    The Ecology Of Marten In Southcentral Alaska

    Thesis (Ph.D.) University of Alaska Fairbanks, 1983The ecology of marten in the upper Susitna Basin, southcentral Alaska was studied from January 1980 to June 1982. Data were gathered on home range and movements, seasonal food habits, habitat use and winter energetic strategies. Radio telemetry was used to obtain a total of 560 locations for 17 marten. Mean home range sizes of marten along the Susitna River were 3.71 km('2) for females, 6.82 km('2) for males and 6.75 km('2) for adult males (2+ years). Marten were found to be nocturnal in autumn and to show strong variability in their diel activity patterns in late winter. Marten tended to move upward in elevation during spring and downward in autumn, contrary to the prevailing views of trappers. Analyses of marten scats and colon contents collected during four seasons showed the most important foods to be microtine rodents, squirrels, fruits and birds. Major foods showed strong seasonal variation in utilization. Microtines were most important in autumn and showed declining use over winter. Northern red-backed voles were the most important microtine species. Sciurids were most important in spring and appeared to be a nonpreferred alternative food. Marten made little use of shrews, snowshoe hares, porcupines or insects. Carrion and human foods were highly preferred and consumed when available. Habitat utilization was studied through the use of aerial transects and snow tracking and by identifying resting sites. Marten foraged for microtines more frequently than expected in vegetation types dominated by black spruce. Marten rested in winter primarily in active red squirrel middens in stands dominated by old-growth white spruce. Fat depot and organ weights and total body fat of marten were measured to find an indicator to total body fat. Marten were found to have extremely low body fat ratios which did not show a significant change over winter

    Capturing and radio ear-tagging neonatal vicuñas

    We used and evaluated the effectiveness of a hand-capture technique to capture and mark neonatal vicuñas (Vicugna vicugna) at San Guillermo National Park, west-central Argentina. We captured and marked 98 neonates during 5-31 January, 2008-2010. Capture success was 92% (106 attempts), with stationary marking teams being 2.5 times as successful in detecting and capturing neonates as mobile ones. Of neonates captured, 99% were 20 minutes old typically were too mobile to capture. Sex ratio at capture was biased toward males (1.5:1), while mean body weight and neck circumference were similar between sexes. Generally, mothers remained in sight during, and immediately reunited with their neonates after, capture. Four percent of the mothers ran out of sight but then returned, while 2% failed to return. Capture-related mortality was 5%, with maternal abandonment in response to capture activities (n = 3) the leading cause. Ear-tag transmitter had minimal effects on subsequent ear position. The success of our technique likely was based on terrain (flat) and vegetative conditions (short grass), high densities of animals for monitoring (particularly in a meadow), and a predictable, pulsed birth season. This hand-capturing method was efficient for handling and marking neonatal vicuñas.     Pulmonary immune responses to Aspergillus fumigatus in an immunocompetent mouse model of repeated exposures

    Aspergillus fumigatus is a filamentous fungus that produces abundant pigmented conidia. Several fungal components have been identified as virulence factors, including melanin; however, the impact of these factors in a repeated exposure model resembling natural environmental exposures remains unknown. This study examined the role of fungal melanin in the stimulation of pulmonary immune responses using immunocompetent BALB/c mice in a multiple exposure model. It compared conidia from wild-type A. fumigatus to two melanin mutants of the same strain, Δarp2 (tan) or Δalb1 (white). Mass spectrometry-based analysis of conidial extracts demonstrated that there was little difference in the protein fingerprint profiles between the three strains. Field emission scanning electron microscopy demonstrated that the immunologically inert Rodlet A layer remained intact in melanin-deficient conidia. Thus, the primary difference between the strains was the extent of melanization. Histopathology indicated that each A. fumigatus strain induced lung inflammation, regardless of the extent of melanization. In mice exposed to Δalb1 conidia, an increase in airway eosinophils and a decrease in neutrophils and CD8(+) IL-17(+) (Tc17) cells were observed. Additionally, it was shown that melanin mutant conidia were more rapidly cleared from the lungs than wild-type conidia. These data suggest that the presence of fungal melanin may modulate the pulmonary immune response in a mouse model of repeated exposures to A. fumigatus conidia

    The Role of Preoperative Bilateral Breast Magnetic Resonance Imaging in Patient Selection for Partial Breast Irradiation in Ductal Carcinoma In Situ

