    Effects on Berry Shrinkage in Vitis vinifera. L cv. ‘Merlot’ From Changes in Canopy/Root Ratio: A Preliminary Approach

    A trial was conducted to find a possible relationship between the canopy/root ratio and the incidence and severity of premature berry shrinkage, and to propose an alternative to avoid this phenomenon in ‘Merlot’ grapevines. The ratio was changed by cutting foliage at a certain height 15 days before véraison, and by delaying the removal of trunk shoots. Treatments were the control (T1), 50% foliage area of control (T2), 75% foliage area of control (T3), and delayed trunk shoot removal (T4). Foliage area and the canopy/root ratio were lower in the T2 and T3 treatments. T4 was ineffective in changing the parameters. The incidence of berry shrinkage was lower for the T2 and T3 treatments, with the percentage of affected plants dropping from the 52% of the control to 22.9% and 31.3% for T2 and T3 respectively, and from 52.4% of the affected bunches to 16.6% and 21.2% for the same treatments respectively. The percentage of affected bunches falling into the range of moderate to severe damage fell from the 24% of the control to 5.2% and 3.9% for T2 and T3 respectively. Therefore, it is possible to avoid the incidence and severity of berry shrinkage by decreasing the canopy/root ratio in ‘Merlot’ grapevines

    Evidence for Gender-Specific Transcriptional Profiles of Nigral Dopamine Neurons in Parkinson Disease

    Epidemiological data suggest that the male gender is one of the risks factors for the development of Parkinson Disease (PD). Also, differences in the clinical manifestation and the course of PD have been observed between males and females. However, little is known about the molecular aspects underlying gender-specificity in PD. To address this issue, we determined the gene expression profiles of male and female dopamine (DA) neurons in sporadic PD.We analyzed Affymetrix-based microarrays on laser microdissected DA neurons from postmortem brains of sporadic PD patients and age-matched controls across genders. Pathway enrichment demonstrated that major cellular pathways involved in PD pathogenesis showed different patterns of deregulation between males and females with more prominent downregulation of genes related to oxidative phosphorylation, apoptosis, synaptic transmission and transmission of nerve impulse in the male population. In addition, we found upregulation of gene products for metabolic processes and mitochondrial energy consumption in the age-matched male control neurons. On the single cell level, selected data validation using quantitative Real-Time (qRT)-PCR was consistent with microarray raw data and supported some of the observations from data analysis.On the molecular level, our results provide evidence that the expression profiles of aged normal and PD midbrain DA neurons are gender-specific. The observed differences in the expression profiles suggest a disease bias of the male gender, which could be in concordance with clinical observations that the male gender represents a risk factor for sporadic PD. Validation of gene expression by qRT-PCR supported the microarray results, but also pointed to several caveats involved in data interpretation

    Spatiotemporal Compound Wavelet Matrix Framework for Multiscale/Multiphysics Reactor Simulation: Case Study of a Heterogeneous Reaction/Diffusion System

    We present a mathematical method for efficiently compounding information from different models of species diffusion from a chemically reactive boundary. The proposed method is intended to serve as a key component of a multiscale/ multiphysics framework for heterogeneous chemically reacting processes. An essential feature of the method is the merging of wavelet representations of the different models and their corresponding time and length scales. Up-and-downscaling of the information between the scales is accomplished by application of a compounding wavelet operator, which is assembled by establishing limited overlap in scales between the models. We show that the computational efficiency gain and potential error associated with the method depend on the extent of scale overlap and wavelet filtering used.     Liposome Co-sedimentation and Co-flotation Assays to Study Lipid-Protein Interactions

    A large proportion of proteins are expected to interact with cellular membranes to carry out their physiological functions in processes such as membrane transport, morphogenesis, cytoskeletal organization, and signal transduction. The recruitment of proteins at the membrane-cytoplasm interface and their activities are precisely regulated by phosphoinositides, which are negatively charged phospholipids found on the cytoplasmic leaflet of cellular membranes and play critical roles in membrane homeostasis and cellular signaling. Thus, it is important to reveal which proteins interact with phosphoinositides and to elucidate the underlying mechanisms. Here, we present two standard in vitro methods, liposome co-sedimentation and co-flotation assays, to study lipid-protein interactions. Liposomes can mimic various biological membranes in these assays because their lipid compositions and concentrations can be varied. Thus, in addition to mechanisms of lipid-protein interactions, these methods provide information on the possible specificities of proteins toward certain lipids such as specific phosphoinositide species and can hence shed light on the roles of membrane interactions on the functions of membrane-associated proteins.Peer reviewe

