenses (such as cleanup, health care, and relocation) on the population. The real costs of ignoring the environment are life-threatening health and safety issues, including lung damage and cancer resulting from radon exposure, (which the EPA estimates kills 20,000 people per year), and indoor air pollution (from Volatile Organic Compounds (VOCs), for example), the effects of mold and polluted water, and heart and lung conditions related to poor air quality, result in thousands of premature deaths each year.
Environmental issues include both anthropogenic sources and naturally occurring phenomena. The problem is that, for some buyers, sellers and professionals, obtaining accurate data is difficult. Many know there are issues, but they are unable to get straight-forward, manageable information. Some do not want to know of the issues, and others are overwhelmed. Complicating the matter for everyone is the reality that the laws related to disclosure and duties to prevent or mitigate harm vary significantly by jurisdiction, creating unpredictable rights and duties that range from caveat emptor to duties of reasonable inquiry. Consistency between jurisdictions is of greater importance than ever because of the mobility of the population. Even with guidance and reports from the EPA, the tools available to the majority of individuals seeking to make this most significant purchase, the real estate market does not address the health and safety risks caused by environmental degradation.
Given this deficit in information, we propose a voluntary checklist to alert consumers, owners, and professionals of environmental issues that can impose significant costs for health care, remediation, and property devaluation. Knowledge of the issues should reduce disputes, and, over time, consumers may demand properties that are safer, with economic variables that are better quantified. That in turn should encourage sellers, builders and producers to satisfy the expectations of the consuming public with greener and more sustainable housing
Detecting the direction of a signal on high-dimensional spheres: Non-null and Le Cam optimality results
We consider one of the most important problems in directional statistics,
namely the problem of testing the null hypothesis that the spike direction
of a Fisher-von Mises-Langevin distribution on the -dimensional
unit hypersphere is equal to a given direction . After a reduction
through invariance arguments, we derive local asymptotic normality (LAN)
results in a general high-dimensional framework where the dimension goes
to infinity at an arbitrary rate with the sample size , and where the
concentration behaves in a completely free way with , which
offers a spectrum of problems ranging from arbitrarily easy to arbitrarily
challenging ones. We identify various asymptotic regimes, depending on the
convergence/divergence properties of , that yield different
contiguity rates and different limiting experiments. In each regime, we derive
Le Cam optimal tests under specified and we compute, from the Le Cam
third lemma, asymptotic powers of the classical Watson test under contiguous
alternatives. We further establish LAN results with respect to both spike
direction and concentration, which allows us to discuss optimality also under
unspecified . To investigate the non-null behavior of the Watson test
outside the parametric framework above, we derive its local asymptotic powers
through martingale CLTs in the broader, semiparametric, model of rotationally
symmetric distributions. A Monte Carlo study shows that the finite-sample
behaviors of the various tests remarkably agree with our asymptotic results.Comment: 47 pages, 4 figure
Influences of Neural Pathway Integrity on Children's Response to Reading Instruction
As the education field moves toward using responsiveness to intervention to identify students with disabilities, an important question is the degree to which this classification can be connected to a student's neurobiological characteristics. A few functional neuroimaging studies have reported a relationship between activation and response to instruction; however, whether a similar correlation exists with white matter (WM) is not clear. To investigate this issue, we acquired high angular resolution diffusion images from a group of first grade children who differed in their levels of responsiveness to a year-long reading intervention. Using probabilistic tractography, we calculated the strength of WM connections among nine cortical regions of interest and correlated these estimates with participants’ scores on four standardized reading measures. We found eight significant correlations, four of which were connections between the insular cortex and angular gyrus. In each of the correlations, a relationship with children's response to intervention was evident
Using Co-operative Inquiry to embed Education for Sustainable Development (ESD) across all programmes offered by Plymouth Institute of Education (PIoE)
Our aim is to embed Education for Sustainable Development across all of our courses, ensuring that all students benefit from an education where social, economic and environmental sustainability is woven into the very fabric of their learning experience, thereby producing graduates who take this learning forward and embed it in their own teaching. To achieve this, the aims of the projects were:
To embed elements of ESD in all PIoE programmes utilising a co-operative inquiry approach (Heron and Reason 2001)
• To critically evaluate co-operative inquiry (CI) as an effective mechanism for the implementation of ESD in HE.PedRI
Human recognition based on gait poses
This paper introduces a new approach for gait analysis based on the Gait Energy Image (GEI). The main idea is to segment the gait cycle into some biomechanical poses, and to compute a particular GEI for eachpose. Pose-based GEIs can better represent body parts and dynamics descriptors with respect to the usually blurred depiction provided by a general GEI. Gait classification is carried out by fusing separatedpose-based decisions. Education for Sustainable Development: Towards the Sustainable University
We planned this conference in anticipation of the end of the UN Decade of Education for Sustainable Development (DESD), and the start of the next phase for those involved in ESD here and internationally. At Plymouth University, 2015 marks ten year anniversary since cross-‐institutional work on sustainability and sustainability education was spearheaded by the founding of the Centre for Sustainable Futures (CSF). Coincidentally, 2015 also marks a ten years since the influential HEFCE policy document ‘Sustainable Development in Higher Education’ was released. Holding the conference in January – named after the Roman god of doorways, of endings and beginnings – we sought to look at some of what has been achieved in sustainability education to date and explore its prospects as we move forward. Following an enthusiastic response to the call for abstracts, the conference featured a diverse range of research papers, posters, and roundtable presentations from academics and practitioners across the UK and beyond. The conference was arranged around three overarching themes: ESD Pedagogy: Criticality, Creativity, and Collaboration What are the teaching and learning processes that enable students to develop their own capacity to think critically and creatively in the face of global sustainability challenges and, secondly, to act collaboratively in ways that pursue more hopeful and sustainable futures? Innovative Learning Spaces for ESD What are the physical environments that provide opportunities for new forms of sustainability education to flourish? What lies beyond the lecture hall that is conducive to student learning through inquiry-‐based, active, participatory, interdisciplinary and experiential methods? Towards the Sustainable University What are effective approaches for leading institutional change, organisational learning, and staff CPD towards sustainability? This publication focuses on the last theme – Towards the Sustainable University. Extracting collective trends from Twitter using social-based data mining

The interactions amongst users provide information of their preferences and relationships. This information can be used to measure the influence of ideas, or opinions, and how they are spread within the Network. Currently, one of the most relevant and popular Social Network is Twitter. This Social Network was created to share comments and opinions. The information provided by users is specially useful in different fields and research areas such as marketing. This data is presented as short text strings containing different ideas expressed by real people. With this representation, different Data Mining and Text Mining techniques (such as classification and clustering) might be used for knowledge extraction trying to distinguish the meaning of the opinions.
