Last Glacial Maximum in an Andean cloud forest environment (Eastern Cordillera, Bolivia): Comment and Reply
Whether the climate of tropical South America during the Last Glacial Maximum (LGM) was colder and drier or colder and wetter than present day has been widely debated. It is accepted, however, that the LGM in tropical South America was 2–9 °C colder than today (e.g., Betts and Ridgway, 1992; Bush et al., 2001). Without debating the merits of the following choices, if we assume a lapse rate in the LGM similar to the modern one of ~0.6 °C·100 m−1, then an intermediate cooling of 5 °C would lower the boundary between montane cloud forest and the overlying puna grasslands by ~800 or 900 m. Palynologists on both sides of the wet/dry debate have come to similar conclusions about forest-boundary lowering due to temperature decrease (reviewed by Flenley, 1998). In the Eastern Cordillera of Bolivia the modern puna–cloud forest boundary lies ~3400 m above sea level (masl). Ignoring any other environmental changes, LGM cooling would have lowered this boundary to 2500 or 2600 masl
Ecological Legacies and Ethnotourism:Bridging Science and Community in Ecuador’s Amazonia
This paper offers paradigmatic insights from an international workshop on Ecological Legacies: Bridge Between Science and Community, in Ecuador, in the summer of 2023. The conference brought together foreign and local scholars, tour operators, village community, and Indigenous leaders in the upper Amazonia region of Ecuador with the goal of developing a vision for a sustainable and regenerative future of the upper Amazon. The conference offered three epistemological contributions to the existing literature in the emergent field of Montology, including addressing issues of (a) understanding the existing linguistic hegemony in describing tropical environments, (b) the redress of mistaken notions on pristine jungle environments, and (c) the inclusion of traditional knowledge and transdisciplinary approaches to understand the junglescape from different perspectives and scientific traditions. Methodologically, the conference bridged the fields of palaeoecological and ethnobotanical knowledge (as part of a wider conversation between science and local communities). Results show that local knowledge should be incorporated into the study of the junglescape and its conservation, with decolonial approaches for tourism, sharing language, methodology, tradition, and dissemination of the forest’s attributes. Our research helped co-create and formulate the “Coca Declaration” calling for a philosophical turn in research, bridging science and ethnotourism in ways that are local, emancipatory, and transdisciplinary. We conclude that facilitating new vocabulary by decolonial heightening of Indigenous perspectives of the junglescape helps to incorporate the notion of different Amazons, including the mountainscape of the Andean–Amazonian flanks. We also conclude that we can no consider Ecuador the country of “pure nature” since we helped demystify pristine nature for foreign tourists and highlighted local views with ancestral practices. Finally, we conclude that ethnotourism is a viable alternative to manage heritagization of the junglescape as a hybrid territory with the ecological legacies of the past and present inhabitants of upper Amazonia.</p

Characterization and identification of dityrosine cross-linked peptides using tandem mass spectrometry
The use of mass spectrometry coupled with chemical cross-linking of proteins has become a powerful tool for proteins structure and interactions studies. Unlike structural analysis of proteins using chemical reagents specific for lysine or cysteine residues, identification of gas-phase fragmentation patterns of endogenous dityrosine cross-linked peptides have not been investigated. Dityrosine cross-linking in proteins and peptides are clinical markers of oxidative stress, aging, and neurodegenerative diseases including Alzheimer’s disease and Parkinson’s disease. In this study, we investigated and characterized the fragmentation pattern of a synthetically prepared dityrosine cross-linked dimer of Aβ(1–16) using ESI tandem mass spectrometry. We then detailed the fragmentation pattern of dityrosine cross-linked Aβ(1–16), using collision induced dissociation (CID), higher-energy collision induced dissociation (HCD), electron transfer dissociation (ETD), and electron capture dissociation (ECD). Application of these generic fragmentation rules of dityrosine cross-linked peptides allowed for the identification of dityrosine cross-links in peptides of Aβ and α-synuclein generated in vitro by enzymatic peroxidation. We report, for the first time, the dityrosine cross-linked residues in human hemoglobin and α-synuclein under oxidative conditions. Together these tools open up the potential for automated analysis of this naturally occurring post-translation modification in neurodegenerative diseases as well as other pathological conditions
Quantitative determination of underivatized anti-convulsant drugs by high resolution gas chromatography with support-coated open tubular columns
