    Steady supersonic Navier-Stokes solutions of a 75 deg delta wing

    Steady solutions about a slender sharp edged delta wing in a supersonic freestream for moderate and high angles of attack are obtained numerically by time integration of the unsteady compressible three dimensional laminar Navier-Stokes equations. The main features of the flow, including primary and secondary separation, and vortex position and strength, are adequately simulated in the numerical solutions. Improved resolution of the computational grid in the leading edge region from a previous solution had considerable effect on the accuracy of the solutions. Good agreement between numerical solutions and experimental data was obtained for two cases. A local timestepping procedure is used to speed convergence by approximately a factor of two

    Force and moment measurements on a 74 deg delta wing with an apex flap

    Results are presented of a subsonic experimental investigation of an apex flap concept on a 74 deg swept delta wing with trailing-edge flaps. The apex flap comprised approximately 6 percent of the wing area forward of a transverse hinge, allowing for upward and downward deflection angles from +40 deg to -20 deg. Upward deflection forces leading-edge vortex formation on the apex flap, resulting in an increased lift component on the apex area. The associated nose-up moment balances the nose-down moment due to trailing-edge flaps, resulting in sizeable increase in the trimmed lift coefficient particularly at low angles of attack. Nose-down apex deflection may be used to augment the pitch control for rapid recovery from high-alpha maneuvers. This report presents the balance data without analysis

    Leading-edge receptivity to a vortical freestream disturbance: A numerical analysis

    The receptivity to freestream vorticity of the boundary layer over a flat plate with an elliptic leading edge is investigated numerically. The flow is simulated by solving the incompressible Navier-Stokes system in general curvilinear coordinates with the vorticity and stream function as dependent variables. A finite-difference scheme which is second-order accurate in both space and time is used. As a first step, the steady basic-state solution is computed. Then a small amplitude vortical disturbance is introduced at the upstream boundary and the governing equations are solved time-accurately to evaluate the spatial and temporal growth of the perturbations leading to instability waves (Tollmien-Schlichting waves) inside the boundary layer. Preliminary results for a symmetric, 2-D disturbance reveal the presence of Tollmien-Schlichting waves aft of the flat-plate/ellipse juncture

    Virgin females of the parasitoid wasp Trichogramma turkestanica produce minute amounts of a sex pheromone, the identity of which has not been fully established. The enantioselective synthesis of a putative component of this pheromone, (6S,8S,10S)-4,6,8,10-tetramethyltrideca-2E,4E-dien-1-ol (2), is reported as a contribution to this identification. Catalytic asymmetric conjugate addition of methylmagnesium bromide and stereoselective Horner–Wadsworth–Emmons olefinations are used as the key steps, and 2 was obtained in 16 steps with an overall yield of 4.4%

    Immunomodulatory effects of intravenous BIS-1 F(ab')2 administration in renal cell cancer patients.

    We report the immunomodulatory effects of an intravenous treatment with F(ab')2 fragments of the bispecific monoclonal antibody BIS-1 during subcutaneous recombinant interleukin 2 (rIL-2) therapy of renal cell cancer (RCC) patients. BIS-1 is directed against both the CD3 antigen on T cells and the EGP-2 molecule on carcinoma cells and some normal epithelia. The amount of BIS-1 F(ab')2 bound to peripheral blood lymphocytes (PBLs) increased dose-dependently. This occupation degree was highest at the end of the 2 h infusion and rapidly decreased subsequently. During the first hour of BIS-1 F(ab')2 infusion the number of PBLs decreased slowly. This was followed by an increase in serum tumour necrosis factor alpha (TNF-alpha) concentrations and a rapid decrease in the numbers of peripheral blood lymphocytes, monocytes and eosinophils. In our view, the most likely explanation for the observed decrease in occupation degree of BIS-1 F(ab')2 and the rise in TNF-alpha levels is based on the assumption that BIS-1-carrying T cells leave the circulation. The CD3 antigens on these extravasated T cells become cross-linked by EGP-2 antigens, inducing TNF-alpha secretion. This results in an enhanced decrease in the numbers of PBLs, monocytes and eosinophils. These preliminary results suggest that BIS-1 F(ab')2 treatment during IL-2 therapy may induce local T-cell activation

    Immune response to varicella-zoster virus before and after renal transplantation

    Background: Herpes zoster (HZ) risk is high in renal transplant recipients. Vaccination prior to transplantation may provide a useful strategy for the prevention of HZ in the posttranplantation period. However, it is not known whether immunity to varicella-zoster virus (VZV) is affected due to treatment surrounding transplantation. Methods: Both humoral and cellular immunity to VZV were determined prior to and 2-3 years after renal transplantation in 60 adult patients, and 62 matched healthy controls. VZV-specific cellular immunity was measured by an interferon gamma (IFN gamma) enzyme-linked immunospot (ELISpot) assay and by analyzing T-cell functionality using flowcytometry. VZV-IgG levels were measured using an in-house glycoprotein enzyme-linked immunosorbent assay (gpELISA). Results: Using paired analysis, it was determined that numbers of IFN gamma-producing cells did not change after transplantation, but were significantly lower in transplant recipients after transplantation than in controls (p = 0.028). Patients in whom the post-transplant period was complicated by rejection or any acute infection (excluding HZ) had a lower number of IFN gamma-producing cells than patients who did not. VZV IgG levels did not differ from controls, but a significant decrease was observed after transplantation (p <0.0001). Conclusions: VZV-specific cellular immunity, which is essential in the prevention of HZ, did not markedly change in patients following renal transplantation. This suggests that preventive vaccination before transplantation may be beneficial. Our results extend knowledge on VZV immunity after transplantation, vital when considering strategies for the prevention of HZ in these patients

    Total Synthesis of a Mycolic Acid from Mycobacterium tuberculosis

    In Mycobacterium tuberculosis, mycolic acids and their glycerol, glucose, and trehalose esters ("cord factor") form the main part of the mycomembrane. Despite their first isolation almost a century ago, full stereochemical evaluation is lacking, as is a scalable synthesis required for accurate immunological, including vaccination, studies. Herein, we report an efficient, convergent, gram-scale synthesis of four stereo-isomers of a mycolic acid and its glucose ester. Binding to the antigen presenting protein CD1b and T cell activation studies are used to confirm the antigenicity of the synthetic material. The absolute stereochemistry of the syn-methoxy methyl moiety in natural material is evaluated by comparing its optical rotation with that of synthetic material
