    Effects of type of physical exercise and leisure activities on the depression scores of obese Brazilian adolescent girls

    Several studies have indicated that depressive states may lead to hypokinesia with diminished metabolic rate and energy use. Hypokinesia associated with certain eating behaviors may lead to an unfavorable energy balance that can contribute to the emergence and prevalence of obesity among children and adults. The purpose of the present study was to examine the possibility of reducing depression inventory scores in female adolescents with third-degree obesity while testing the effectiveness of different exercise programs in reducing anxiety and depression scores. The sample consisted of 40 female subjects (mean age 16 ± 1.56 years) divided into 4 groups (aerobic training, anaerobic training, leisure activities, and control). Subjects had a body mass index of 95% or more in relation to the 50th percentile. The aerobic program consisted of three ergometric bicycle sessions per week over a 3-month period (12 weeks) and the activities were prescribed after determining the anaerobic ventilatory threshold (VO2 threshold). Anaerobic training was based on the Wingate anaerobic power test. The leisure program consisted of a varied range of activities (games, exercises, etc.). A nutritionist interviewed the members of these two groups and the control group every week in order to adapt them to the nutritional guidelines proposed for the study. The study showed that all three programs (aerobic exercise, anaerobic exercise and leisure activities) were effective in reducing body mass.     Lactate transporters and vascular factors in HPV-induced squamous cell carcinoma of the uterine cervixBACKGROUND: Tumour microenvironment is a fundamental aspect of tumour behaviour, modulating important events as cancer cell migration and invasion, as well as angiogenesis and metastisation. Among other microenvironment features, hypoxia and acidity play important roles in this modulation. As the metabolic reprogramming of cancer cells induces extracellular acidity, which in turn induces angiogenesis, and hypoxia induces both the metabolic reprogramming and angiogenesis, the present study aims to evaluate the immunohistochemical expression of a variety of metabolic and vascular markers as common targets of the hypoxic microenvironment in a series of cervical squamous cells carcinoma, as well as using an in vitro 3D culture model. METHODS: Immunohistochemical expression of MCT1, MCT4, CD147, GLUT1 and CAIX was assessed in a series of 28 chronic cervicitis, 34 LSIL, 29 HSIL, 38 cases of squamous cells carcinoma (SCC), as well as in in vitro 3D culture of keratinocytes expressing HPV genes. Furthermore, VEGF family members' expression was assessed in the SCC cases. The expression profiles were associated with patients' clinicopathological parameters. RESULTS: We found an increase of MCT4 expression along progression to malignancy in cervical samples. Also, MCT4 was associated with CD147 and CAIX expression. VEGF-A expression was more frequently found in cases without MCT1 expression. Both MCT4 and CD147 were more frequently expressed in younger patients at diagnosis while no associations were found between VEGF family and clinicopathological parameters. Finally, we show evidence for the upregulation of MCT4, as well as CD147 and CAIX, after HPV transfection. CONCLUSIONS: The results herein presented point at MCT4 as a promising therapeutic target in squamous cells carcinoma of the uterine cervix.     Ostreid herpesvirus 1 detection and relationship with Crassostrea gigas spat mortality in France between 1998 and 2006

    Since its molecular characterisation, Ostreid herpesvirus 1 (OsHV-1) has been regularly detected in Crassostrea gigas in France. Although its pathogenicity was demonstrated on larval stages, its involvement during mortality outbreaks at the juvenile stage was highly suspected but not evidenced. To investigate mortality outbreaks, the French National Network for Surveillance and Monitoring of Mollusc Health (REPAMO) carried out two surveys in juvenile C. gigas. The first survey lasted from 1998 to 2006 and was an epidemiological inquiry occurring when oyster farmers reported mortality outbreaks. The second survey, a longitudinal one, was set up in 1998 to complete the network observations on OsHV-1. Data analysis showed a specific pattern of mortality outbreaks associated with OsHV-1 detection. Ostreid herpesvirus 1 detection mainly appeared during the summer, suggesting the influence of the seawater temperature on its occurrence. It mostly presented a patchy distribution in the field in contrast to the nursery. Significant relationship between OsHV-1 detection and spat mortality was found, preferentially in sheltered and closed environments. The longitudinal survey confirmed most of the network observations. Although subsequent works particularly epidemiological surveys would be useful to confirm the causal link between the detection of OsHV-1 and the mortality outbreaks in juvenile C. gigas, the role of OsHV-1 in oyster mortality is progressing