    Morphology of two dimensional fracture surface

    We consider the morphology of two dimensional cracks observed in experimental results obtained from paper samples and compare these results with the numerical simulations of the random fuse model (RFM). We demonstrate that the data obey multiscaling at small scales but cross over to self-affine scaling at larger scales. Next, we show that the roughness exponent of the random fuse model is recovered by a simpler model that produces a connected crack, while a directed crack yields a different result, close to a random walk.     Widespread sex differences in gene expression and splicing in the adult human brain

    There is strong evidence to show that men and women differ in terms of neurodevelopment, neurochemistry and susceptibility to neurodegenerative and neuropsychiatric disease. The molecular basis of these differences remains unclear. Progress in this field has been hampered by the lack of genome-wide information on sex differences in gene expression and in particular splicing in the human brain. Here we address this issue by using post-mortem adult human brain and spinal cord samples originating from 137 neuropathologically confirmed control individuals to study whole-genome gene expression and splicing in 12 CNS regions. We show that sex differences in gene expression and splicing are widespread in adult human brain, being detectable in all major brain regions and involving 2.5% of all expressed genes. We give examples of genes where sex-biased expression is both disease-relevant and likely to have functional consequences, and provide evidence suggesting that sex biases in expression may reflect sex-biased gene regulatory structures

    Effect of colour vision status on insect prey capture efficiency of captive and wild tamarins (Saguinus spp.)

    The colour vision polymorphism of most New World primates is a model system to study the function of colour vision. Theories for the evolution of primate trichromacy focus on the efficient detection and selection of ripe fruits and young leaves amongst mature leaves, when trichromats are likely to be better than dichromats. We provide data on whether colour vision status affects insect capture in primates. Trichromatic tamarins (Saguinus spp.) catch more prey than dichromats, but dichromats catch a greater proportion of camouflaged prey than trichromats. The prey caught does not differ in size between the two visual phenotypes. Thus two factors may contribute to the maintenance of genetic polymorphism of middle- to long-wavelength photopigments in Platyrrhines: the advantage in finding fruit and leaves, which supports the maintenance of the polymorphism through a heterozygote advantage, and the dichromats’ exploitation of different (e.g., camouflaged) food, which results in frequency-dependent selection on the different colour vision phenotypes

    Globular Cluster UVIT legacy Survey (GlobUleS) III. Omega Centauri in Far-Ultraviolet

    We present the first comprehensive study of the most massive globular cluster Omega Centauri in the far-ultraviolet (FUV) extending from the center to ~ 28% of the tidal radius using the Ultraviolet Imaging Telescope aboard AstroSat. A comparison of the FUV-optical color-magnitude diagrams with available canonical models reveals that the horizontal branch (HB) stars bluer than the knee (hHBs) and the white dwarfs (WDs) are fainter in the FUV by ~ 0.5 mag than model predictions. They are also fainter than their counterparts in M13, another massive cluster. We simulated HB with at least five subpopulations, including three He-rich populations with a substantial He enrichment of Y up to 0.43 dex, to reproduce the observed FUV distribution. We find the He-rich younger subpopulations to be radially more segregated than the He-normal older ones, suggesting an in-situ enrichment from older generations. The Omega Cen hHBs span the same effective temperature range as their M13 counterparts, but some have smaller radii and lower luminosities. This may suggest that a fraction of Omega Cen hHBs are less massive than those of M13, similar to the result derived from earlier spectroscopic studies of outer extreme HB stars. The WDs in Omega Cen and M13 have similar luminosity-radius-effective temperature parameters, and 0.44 - 0.46 M_\odot He-core WD model tracks evolving from progenitors with Y = 0.4 dex are found to fit the majority of these. This study provides constraints on the formation models of Omega Cen based on the estimated range in age, [Fe/H] and Y (in particular), for the HB stars.Comment: Accepted for publication in ApJL; 13 pages, 5 figures, 1 tabl