    Purpose. Women with ductal carcinoma in situ (DCIS) are often candidates for breast-conserving therapy, and one option for radiation treatment is partial breast irradiation (PBI). This study evaluates the use of preoperative breast magnetic resonance imaging (MRI) for PBI selection in DCIS patients. Methods. Between 2002 and 2009, 136 women with newly diagnosed DCIS underwent a preoperative bilateral breast MRI at Mayo Clinic in Florida. One hundred seventeen women were deemed eligible for PBI by the NSABP B-39 (National Surgical Adjuvant Breast and Bowel Project, Protocol B-39) inclusion criteria using physical examination, mammogram, and/or ultrasound. MRIs were reviewed for their impact on patient eligibility, and findings were pathologically confirmed. Results. Of the 117 patients, 23 (20%) were found ineligible because of pathologically proven MRI findings. MRI detected additional ipsilateral breast cancer in 21 (18%) patients. Of these women, 15 (13%) had more extensive disease than originally noted before MRI, and 6 (5%) had multicentric disease in the ipsilateral breast. In addition, contralateral breast cancer was detected in 4 (4%). Conclusions. Preoperative breast MRI altered the PBI recommendations for 20% of women. Bilateral breast MRI should be an integral part of the preoperative evaluation of all patients with DCIS being considered for PBI

    An Examination of the Association between FOXA1 Staining Level and Biochemical Recurrence following Salvage Radiation Therapy for Recurrent Prostate Cancer.

    BACKGROUND: Standardly collected clinical and pathological patient information has demonstrated only moderate ability to predict risk of biochemical recurrence (BCR) of prostate cancer in men undergoing salvage radiation therapy (SRT) for a rising PSA after radical prostatectomy (RP). Although elevated FOXA1 staining has been associated with poor patient outcomes following RP, it has not been studied in the specific setting of SRT after RP. The aim of this study was to evaluate the association between FOXA1 staining level and BCR after SRT for recurrent prostate cancer. METHODS: A total of 141 men who underwent SRT at our institution were included. FOXA1 staining levels in primary tumor samples were detected using immunohistochemistry. FOXA1 staining percentage and intensity were measured and multiplied together to obtain a FOXA1 H-score (range 0-12) which was our primary staining measure. P-values ≤ 0.0056 were considered as statistically significant after applying a Bonferroni correction for multiple comparisons. RESULTS: There was not a significant association between FOXA1 H-score and risk of BCR when considering H-score as an ordinal variable or as a categorical variable (all P ≥ 0.090). Similarly, no significant associations with BCR were observed for FOXA1 staining percentage or staining intensity (all P ≥ 0.14). CONCLUSIONS: FOXA1 staining level does not appear to have a major impact on risk of BCR after SRT

    Assessment of bladder filling during prostate cancer radiation therapy with ultrasound and cone-beam CT

    Prostate cancer patients undergoing external beam radiation therapy (EBRT) benefit from a full bladder to decrease bowel and bladder toxicity. Ultrasound may offer a proxy metric for evaluation, sparing CBCT dosing. Patients were prospectively enrolled pre-simulation from January 2017 to February 2018. Bladder volume was evaluated prior to RT using US daily and CBCT for three daily treatments and then weekly unless otherwise indicated. 29 patients completed median 40 days of RT, resulting in 478 CBCT and 1,099 US bladder volumes. 21 patients were treated to intact glands and 8 to the post-prostatectomy bed. Median patient age was 70 years. Bladder volume on CBCT and US positively correlated (r = 0.85), with average bladder volume for all patients of 162 mL versus 149 mL, respectively. Bladder volume during treatment was consistently lower than the volume at CT simulation (153 mL vs 194 mL, p<0.01) and progressively declined during treatment. Patients older than 70 years presented with lower average bladder volumes than those < 70 years (122 mL vs 208 mL, respectively, p<0.01). Patients with the highest agreement between CBCT and US (<10% variability) had higher average bladder volumes (192 mL vs 120 mL, p=0.01). US was found to be an accurate measure of bladder volume and may be used to monitor daily bladder volumes in patients being treated with radiation for prostate cancer

    Tick Extracellular Vesicles Enable Arthropod Feeding and Promote Distinct Outcomes of Bacterial Infection

    Extracellular vesicles are thought to facilitate pathogen transmission from arthropods to humans and other animals. Here, we reveal that pathogen spreading from arthropods to the mammalian host is multifaceted. Extracellular vesicles from Ixodes scapularis enable tick feeding and promote infection of the mildly virulent rickettsial agent Anaplasma phagocytophilum through the SNARE proteins Vamp33 and Synaptobrevin 2 and dendritic epidermal T cells. However, extracellular vesicles from the tick Dermacentor andersoni mitigate microbial spreading caused by the lethal pathogen Francisella tularensis. Collectively, we establish that tick extracellular vesicles foster distinct outcomes of bacterial infection and assist in vector feeding by acting on skin immunity. Thus, the biology of arthropods should be taken into consideration when developing strategies to control vector-borne diseases