Neutron and hard X-ray measurements during pellet deposition in TFTR
Measurements of neutrons and hard x rays are made with a pair of plastic scintillators during injection of deuterium pellets into deuterium TFTR plasmas. Three cases are investigated. During ohmic heating in plasmas with few runaway electrons, the neutron emission does not increase when a pellet is injected, indicating that strong acceleration of the pellet ions does not occur. In ohmic plasmas with low but detectable levels of runaway electrons, an x-ray burst is observed on a detector near the pellet injector as the pellet ablates, while a detector displaced 126/sup 0/ toroidally from the injector does not measure a synchronous burst. Reduced pellet penetration correlates with the presence of x-ray emission, suggesting that the origin of the burst is bremsstrahlung from runaway electrons that strike the solid pellet. In deuterium beam-heated discharges, an increase in the d-d neutron emission is observed when the pellet ablates. In this case, the increase is due to fusion reactions between beam ions and the high density neutral and plasma cloud produced by ablation of the pellet; this localized density perturbation equilibrates in about 700 ..mu..sec. Analysis of the data indicates that the density propagates without forming a sharp shock front with a rapid initial propagation velocity (greater than or equal to 2 x 10/sup 7/ cm/sec) that subsequently decreases to around 3 x 10/sup 6/ cm/sec. Modelling suggests that the electron heat flux into the pellet cloud is much less than the classical Spitzer value
Latent Class Analysis of Prenatal Substance Exposure and Child Behavioral Outcomes.
OBJECTIVES: To predict behavioral disruptions in middle childhood, we identified latent classes of prenatal substance use. STUDY DESIGN: As part of the Environmental influences on Child Health Outcomes Program, we harmonized prenatal substance use data and child behavior outcomes from 2195 women and their 6- to 11-year-old children across 10 cohorts in the US and used latent class-adjusted regression models to predict parent-rated child behavior. RESULTS: Three latent classes fit the data: low use (90.5%; n = 1986), primarily using no substances; licit use (6.6%; n = 145), mainly using nicotine with a moderate likelihood of using alcohol and marijuana; and illicit use (2.9%; n = 64), predominantly using illicit substances along with a moderate likelihood of using licit substances. Children exposed to primarily licit substances in utero had greater levels of externalizing behavior than children exposed to low or no substances (P = .001, d = .64). Children exposed to illicit substances in utero showed small but significant elevations in internalizing behavior than children exposed to low or no substances (P < .001, d = .16). CONCLUSIONS: The differences in prenatal polysubstance use may increase risk for specific childhood problem behaviors; however, child outcomes appeared comparably adverse for both licit and illicit polysubstance exposure. We highlight the need for similar multicohort, large-scale studies to examine childhood outcomes based on prenatal substance use profiles

Comprehensive analysis of epigenetic clocks reveals associations between disproportionate biological ageing and hippocampal volume
The concept of age acceleration, the difference between biological age and chronological age, is of growing interest, particularly with respect to age-related disorders, such as Alzheimer’s Disease (AD). Whilst studies have reported associations with AD risk and related phenotypes, there remains a lack of consensus on these associations. Here we aimed to comprehensively investigate the relationship between five recognised measures of age acceleration, based on DNA methylation patterns (DNAm age), and cross-sectional and longitudinal cognition and AD-related neuroimaging phenotypes (volumetric MRI and Amyloid-β PET) in the Australian Imaging, Biomarkers and Lifestyle (AIBL) and the Alzheimer’s Disease Neuroimaging Initiative (ADNI). Significant associations were observed between age acceleration using the Hannum epigenetic clock and cross-sectional hippocampal volume in AIBL and replicated in ADNI. In AIBL, several other findings were observed cross-sectionally, including a significant association between hippocampal volume and the Hannum and Phenoage epigenetic clocks. Further, significant associations were also observed between hippocampal volume and the Zhang and Phenoage epigenetic clocks within Amyloid-β positive individuals. However, these were not validated within the ADNI cohort. No associations between age acceleration and other Alzheimer’s disease-related phenotypes, including measures of cognition or brain Amyloid-β burden, were observed, and there was no association with longitudinal change in any phenotype. This study presents a link between age acceleration, as determined using DNA methylation, and hippocampal volume that was statistically significant across two highly characterised cohorts. The results presented in this study contribute to a growing literature that supports the role of epigenetic modifications in ageing and AD-related phenotypes
