    Strategies for Prevention of Pertussis

    Introdução: Nos últimos 30 anos, em vários países, tem sido relatado um aumento da incidência de tosse convulsa, sobretudo em adolescentes e adultos, apesar das altas taxas de cobertura de imunização primária. Objectivos: Rever aspectos da epidemiologia da tosse convulsa, descrever algumas estratégias de controlo, com ênfase nas que incluem a utilização de vacinas com menores quantidades do toxóide diftérico e de alguns componentes pertussis (dTpa), e a avaliação da eficácia e efectividade destas vacinas. Métodos: Selecção de artigos relevantes, através da base de dados PubMed e sítios de acesso livre da internet, publicados entre 1991 e 2011. Resultados: As alterações do padrão epidemiológico são atribuídas, principalmente, à diminuição da imunidade ao longo dos anos, após a vacinação ou infecção natural. Os adolescentes e adultos foram identificados como importantes fontes de transmissão da doença para lactentes muito jovens que, uma vez não imunizados ou parcialmente imunizados, são mais vulneráveis às complicações relacionadas com a doença e apresentam maior mortalidade. A vacina (dTpa), formulada para o uso em adolescentes e adultos é segura e eficaz. A sua utilização também reduz a transmissão da tosse convulsa para os grupos etários com alto risco de complicações. Conclusão: A disponibilidade da dTpa oferece novas oportunidades para reduzir o impacto da tosse convulsa. A modificação das estratégias preventivas pode levar a um melhor controlo global da doença

    Spitzer-MIPS survey of the young stellar content in the Vela Molecular Cloud-D

    A new, unbiased Spitzer-MIPS imaging survey (~1.8 square degs) of the young stellar content of the Vela Molecular Cloud-D is presented. The survey is complete down to 5mJy and 250mJy at 24micron (mu) and 70mu, respectively. 849 sources are detected at 24mu and 52 of them also have a 70mu counterpart. The VMR-D region is one that we have already partially mapped in dust and gas millimeter emission, and we discuss the correlation between the Spitzer compact sources and the mm contours. About half of the 24mu sources are located inside the region delimited by the 12CO(1-0) contours (corresponding to only one third of the full area mapped with MIPS) with a consequent density increase of about 100% of the 24mu sources [four times for 70mu ones] moving from outside to inside the CO contours. About 400 sources have a 2MASS counterpart. So we have constructed a Ks vs. Ks-[24] diagram and identified the protostellar population. We find an excess of Class I sources in VMR-D in comparison with other star forming regions. This result is reasonably biased by the sensitivity limits, or, alternatively, may reflect a very short lifetime (<=10^6yr) of the protostellar content in this cloud. The MIPS images have identified embedded cool objects in most of the previously identified starless cores; in addition, there are 6 very young, possibly Class 0 objects identified. Finally we report finding of the driving sources for a set of five out of six very compact protostellar jets previously discovered in near-infrared images.Comment: 29 pages, 14 figures.     AB0563 DISCONTINUATION OF ANTI-TNFα IN PATIENTS WITH PSORIATIC ARTHRITIS: A SINGLE-CENTER EXPERIENCE

    Background:TNF inhibitors have been largely demonstrated to be effective and reasonably safe for the treatment of psoriatic arthritis (PsA). Current EULAR guidelines recommend the use of an anti-TNF as first choice treatment in patients with PsA for whom a synthetic DMARD (usually methotrexate or leflunomide) is not efficacious or not well tolerated [1]. In a scenario where biologic treatments are easily available, and the treat to target strategy is widely accepted, a complete disease remission or at least a minimal disease activity are considered realistic goals to be achieved in a growing proportion of patients [2]. However, there remains very little research regarding anti-TNF discontinuation in patients who achieved a complete remission [3-5].Objectives:The primary aim of this study was to measure the disease-free interval after anti-TNF discontinuation, secondary it was investigated whether the use of Power Doppler Ultrasound (PDUS) and Contrast Enhanced Ultrasound (CEUS) could improve the diagnostic accuracy in the recognition of the relapse. Finally, we wanted to characterize the clinical features of the disease recurrence.Methods:From June 2018, 35 patients with PsA (27 males and 8 female) treated with anti-TNF, in stable remission were prospectively monitored for 1 year after treatment discontinuation. Remission was defined as documented absence of clinical and ultrasonographic signs of arthritis or enthesitis. Complete rheumatological and dermatological examinations were performed in all participants, at baseline and every 8-12 weeks: American College of Rheumatology (ACR) 66-68 joint count; Psoriasis Area Severity Index (PASI); patient pain visual analog score (VAS); patient global disease activity VAS; Health Assessment Questionnaire (HAQ); Leeds Enthesitis Index (LEI); Bath Ankylosing Spondylitis Disease Activity Index (BASDAI); Bath Ankylosing Spondylitis Functional Index (BASFI); Power Doppler Ultrasound (PDUS) of the involved joints and entheses, Contrast Enhanced Ultrasound (CEUS) of a selected joint or enthesis and laboratory inflammation tests.Results:31 out of the 35 enrolled patients, experienced a disease recurrence with an average disease-free interval of 27.9±21.1 weeks (Figure 1). In 3 patients the treatment was restored for a relapse of the skin psoriasis, 8 patients reported only axial symptoms of disease relapse and 20 patients had both axial and peripheral joints involvement (average DAPSA score of 23.6±11.1; average BASDAI score of 4.7±2.6; average BASFI score 4.5±2.9). In all cases the disease flare was moderate and all patients promptly regained remission after restarting the treatment. Both PDUS and CEUS were safe and reliable showing a good percentage of accordance (95,4%) in detecting synovitis and enthesitis.Conclusion:The rate of disease relapse of PsA after anti-TNF discontinuation is relevant.     Transcriptome analysis of the venom gland of the Mexican scorpion Hadrurus gertschi (Arachnida: Scorpiones)