    Lipid polymers in plant cell walls, such as cutin and suberin, build recalcitrant hydrophobic protective barriers. Their degradation is of foremost importance for both plant pathogenic and saprophytic fungi. Regardless of numerous reports on fungal degradation of emulsified fatty acids or cutin, and on fungi-plant interactions, the pathways involved in the degradation and utilisation of suberin remain largely overlooked. As a structural component of the plant cell wall, suberin isolation, in general, uses harsh depolymerisation methods that destroy its macromolecular structure.     Solution approaches based purely on economics do not lead to optimal success – at least, not in the medium to long term. Rather, a sustainable economy requires an equal consideration of the ecological and social dimensions. The green economy concept, or, better said, the green and fair economy concept, is in principle suitable for, if not avoiding an economic and financial crisis as experienced in the last five years, at least moderating its negative effects. The important thing here is to integrate the design of the concept as much as possible at the political, economic and individual level

    Release of hydro-soluble microalgal proteins using mechanical and chemical treatments

    In order to release proteins in the aqueous phase, high-pressure homogenization and alkaline treatments were applied to rupture the cell walls of five intensively grown microalgae. Protein characterisation was carried out by analysing the amino acid profiles of both the crude microalgae and the protein extracts, obtained after both types of treatment. The results showed that the proportion of proteins released from microalgae following both treatments was, in descending order: Porphyridium cruentum>Arthrospira platensis>Chlorella vulgaris>Nannochloropsis oculata>Haematococcus pluvialis, reflecting the increasingly protective, cell walls. Nonetheless, mechanical treatment released more proteins from all the microalgae compared to chemical treatment. The highest yield was for the fragile cell walled P. cruentum with 88% hydro-soluble proteins from total proteins, and the lowest from the rigid cell walled H. pluvialis with 41%. The proportion of essential and non-essential amino acids in the extract was assessed and compared to the crude microalgae profile. It was higher after alkaline treatment and much higher after high-pressure homogenization. These results suggest that non-essential amino acids are more concentrated actually inside the cells and that different types of proteins are being released by these two treatments

    The microalgal structure has been investigated to evaluate the release of proteins in aqueous media from five microalgae after conducting different cell disruption techniques: manual grinding, ultrasonication, alkaline treatment, and high-pressure treatment. After conducting cell disruption, the protein concentration in water was determined for all the microalgae and the results are discussed within the context of their cell wall structure. It was found that the aqueous media containing most protein concentration followed the order: high-pressure cell disruption>chemical treatment>ultrasonication>manual grinding. Fragile cell-walled microalgae were mostly attacked according to the following order: Haematococcus pluvialis<Nannochloropsis oculata<Chlorella vulgaris<Porphyridium cruentum≤Arthrospira platensis

    Understanding the effect of cell disruption methods on the diffusion of Chlorella vulgaris proteins and pigments in the aqueous phase

    Cell disruption of microalgae is usually evaluated by microscopic observations and quantification of the target molecules before and after cell disruption. The following study considers a new approach by analysing the diffusion behaviour of proteins and pigments of Chlorella vulgaris in an aqueous medium after applying different cell disruption methods. Results were revealed by microscopic observations, quantifying the concentration of the molecules of interest, and calculating their diffusion coefficient. Microscopic observations showed intact cells after applying chemical hydrolysis and ultrasonication. However, the majority of the cells lost their globular shape after bead milling and high-pressure homogenization. The protein concentration increased in the following order: ultrasonication bead milling > ultrasonication > high-pressure homogenization. Pigments were not detected in the aqueous phase of the chemical hydrolysis treatment, but their concentration and their diffusion were in the same order as proteins in the mechanical treatments. The study implied that diffusivity of the target molecules was not directly correlated to their increase concentration in the aqueous phase. Therefore, even if the cells were completely broken, diffusivity followed the hindered molecule diffusion phenomenon, which implies that somehow cells are not completely disrupted
