    Electron energy-loss spectrometry on lithiated graphite

    Transmission electron energy-loss spectrometry was used to investigate the electronic states of metallic Li and LiC6, which is the Li-intercalated graphite used in Li-ion batteries. The Li K edges of metallic Li and LiC6 were nearly identical, and the C K edges were only weakly affected by the presence of Li. These results suggest only a small charge transfer from Li to C in LiC6, contrary to prior results from surface spectra obtained by x-ray photoelectron spectroscopy. Effects of radiation damage and sample oxidation in the transmission electron microscopy are also reported

    White Lines and 3d-Occupancy for the 3d Transition-Metal Oxides

    Electron energy-loss spectrometry was employed to measure the white lines at the L23 absorption edges of the 3d transition-metal oxides and lithium transition-metal oxides. The white-line ratio (L3/L2) was found to increase between d^0 and d^5 and decrease between d^5 and d^10, consistent with previous results for the transition metals and their oxides. The intensities of the white lines, normalized to the post-edge background, are linear for the 3d transition-metal oxides and lithium transition-metal oxides. An empirical correlation between normalized white-line intensity and 3d occupancy is established. It provides a method for measuring changes in the 3d-state occupancy. As an example, this empirical relationship is used to measure changes in the transition-metal valences of Li_{1-x}Ni_{0.8}Co_{0.2}O_2 in the range of 0 < x < 0.64. In these experiments the 3d occupancy of the nickel ion decreased upon lithium deintercalation, while the cobalt valence remained constant.Comment: 6 pages, 7 figure

    P?ginas 39-44El estado de madurez es considerado uno de los factores determinantes en la comercializaci?n de los productos hortofrut?colas, principalmente, por su relaci?n directa con la calidad y el tiempo de conservaci?n en poscosecha. La mora de Castilla (Rubus glaucus Benth), considerada un fruto no climat?rico altamente perecedero, evidencia en su recolecci?n heterogeneidad en forma, tama?o, color, peso y composici?n, que genera altas p?rdidas durante las etapas de cosecha, poscosecha y comercializaci?n, lo que, en consecuencia, afecta vol?menes en el mercado y produce p?rdidas econ?micas. Por lo anterior, el objetivo de este estudio fue establecer el grado de madurez de cosecha adecuado para la comercializaci?n de la mora proveniente del ca??n del Combeima, para lo cual se recurri? a la evaluaci?n de par?metros f?sicos, qu?micos y fisiol?gicos en producto con grado de madurez 4, 5 y 6 seg?n la NTC 4106. Los resultados obtenidos evidencian diferencias significativas en el estado de madurez 4 respecto del peso fresco que se reduce en promedio 12 %, el m?nimo contenido de s?lidos solubles (6.93 ? 0.12 ?Bx) que indicen en el ?ndice de madurez y la superior intensidad respiratoria durante su almacenamiento refrigerado, aspectos que reducen la calidad y vida ?til de este fruto. Se concluy? que el estado de madurez recomendable para la cosecha y comercializaci?n del producto evaluado es el 5, teniendo en cuenta que en este se alcanza la adecuada acumulaci?n de ?cidos, s?lidos solubles, tama?o y forma caracter?stica del fruto, sin diferencias significativas en peso al compararse con la madurez comercial actual (GM 6).ABSTRACT. The maturity stage is considered a factor decisive in the marketing of horticultural products, mainly because its direct related with the quality and postharvest shelf life. Castilla blackberry (Rubus glaucus Benth) is considered a non-climateric highly perishable fruit, presented in its harvest heterogeneity, in shape, size, color, weight and composition, generating high losses during the harvest, post-harvest and marketing that in consequence affect the market stock and economic losses. Therefore, the aim of this study was establish the proper harvest maturity for marketing of blackberry from Canyon Combeima, which was assement of physical, chemical and physiological with maturity level 4, 5 and 6 according to standard Icontec 4106. The results showed significant differences in maturity stage 4 respect to fresh weight was reduced 12 %, minimum soluble solids content (6.93 ? 0.12 ?Bx) influencing maturity index and higher respiratory rate during refrigerated storage, both of which reduce the quality and shelf life of this fruit. It was concluded that the state of maturity harvest and marketing recommended of the product evaluated is five, considering that in that state it reached adequate accumulation of acids, soluble solids, size and characteristic form of the fruit, without significant differences in weight when compared with the actual commercial maturity (GM 6)

    Dose, exposure time, and resolution in Serial X-ray Crystallography

    The resolution of X-ray diffraction microscopy is limited by the maximum dose that can be delivered prior to sample damage. In the proposed Serial Crystallography method, the damage problem is addressed by distributing the total dose over many identical hydrated macromolecules running continuously in a single-file train across a continuous X-ray beam, and resolution is then limited only by the available molecular and X-ray fluxes and molecular alignment. Orientation of the diffracting molecules is achieved by laser alignment. We evaluate the incident X-ray fluence (energy/area) required to obtain a given resolution from (1) an analytical model, giving the count rate at the maximum scattering angle for a model protein, (2) explicit simulation of diffraction patterns for a GroEL-GroES protein complex, and (3) the frequency cut off of the transfer function following iterative solution of the phase problem, and reconstruction of an electron density map in the projection approximation. These calculations include counting shot noise and multiple starts of the phasing algorithm. The results indicate counting time and the number of proteins needed within the beam at any instant for a given resolution and X-ray flux. We confirm an inverse fourth power dependence of exposure time on resolution, with important implications for all coherent X-ray imaging. We find that multiple single-file protein beams will be needed for sub-nanometer resolution on current third generation synchrotrons, but not on fourth generation designs, where reconstruction of secondary protein structure at a resolution of 0.7 nm should be possible with short exposures.Comment: 19 pages, 7 figures, 1 tabl