    <p>Abstract</p> <p>Background</p> <p>Scorpions like other venomous animals posses a highly specialized organ that produces, secretes and disposes the venom components. In these animals, the last postabdominal segment, named telson, contains a pair of venomous glands connected to the stinger. The isolation of numerous scorpion toxins, along with cDNA-based gene cloning and, more recently, proteomic analyses have provided us with a large collection of venom components sequences. However, all of them are secreted, or at least are predicted to be secretable gene products. Therefore very little is known about the cellular processes that normally take place inside the glands for production of the venom mixture. To gain insights into the scorpion venom gland biology, we have decided to perform a transcriptomic analysis by constructing a cDNA library and conducting a random sequencing screening of the transcripts.</p> <p>Results</p> <p>From the cDNA library prepared from a single venom gland of the scorpion <it>Hadrurus gertschi</it>, 160 expressed sequence tags (ESTs) were analyzed. These transcripts were further clustered into 68 unique sequences (20 contigs and 48 singlets), with an average length of 919 bp. Half of the ESTs can be confidentially assigned as homologues of annotated gene products. Annotation of these ESTs, with the aid of Gene Ontology terms and homology to eukaryotic orthologous groups, reveals some cellular processes important for venom gland function; including high protein synthesis, tuned posttranslational processing and trafficking. Nonetheless, the main group of the identified gene products includes ESTs similar to known scorpion toxins or other previously characterized scorpion venom components, which account for nearly 60% of the identified proteins.</p> <p>Conclusion</p> <p>To the best of our knowledge this report contains the first transcriptome analysis of genes transcribed by the venomous gland of a scorpion. The data were obtained for the species <it>Hadrurus gertschi</it>, belonging to the family Caraboctonidae. One hundred and sixty ESTs were analyzed, showing enrichment in genes that encode for products similar to known venom components, but also provides the first sketch of cellular components, molecular functions, biological processes and some unique sequences of the scorpion venom gland.</p

    A necklace of dense cores in the high-mass star forming region G35.20-0.74N: ALMA observations

    The present study aims at characterizing the massive star forming region G35.20N, which is found associated with at least one massive outflow and contains multiple dense cores, one of them recently found associated with a Keplerian rotating disk. We used ALMA to observe the G35.20N region in the continuum and line emission at 350 GHz. The observed frequency range covers tracers of dense gas (e.g. H13CO+, C17O), molecular outflows (e.g. SiO), and hot cores (e.g. CH3CN, CH3OH). The ALMA 870 um continuum emission map reveals an elongated dust structure (0.15 pc long and 0.013 pc wide) perpendicular to the large-scale molecular outflow detected in the region, and fragmented into a number of cores with masses 1-10 Msun and sizes 1600 AU. The cores appear regularly spaced with a separation of 0.023 pc. The emission of dense gas tracers such as H13CO+ or C17O is extended and coincident with the dust elongated structure. The three strongest dust cores show emission of complex organic molecules characteristic of hot cores, with temperatures around 200 K, and relative abundances 0.2-2x10^(-8) for CH3CN and 0.6-5x10^(-6) for CH3OH. The two cores with highest mass (cores A and B) show coherent velocity fields, with gradients almost aligned with the dust elongated structure. Those velocity gradients are consistent with Keplerian disks rotating about central masses of 4-18 Msun. Perpendicular to the velocity gradients we have identified a large-scale precessing jet/outflow associated with core B, and hints of an east-west jet/outflow associated with core A. The elongated dust structure in G35.20N is fragmented into a number of dense cores that may form massive stars. Based on the velocity field of the dense gas, the orientation of the magnetic field, and the regularly spaced fragmentation, we interpret this elongated structure as the densest part of a 1D filament fragmenting and forming massive stars.Comment: 24 pages, 26 figures, accepted for publication in Astronomy and Astrophysics (abstract modified to fit arXiv restrictions

    Stability of quantized time-delay nonlinear systems: A Lyapunov-Krasowskii-functional approach

    Lyapunov-Krasowskii functionals are used to design quantized control laws for nonlinear continuous-time systems in the presence of constant delays in the input. The quantized control law is implemented via hysteresis to prevent chattering. Under appropriate conditions, our analysis applies to stabilizable nonlinear systems for any value of the quantization density. The resulting quantized feedback is parametrized with respect to the quantization density.     Do dogs and cats passively carry sars-cov-2 on hair and pads?

    The epidemiological role of domestic animals in the spread and transmission of SARS-CoV-2 to humans has been investigated in recent reports, but some aspects need to be further clarified. To date, only in rare cases have dogs and cats living with COVID-19 patients been found to harbour SARS-CoV-2, with no evidence of pet-to-human transmission. The aim of the present study was to verify whether dogs and cats act as passive mechanical carriers of SARS-CoV-2 when they live in close contact with COVID-19 patients. Cutaneous and interdigital swabs collected from 48 dogs and 15 cats owned by COVID-19 patients were tested for SARS-CoV-2 by qRT-PCR. The time elapsed between owner swab positivity and sample collection from pets ranged from 1 to 72 days, with a median time of 23 days for dogs and 39 days for cats. All samples tested negative, suggesting that pets do not passively carry SARS-CoV-2 on their hair and pads, and thus they likely do not play an important role in the virus transmission to humans. This data may contribute to confirming that the direct contact with the hair and pads of pets does not represent a route for the transmission of SARS-CoV-2
