    Physiological response of Cistus salviifolius L. to high arsenic concentrations

    Arsenic is a trace element found in the environment which can be particularly toxic to living organisms. However, some plant species such as those of the genus Cistus are able to grow in soils with high As concentrations and could be used in the sustainable rehabilitation of mining areas 19 through phytostabilization. In this work, the growth and the physiological response of Cistus salviifolius L. to As induced oxidative stress at several concentrations (reaching 30 mg L-1) in an hydroponic system were evaluated for 30 days. Several growth parameters, chlorophyll content, chemical composition, one indicator of oxidative stress (H2O2) and two of the major anti-oxidative metabolites (ascorbate and glutathione) were analyzed. The toxic effect of As was better perceived in the plants submitted to treatments with concentrations of 20 and 30 mg As L-1. Plants subjected to these treatments had higher concentration of As in roots and shoots. The concentrations of Ca, Mg, K and Fe in the plants, as well as a large part of the evaluated growth parameters were also affected. Arsenic did not interfere with the ability of the plant to perform photosynthesis, as there were no significant differences in the contents of chlorophyll a, b and total between the different treatments. Plants from all treatments accumulated higher amount of As in roots than in shoots, and it was also in the roots that the concentrations of H2O2, AsA and GSH were higher.     Influence of different finishing treatments over mechanical and thermal properties of bed linen

    This work aims to assess the relationship between the mechanical and the thermal properties of summer bed linen using different softener formulations in home textiles finishing. Objectively, we intend to study the effect of different softeners and their concentrations, based on non-ionic polyethylene dispersions and a cationic silicone softener micro-emulsion on textile properties. The research shown that thermal related properties are influenced by polyethylene softeners, while the silicone softener influences the mechanical behavior of the tested samples. Finally, selected softened samples of bed linen were evaluated by a dry thermal manikin to validate some conclusions.(undefined

    The perception of tactile feeling and corresponding textile attributes worldwide

    The textile industry has been exploring sensory analysis as means to evaluate different products as a strategy to improve product comfort. The sensory analysis uses a set of attributes standardized for sensory evaluation: the lexicons. This paper compares three lexicons developed to assess the sensory comfort of textile fabrics, by researchers from France, Portugal and Brazil. Quantitative Descriptive Analysis was conducted for the development of these lexicons.     Antibacterial activity of phenyl isothiocyanate on Escherichia coli and Staphylococcus aureus

    The present study has been aimed to assess the antibacterial effects of the glucosinolate hydrolysis product phenyl isothiocyanate (PITC) against Escherichia coli and Staphylococcus aureus. Aspects on the antibacterial mode of action of PITC have also been characterized, such as the changes on surface physicochemical characteristics and membrane damage. The minimum inhibitory concentration of PITC was 1000 µg/mL, for both bacteria. The antimicrobial potential of PITC was compared with selected antibiotics (ciprofloxacin, erythromycin, streptomycin, tetracycline and spectinomycin),that reported a moderate effect. The combination of PITC with ciprofloxacin and erythromycin against S.aureus exhibited a good antimicrobial efficacy, due to an additive effect (the diameter of inhibition zones increased from 30 to 40 mm for ciprofloxacin and almost the double for erythromycin). The other combinations reported unsatisfactory results against both bacteria. The study of the physiological changes induced by PITC action demonstrated the interaction between the electrophilic compound and the bacterial cells at several points that causes changes in membrane properties (decreases negative surface charge, increases surface hydrophilicity and electron donor characteristics).     Cost and efficiency analysis of commercial softeners in the sewability behavior of cotton fabrics

    This paper reports a comparative case study on the use of different softening products for bed linen fabrics, specifically regarding the sewability of the fabrics. The market offers a wide variety of commercial formulations of softeners for this purpose, but the composition and price varies considerably. This work was aimed to assess the relationship between the cost and effectiveness of different softener formulations in home textiles finishing. Objectively, the effect of different softeners and their concentrations on sewability of the fabrics was studied. Non-ionic polyethylene dispersions and a cationic silicone softener micro-emulsion in different concentrations and combinations were considered in this investigation.     SPATIAL AND TEMPORALVARIATION OF DISSOLVED INORGANIC NUTRIENTS, AND CHLOROPHYLL-α IN A TROPICAL ESTUARY IN NORTHEASTERN BRAZIL: DYNAMICS OF NUTRIENT REMOVAL

    Monthly sampling campaigns were carried out between February 2010 and January 2011 to evaluate the spatial and temporal distribution of nutrients (ammonium, nitrite, nitrate, dissolved organic nitrogen, phosphate, dissolved organic phosphorus and silicate) and chlorophyll-α along a salinity gradient in the tropical Cachoeira River estuary, subject to the untreated effluents of a sewage treatment plant (STP). During the study period the lowest and highest river discharge occurred in February and April 2010, respectively. High river outflow promoted increased concentrations of inorganic nitrogen and silicate but did not affect the concentration of phosphate. Based on the chlorophyll-α concentration the estuary may be classified as eutrophic / hypereutrophic in its inner portion and mesotrophic in the lower region. The inner portion is more affected by the nutrient load carried out by the river and STP, while dilution by seawater contributed to the reduction of the nutrient concentrations in the lower reaches of the estuary. The results indicate that nutrient uptake by the phytoplankton is the most effective dissolved inorganic nutrient removal processes, especially for phosphate. Mixing diagrams suggest that the coupling of nitrification and denitrification processes is also responsible for the elimination of nitrogen from this ecosystem.Campanhas de amostragens mensais foram realizadas entre fevereiro de 2010 e janeiro de 2011 para avaliar a distribuição espacial e temporal de nutrientes (amônia, nitrito, nitrato, nitrogênio orgânico dissolvido, fosfato, fósforo orgânico dissolvido e silicato) e clorofila-α, ao longo do gradiente de salinidade no estuário tropical do Rio Cachoeira. Este estuário é sujeito aos efluentes de esgotos não tratados de uma estação de tratamento de esgoto (ETE). No período estudado a maior e menor vazão do rio ocorreram em fevereiro e abril de 2010, respectivamente. A alta vazão do rio promoveu aumento das concentrações de nitrogênio inorgânico e silicato, mas não afetou as concentrações de fosfato. Baseado nas concentrações de clorofila-α, o estuário pode ser classificado como eutrófico/hipereutrófico na porção interna e mesotrófico na região externa. A porção interna é mais afetada pela carga de nutrientes do rio e da ETE, enquanto a diluição pela água marinha contribuiu para diminuir as concentrações de nutrientes na porção externa. Os resultados indicam que a absorção de nutrientes pelo fitoplâncton é o processo mais eficiente na remoção desses nutrientes, especialmente do fosfato. No entanto, os diagramas de mistura sugerem que a nitrificação e denitrificação acopladas no rio também são responsáveis pela eliminação do nitrogênio do ecossistema