    Systemic rheumatic diseases are a heterogeneous group of autoimmune disorders that affect the connective tissue, characterized by the involvement of multiple organs, leading to disability, organ failure and premature mortality. Despite the advances in recent years, the therapeutic options for these diseases are still limited and some patients do not respond to the current treatments. Interleukin-17 (IL-17) is a cytokine essential in the defense against extracellular bacteria and fungi. Disruption of IL-17 homeostasis has been associated with the development and progression of rheumatic diseases, and the approval of different biological therapies targeting IL-17 for the treatment of psoriatic arthritis (PsA) and ankylosing spondylitis (AS) has highlighted the key role of this cytokine. IL-17 has been also implicated in the pathogenesis of systemic rheumatic diseases, including systemic lupus erythematosus (SLE), Sjogren's syndrome (SS) and systemic sclerosis (SSc). The aim of this review is to summarize and discuss the most recent findings about the pathogenic role of IL-17 in systemic rheumatic and its potential use as a therapeutic option

    Semaphorin (Sema)4A is a transmembrane glycoprotein that is elevated in several autoimmune diseases such as systemic sclerosis, rheumatoid arthritis and multiple sclerosis. Sema4A has a key role in the regulation of Thelper Th1 and Th2 differentiation and we recently demonstrated that CD4(+) T cell activation induces the expression of Sema4A. However, the autocrine role of Sema4A on Th cell differentiation remains unknown. Naive Th cells from healthy controls were cell sorted and differentiated into Th1, Th2 and Th17 in the presence or absence of a neutralizing antibody against the Sema4A receptor PlexinD1. Gene expression was determined by quantitative PCR and protein expression by ELISA and flow cytometry. We found that the expression of Sema4A is induced during Th1, Th2 and Th17 differentiation. PlexinD1 neutralization induced the differentiation of Th1 cells, while reduced the Th2 and Th17 skewing. These effects were associated with an upregulation of the transcription factor T-bet by Th1 cells, and to downregulation of GATA3 and RORgammat in Th2 cells and Th17 cells, respectively. Finally, PlexinD1 neutralization regulates the systemic sclerosis patients serum-induced cytokine production by CD4(+) T cells. Therefore, the autocrine Sema4A-PlexinD1 signaling acts as a negative regulator of Th1 skewing but is a key mediator on Th2 and Th17 differentiation, suggesting that dysregulation of this axis might be implicated in the pathogenesis of CD4(+) T cell-mediated diseases

    Identification of candidate host serum and saliva biomarkers for a better diagnosis of active and latent tuberculosis infection

    In our work, we aim to identify new candidate host biomarkers to discriminate between active TB patients (n = 28), latent infection (LTBI; n = 27) and uninfected (NoTBI; n = 42) individuals. For that, active TB patients and their contacts were recruited that donated serum and saliva samples. A multiplex assay was performed to study the concentration of different cytokines, chemokines and growth factors. Proteins with significant differences between groups were selected and logistic regression and the area under the ROC curve (AUC) was used to assess the diagnostic accuracy. The best marker combinations that discriminate active TB from NoTBI contacts were [IP-10 + IL-7] in serum and [Fractalkine + IP-10 + IL-1alpha + VEGF] in saliva. Best discrimination between active TB and LTBI was achieved using [IP-10 + BCA-1] in serum (AUC = 0.83) and IP-10 in saliva (p = 0.0007; AUC = 0.78). The levels of TNFalpha (p = 0.003; AUC = 0.73) in serum and the combination of [Fractalkine+IL-12p40] (AUC = 0.83) in saliva, were able to differentiate between NoTBI and LTBI contacts. In conclusion, different individual and combined protein markers could help to discriminate between active TB and both uninfected and latently-infected contacts. The most promising ones include [IP-10 + IL-7], [IP-10 + BCA-1] and TNFalpha in serum and [Fractalkine + IP-10 + IL-1alpha + VEGF], IP-10 and [Fractalkine+IL-12p40] in saliva

    Partially and Fully Frustrated Coupled Oscillators With Random Pinning Fields

    We have studied two specific models of frustrated and disordered coupled Kuramoto oscillators, all driven with the same natural frequency, in the presence of random external pinning fields. Our models are structurally similar, but differ in their degree of bond frustration and in their finite size ground state properties (one has random ferro- and anti-ferromagnetic interactions; the other has random chiral interactions). We have calculated the equilibrium properties of both models in the thermodynamic limit using the replica method, with emphasis on the role played by symmetries of the pinning field distribution, leading to explicit predictions for observables, transitions, and phase diagrams. For absent pinning fields our two models are found to behave identically, but pinning fields (provided with appropriate statistical properties) break this symmetry. Simulation data lend satisfactory support to our theoretical predictions.Comment: 37 pages, 7 postscript figure
