Riemann–Hilbert problems, Toeplitz operators and Q-classes
We generalize the notion of Q-classes C(Q1,Q2) , which was introduced in the context of Wiener–Hopf factorization, by considering very general 2 × 2 matrix functions Q1, Q2. This allows us to use a
mainly algebraic approach to obtain several equivalent representations for each class, to study the intersections of Q-classes and to explore their close connection with certain non-linear scalar equations. The results are applied to various factorization problems and to the study of Toeplitz operators with symbol in a Q-class. Hereditary breast cancer and ancestry in the Madeira archipelago: An exploratory study
Access to genetic testing and counselling in remote areas such as the Madeira archipelago, in the Northern Atlantic Ocean, may be complex. Different counselling methods, including telegenetics, should be explored. In this study, we characterise the Hereditary Breast/Ovarian Cancer (HBOC) families with Madeira ancestry enrolled in our programme. Of a total of 3, 566 index patients tested between January 2000 and June 2018, 68 had Madeira ancestry and 22 were diagnosed with a pathogenic germline variant (PV). As in the whole group, BRCA2 PV were more frequent in Madeira patients (68.4%: c.9382C>T (26.3%), c.658_659del (21%), c.156_157insAlu (10.5%), c.793+1G>A (5.3%) and c.298A>T (5.3%). However, the most frequently diagnosed PV in Madeira patients was the BRCA1 c.3331_3334del (31.6%). BRCA1/2 detection rates were 27.9% and 10.5% for Madeira and the whole group, respectively. This study is the first characterisation of HBOC patients with Madeira ancestry. A distinct pattern of BRCA1/2 variants was observed, and the geographic clustering of BRCA1 c.3331_3334del variant may support the possibility of a founder mutation previously described in Northern Portugal. The high detection rate observed reinforces the need to reduce gaps in access to genetic testing in Madeira and other remote areas. According to current guidelines, timely identification of HBOC patients can contribute to their ongoing care and treatment
Nanocomposite gels of poloxamine and Laponite for β-Lapachone release in anticancer therapy
Nano-hybrid systems have been shown to be an attractive platform for drug delivery. Laponite® RD (LAP), a biocompatible synthetic clay, has been exploited for its ability to establish of strong secondary interactions with guest compounds and hybridization with polymers or small molecules that improves, for instance, cell adhesion, proliferation, and differentiation or facilitates drug attachment to their surfaces through charge interaction. In this work, LAP was combined with Tetronics, X-shaped amphiphilic PPO-PEO (poly (propylene oxide)–poly (ethylene oxide) block copolymers. β-Lapachone (BLPC) was selected for its anticancer activity and its limited bioavailability due to very low aqueous solubility, with the aim to improve this by using LAP/Tetronic nano-hybrid systems. The nanocarriers were prepared over a range of Tetronic 1304 concentrations (1 to 20% w/w) and LAP (0 to 3% w/w). A combination of physicochemical methods was employed to characterize the hybrid systems, including rheology, particle size and shape (DLS, TEM), thermal analysis (TG and DSC), FTIR, solubility studies and drug release experiments. In vitro cytotoxicity assays were performed with BALB/3T3 and MCF-7 cell lines. In hybrid systems, a sol-gel transition can occur below physiological temperature. BLPC exhibits the most significant increase in solubility in formulations with a high concentration of T1304 (over 10% w/w) and 1.5% w/w LAP, or systems with only LAP (1.5%), with a 50 and 100-fold increase in solubilisation, respectively. TEM images showed spherical micelles of T1304, which elongated into wormlike micelles with concentration (20%) and in the presence of LAP, a finding that has not been reported before. A sustained release of BLPC over 140 hours was achieved in one of the formulations (10% T1304 with 1.5% laponite), which also showed the best selectivity index towards cancer cells (MCF-7) over BALB/3T3 cell lines. In conclusion, BLPC-loaded T1304/LAP nano-hybrid systems proved safe and highly effective and are thus a promising formulation for anticancer therapy.Financial support for this research was provided by the Coordination of Improvement of Higher-Level Personnel - Brazil (CAPES) - 1575/2015. Processos de ocupação nas novas fronteiras da Amazônia (o interflúvio do Xingu/Iriri).
Este trabalho apresenta os primeiros resultados do esforço conjunto de várias instituições, organizadas em torno da rede Geoma (Rede Temática de Pesquisa em Modelagem Ambiental da Amazônia) para avançar a compreensão dos novos padrões e processos de estruturação do território nas novas frentes no sul do Pará, analisando padrões de desmatamento e os processos que dão origem a esses padrões. Busca-se, aqui, produzir os subsídios necessários para o desenho de políticas públicas responsáveis, que não privilegiem um único aspecto do problema, como a abertura de estradas, por exemplo. Aponta-se, então, a partir desses primeiros resultados, que apenas uma solução integrada que procure estruturar os principais agentes e processos na cadeia produtiva seria possível para minorar os efeitos do desmatamento e nortear o desenvolvimento integrado para a região, com benefícios para a floresta e para as populações que ali vivem
Two-photon Lithography for 3D Magnetic Nanostructure Fabrication
Ferromagnetic materials have been utilised as recording media within data
storage devices for many decades. Confinement of the material to a two
dimensional plane is a significant bottleneck in achieving ultra-high recording
densities and this has led to the proposition of three dimensional (3D)
racetrack memories that utilise domain wall propagation along nanowires.
However, the fabrication of 3D magnetic nanostructures of complex geometry is
highly challenging and not easily achievable with standard lithography
techniques. Here, by using a combination of two-photon lithography and
electrochemical deposition, we show a new approach to construct 3D magnetic
nanostructures of complex geometry. The magnetic properties are found to be
intimately related to the 3D geometry of the structure and magnetic imaging
experiments provide evidence of domain wall pinning at a 3D nanostructured
junction
