Diagnosis of Human Leptospirosis in a Clinical Setting: Real-Time PCR High Resolution Melting Analysis for Detection of Leptospira at the Onset of Disease:
Currently, direct detection of Leptospira can be done in clinical laboratories by conventional and by real-time PCR (qRT-PCR). We tested a biobank of paired samples of serum and urine from the same patient (202 patients) presenting at the hospital in an area endemic for leptospirosis using qRT-PCR followed by high resolution melting (HRM) analysis. The results were compared with those obtained by conventional nested PCR and with the serologic gold standard microscopic agglutination test (MAT). Differences were resolved by sequencing. qRT-PCR-HRM was positive for 46 of the 202 patients (22.7%, accuracy 100%) which is consistent with known prevalence of leptospirosis in the Azores. MAT results were positive for 3 of the 46 patients (6.5%). Analysis of paired samples allowed us to identify the illness point at which patients presented at the hospital: onset, dissemination or excretion. The melting curve analysis of Leptospira species revealed that 60.9% (28/46) of patients were infected with L. interrogans and 39.1% (18/46) were infected with L. borgpetersenii, both endemic to the Azores. We validated the use of qRT-PCR-HRM for diagnosis of leptospirosis and for identification of the Leptospira species at the earliest onset of infection in a clinical setting, in less than 2 hours.publishersversionpublishe
Ensaio em rede de fungicidas para o controle de mancha alvo (Corynespora cassiicola), na cultura da soja, Sinop/MT, safra 2017/2018.
A cultura da soja tem grande expressão social e econômica no Estado de Mato Grosso, com 9.464.344 ha cultivados na safra 2017/2018 e uma estimativa de produção de 31.789.630 de toneladas de grãos, com média de 3.358,8 kg/ha (IMEA, 2018). Um dos fatores que afetam a produtividade é a ocorrência de doenças entre elas a Mancha Alvo, causada pelo fungo Corynespora cassiicola. O objetivo desse trabalho foi avaliar em condições de campo a eficiência de fungicidas para o controle da mancha alvo e de outras doenças na cultura da soja, na safra 2017/2018. Foi instalado um ensaio em condições de campo, na área experimental da Agropel, em Sinop, MT, semeado em 10/11/2017. Utilizou-se a cultivar MonSoy 7739 IPRO e seguiu-se as recomendações agronômicas de acordo com as Tecnologias de Produção de Soja (Tecnologias ...., 2013). As aplicações dos produtos foram efetuadas com pulverizador costal a pressão constante e volume de calda de 150 L/ha. A barra utilizada era de 4 bicos marca Teejet tipo TJ6011/002VS, sob pressão constante de 2 kgf.cm-2
Depressive symptoms and its associated factors in 13-year-old urban adolescents.
The available estimates reveal that 20-50% of adolescents report depressive symptoms, being one of the most prevalent health problems in adolescence. The aim of this study was to assess the prevalence of depressive symptoms in a community sample of 13-year-old adolescents and identify associated features. Thirteen year-old adolescents attending private and public schools in Porto (n = 1,988, 52.2% females) were evaluated from October 2003 to June 2004 and completed a questionnaire including health behaviors and the Beck Depression Inventory II. A questionnaire on parents' socio-demographics and clinical characteristics was sent home. Data were analyzed separately by sex. Odds ratios (ORs) and 95% confidence intervals (CIs) were estimated using logistic regression. The prevalence of depressive symptoms was 18.8% in girls and 7.6% in boys (p < 0.001). Boys with a family history of depression and girls with smoking habits had a significantly increased risk of depressive symptoms (OR = 2.18, 95%CI 1.00-4.71; OR = 2.34, 95%CI 1.46-3.76). Menarche at an early age significantly increased the risk of depressive symptoms. The characteristics most strongly associated with depressive symptoms were family history of depression among boys, tobacco consumption and an early age at menarche among girls. The high prevalence of depressive symptoms early in adolescence calls for the awareness of public health professionals.publishersversionpublishe
Nanostructured Systems Containing Rutin: In Vitro Antioxidant Activity and Photostability Studies
The improvement of the rutin photostability and its prolonged in vitro antioxidant activity were studied by means of its association with nanostructured aqueous dispersions. Rutin-loaded nanocapsules and rutin-loaded nanoemulsion showed mean particle size of 124.30 ± 2.06 and 124.17 ± 1.79, respectively, polydispersity index below 0.20, negative zeta potential, and encapsulation efficiency close to 100%. The in vitro antioxidant activity was evaluated by the formation of free radical ·OH after the exposure of hydrogen peroxide to a UV irradiation system. Rutin-loaded nanostructures showed lower rutin decay rates [(6.1 ± 0.6) 10−3 and (5.1 ± 0.4) 10−3 for nanocapsules and nanoemulsion, respectively] compared to the ethanolic solution [(35.0 ± 3.7) 10−3 min−1] and exposed solution [(40.1 ± 1.7) 10−3 min−1] as well as compared to exposed nanostructured dispersions [(19.5 ± 0.5) 10−3 and (26.6 ± 2.6) 10−3, for nanocapsules and nanoemulsion, respectively]. The presence of the polymeric layer in nanocapsules was fundamental to obtain a prolonged antioxidant activity, even if the mathematical modeling of the in vitro release profiles showed high adsorption of rutin to the particle/droplet surface for both formulations.
