"'One Hundred Per Cent American': Nationalism, Masculinity and American Legion Baseball in the 1920s," provides a sociohistorical analysis of baseball and social attitudes and ideologies of the pre- and post-World War I period, specifically focusing on the joining of nationalism and masculinity through the playing of sport. My work explores amateur baseball in the context of the post-World War I period (1920-1930), focusing on the American Legion's baseball program started during that same era. By incorporating the theorization of "hegemonic masculinity," first popularized by sociologist R.W. Connell and a major theme in the sociology of sport, I argue that amateur baseball constituted a distinct form of nationalist American masculinity that figured prominently in both the status of the sport and the understanding of gender within post-war American culture. By focusing on the instruction of these amateur players, I demonstrate how nationalism and masculinity converged through the kinesthetic "play"ing of baseball by young American males

'Right to the Active City': Public Recreation and Urban Governance in Baltimore

Since the inception of the Baltimore City Department of Recreation and Parks (BCRP) in 1940, public recreation in Baltimore has continued to be restructured in relation to changing modes of urban governance, in particular in regards to the city's network of recreation centers. More recently, the reorganization of recreation resulted in the 2011 Mayor's Recreation Center Task Force plan, which proposed the further reformation of the department and changes to the provision and distribution of recreation centers and recreational services. This dissertation - entitled Right to the Active City: Public Recreation and Urban Governance in Baltimore - draws from a diverse and reflexive theoretical and methodological approach in exploring the historical and contemporary forms, practices and experiences of public recreation in Baltimore, specifically focusing on the city's recreation centers as social and spatial manifestations of the processes of urban governance. In seeking to engage and analyze the individuals, institutions, spaces and practices of urban public recreation, the primary goals of this research are: 1) to examine the intersection of historical and current formations of recreation policy and broader processes of urban governance, including the implications of these changing arrangements for the localized experiences of public recreation; 2) to analyze the spaces of public recreation, in particular the changing forms and practices of planning and design that is embedded within a shift between different `recreation center' models; 3) to draw out and describe the often complex and contradictory inter-relationships between the City government, BCRP, community and non-profit groups and city residents, focusing on the associations that actively construct and constitute an emergent form of public recreation; and 4) to provide a nuanced research approach that both contributes to relevant scholarly fields, including public health, kinesiology, sociology, urban studies and physical cultural studies, and simultaneously seeks to promote the co-production of research that can be engaged by and with those involved in the processes of public recreation. In short, this research attempts to better grasp the lived experiences of the active urban body and urban physical cultures, through an analysis of the planning and provision of recreational sites, services and opportunities in a specific postindustrial metropolis

Conformational stability and activity analysis of two hydroxymethylbilane synthase mutants, K132N and V215E, with different phenotypic association with acute intermittent porphyria

The autosomal dominantly inherited disease AIP (acute intermittent porphyria) is caused by mutations in HMBS [hydroxymethylbilane synthase; also known as PBG (porphobilinogen) deaminase], the third enzyme in the haem biosynthesis pathway. Enzyme-intermediates with increasing number of PBG molecules are formed during the catalysis of HMBS. In this work, we studied the two uncharacterized mutants K132N and V215E comparative with wt (wild- type) HMBS and to the previously reported AIP-associated mutants R116W, R167W and R173W. These mainly present defects in conformational stability (R116W), enzyme kinetics (R167W) or both (R173W). A combination of native PAGE, CD, DSF (differential scanning fluorimetry) and ion-exchange chromatography was used to study conformational stability and activity of the recombinant enzymes. We also investigated the distribution of intermediates corresponding to specific elongation stages. It is well known that the thermostability of HMBS increases when the DPM (dipyrromethane) cofactor binds to the apoenzyme and the holoenzyme is formed. Interestingly, a decrease in thermal stability was measured concomitant to elongation of the pyrrole chain, indicating a loosening of the structure prior to product release. No conformational or kinetic defect was observed for the K132N mutant, whereas V215E presented lower conformational stability and probably a perturbed elongation process. This is in accordance with the high association of V215E with AIP. Our results contribute to interpret the molecular mechanisms for dysfunction of HMBS mutants and to establish genotype–phenotype relations for AIP.publishedVersio

Policing the Void: Recreation, Social Inclusion and the Baltimore Police Athletic League

    Get PDF
In this article, we explore the relationship between public recreation policy and planning and the transformation of urban governance in the context of the Police Athletic League centers in Baltimore, Maryland. In light of contemporary discussions of the role of youth programs for sport and physical activity within post-industrial cities, the origination, development, and eventual demise of Baltimore’s network of Police Activity League centers is an instructive, if disheartening, saga. It illustrates the social and political rationales mobilized in justifying recreation policy and programming, the framing of sport and physical activity as preventative measures towards crime and juvenile delinquency, and the precarity of such initiatives given the efficiency-driven orthodoxies of neoliberal urban entrepreneurialism (Harvey, 1989). This analysis emphasizes how the PAL centers were designed to ‘fill the void’ left by a declining system of public recreation, thereby providing an example of a recreation program as part of the “social problems industry” (Pitter & Andrews 1997)

Characterisation of a common hotspot variant in acute intermittent porphyria sheds light on the mechanism of hydroxymethylbilane synthase function

Hydroxymethylbilane synthase (HMBS) is the third enzyme involved in haem biosynthesis, in which it catalyses the formation of tetrapyrrole 1-hydroxymethylbilane (HMB). In this process, HMBS binds four consecutive substrate molecules, creating the enzyme-intermediate complexes ES, ES2, ES3 and ES4. Pathogenic variants in the HMBS gene are associated with the dominantly inherited disorder acute intermittent porphyria. In this study, we have characterised the p.R26H variant to shed light on the role of Arg26 in the elongation mechanism of HMBS and to provide insights into its effect on the enzyme. With selected biophysical methods, we have been able to show that p.R26H forms a single enzyme-intermediate complex in the ES2-state. We were also able to demonstrate that the p.R26H variant results in an inactive enzyme, which is unable to produce the HMB product.publishedVersio