    Influence of Seed Source and Soil Contamination on Ecophysiological Responses of Lavandula pedunculata in Rehabilitation of Mining Area

    Mining activities have turned many areas of the Iberian Pyrite Belt (IPB) into extreme environments with high concentrations of metal(loid)s. These harsh conditions can inhibit or reduce the colonization and/or development of most vegetation. However, some species or populations have developed ecophysiological responses to tolerate stress factors and contaminated soils. The main objectives of this study are: (i) to assess the differences in germination, growth, development and physiological behaviour against oxidative stress caused by metal(loid)s in Lavandula pedunculata (Mill.) Cav. from two different origins (a contaminated area in São Domingos mine, SE of Portugal and an uncontaminated area from Serra do Caldeirão, S of Portugal) under controlled conditions; and (ii) to assess whether it is possible to use this species for the rehabilitation of mine areas of the IPB. After germination, seedlings from São Domingos (LC) and Caldeirão (L) were planted in pots with a contaminated soil developed on gossan (CS) and in pots with an uncontaminated soil (US) under controlled conditions. Multielemental concentrations were determined in soils (total and available fractions) and plants (shoots and roots). Germination rate, shoot height, dry biomass and leaf area were determined, and pigments, glutathione, ascorbate and H2O2 contents were measured in plant shoots. Total concentrations of As, Cr, Cu, Pb and Sb in CS, and As in US exceed the intervention and maximum limits for ecosystem protection and human health. The main results showed that L. pedunculata, regardless of the seed origin, activated defence mechanisms against oxidative stress caused by high concentrations of metal(loid)s. Plants grown from seeds of both origins increased the production of AsA to preserve its reduction levels and kept the contents of GSH stable to maintain the cell’s redox state. Plants grown from seeds collected in non-contaminated areas showed a high capacity for adaptation to extreme conditions. This species showed a greater growth capacity when seeds from a contaminated area were sown in uncontaminated soils.     Persister cells in a biofilm treated with a biocide

    This study investigated the physiology and behaviour following treatment with ortho-phthalaldehyde (OPA), of Pseudomonas fluorescens in both the planktonic and sessile states. Steady-state biofilms and planktonic cells were collected from a bioreactor and their extracellular polymeric substances (EPS) were extracted using a method that did not destroy the cells. Cell structure and physiology after EPS extraction were compared in terms of respiratory activity, morphology, cell protein and polysaccharide content, and expression of the outer membrane proteins (OMP). Significant differences were found between the physiological parameters analysed. Planktonic cells were more metabolically active, and contained greater amounts of proteins and polysaccharides than biofilm cells. Moreover, biofilm formation promoted the expression of distinct OMP. Additional experiments were performed with cells after EPS extraction in order to compare the susceptibility of planktonic and biofilm cells to OPA. Cells were completely inactivated after exposure to the biocide (minimum bactericidal concentration, MBC ¼ 0.55 + 0.20 mM for planktonic cells; MBC ¼ 1.7 + 0.30 mM for biofilm cells). After treatment, the potential of inactivated cells to recover from antimicrobial exposure was evaluated over time. Planktonic cells remained inactive over 48 h while cells from biofilms recovered 24 h after exposure to OPA, and the number of viable and culturable cells increased over time. The MBC of the recovered biofilm cells after a second exposure to OPA was 0.58 + 0.40 mM, a concentration similar to the MBC of planktonic cells.     Selective flexibility of side-chain residues improves VEGFR-2 docking score using autodock vina

    Selective side-chain residue flexibility is an option available on AutoDock Vina docking software. This approach is promising as it attempts to provide a more realistic ligand-protein interaction environment, without an unmanageable increase in computer processing time. However, studies validating this approach are still scarce. VEGFR-2 (vascular endothelial growth factor receptor 2), a known protein target for anti-angiogenic agents, was used in this study. Four residues present in the VEGFR-2 kinase site were selected and made flexible: Lys866, Glu885, Cys917 and Asp1044. The docking scores for all possible combinations of flexible residues were compared to the docking scores using a rigid conformation. The best overall docking scores were obtained using the Glu883 flexible conformation, with pearson and spearman rank correlation values of 0.568 and 0.543, respectively, and a 51% increase in computer processing time. Using different VEGFR-2 X-ray structures a similar trend was observed with Glu885 flexible conformation presenting the best scores. This study demonstrates that careful use of selective side-chain residue flexibility can improve AutoDock Vina docking score accuracy, without a significant increase in computer processing time.
