    Bishop Peak Natural Reserve Conservation Plan Update

    My master\u27s project is updating the Conservation Plan for the Bishop Peak Natural Reserve (BPNR) in San Luis Obispo, California. It is a professional project for the City of San Luis Obispo, supported by City policy that requires continued management of its open space network through management plans unique to each property. As one of the city\u27s most visited open spaces and one of its most visible natural landmarks, the Bishop Peak Natural Reserve is in need of continued management and oversight. While much of the work in developing the plan focused on identifying issues and developing responsive policy, this background report takes a step back, focusing on the underlying principles and municipal policies which direct those efforts. This paper serves as a background report for the planning process including an overview of methodology and policy development. One of the primary goals set forth in the BPNR update process is to improve of management capacity and increase the potential for successful implementation. To accomplish this, the reports shows how aspects of an Adaptive Management approach can be integrated into plan review and development. This background report is intended to complement the plan itself and therefore issues not covered within this report are covered within the plan

    Organic Anion and Cation Transporter Expression and Function During Embryonic Kidney Development and in Organ Culture Model Systems

    Background Organic anion and cation transporters (OATs, OCTs and OCTNs) mediate the proximal tubular secretion of numerous clinically important compounds, including various commonly prescribed pharmaceuticals. Here, we examine the ontogeny of these transporters in rat embryonic kidney in detail, both in vivo and in two in vitro organ culture models of kidney development, whole embryonic kidney (WEK) culture and culture of induced metanephric mesenchyme (MM). Methods We used QPCR to determine expression levels of transporter genes in rat embryonic kidneys on each day of gestation from ed13 to ed18, in induced and un-induced MM, and on each day of one week of WEK culture. We also used uptake of fluorescein as a novel functional assay of organic anion transporter expression in WEK and MM. Results The developmental induction of the various organic anion and cation transporter genes does not occur uniformly: some genes are induced early (e.g., Oat1 and Oat3, potential early markers of proximal tubulogenesis), and others not till kidney development is relatively advanced (e.g., Oct1, a potential marker of terminal differentiation). We also find that the ontogeny of transporter genes in WEK and MM is similar to that observed in vivo, indicating that these organ culture systems may appropriately model the expression of OATs, OCTs and OCTNs. Conclusion We show that WEK and MM cultures may represent convenient in vitro models for study of the developmental induction of organic anion and cation transporters. Functional organic anion transport as measured by fluorescein uptake was evident by accumulation of the fluorescence in the developing tubule in these organ cultures. By demonstrating the mediated uptake of fluorescein in WEK and MM, we have established a novel in vitro functional assay of transporter function. We find that OATs, OCTs, and OCTNs are differentially expressed during proximal tubule development. Our findings on the renal ontogeny of organic anion and cation transporters could carry implications both for the development of more rational therapeutics for premature infants, as well as for our understanding of proximal tubule differentiation

    eDNAPlus: A unifying modelling framework for DNA-based biodiversity monitoring

    DNA-based biodiversity surveys involve collecting physical samples from survey sites and assaying the contents in the laboratory to detect species via their diagnostic DNA sequences. DNA-based surveys are increasingly being adopted for biodiversity monitoring. The most commonly employed method is metabarcoding, which combines PCR with high-throughput DNA sequencing to amplify and then read `DNA barcode' sequences. This process generates count data indicating the number of times each DNA barcode was read. However, DNA-based data are noisy and error-prone, with several sources of variation. In this paper, we present a unifying modelling framework for DNA-based data allowing for all key sources of variation and error in the data-generating process. The model can estimate within-species biomass changes across sites and link those changes to environmental covariates, while accounting for species and sites correlation. Inference is performed using MCMC, where we employ Gibbs or Metropolis-Hastings updates with Laplace approximations. We also implement a re-parameterisation scheme, appropriate for crossed-effects models, leading to improved mixing, and an adaptive approach for updating latent variables, reducing computation time. We discuss study design and present theoretical and simulation results to guide decisions on replication at different stages and on the use of quality control methods. We demonstrate the new framework on a dataset of Malaise-trap samples. We quantify the effects of elevation and distance-to-road on each species, infer species correlations, and produce maps identifying areas of high biodiversity, which can be used to rank areas by conservation value. We estimate the level of noise between sites and within sample replicates, and the probabilities of error at the PCR stage, which are close to zero for most species considered, validating the employed laboratory processing.Comment: The paper is 35 pages long and it has 8 figure

    The Ransom and Sarah Williams Farmstead: Post-Emancipation Transitions of an African American Family in Central Texas Vol. 1

    In conjunction with the proposed construction of the southwest segment of State Highway 45 in southern Travis County, the Texas Department of Transportation sponsored archeological testing and data recovery efforts at the Ransom Williams farmstead. Prewitt and Associates, Inc., conducted an interdisciplinary community-based historic archeological study of the farmstead from 2005 through 2011. Extensive archival research reveals that the 45-acre farm was owned and occupied by Ransom Williams and his wife Sarah, both former slaves, from about 1871 to ca. 1905. The Williams family lived in the predominantly white rural community of Bear Creek, but they had connections to the nearby freedmen communities of Antioch Colony in northern Hays County and Manchaca in southern Travis County. The stories of the Ransom Williams family and their connections to these communities are enhanced by extensive oral history research, with over 46 hours of taped and transcribed interviews with 27 descendant community members. Data recovery investigations focused on a landscape archeological study to define the layout and design of the entire farmstead, including a stock pond and a network of dry-laid rock walls that facilitated water drainage, demarcated property boundaries, and formed livestock pens. Intensive hand excavations were used to examine features associated with the Williams house, outbuildings and activity areas, and a large trash midden. This work recovered more than 26,000 artifacts. They constitute an impressive material culture assemblage that is associated, with few exceptions, with the Williams family tenure on the land. The combined archival data, oral history interviews, and archeological evidence tell the fascinating story of how one African American farm family lived and thrived in central Texas during Reconstruction and into the Jim Crow era

    Understanding experiences of participating in a weight loss lifestyle intervention trial: a qualitative evaluation of South Asians at high risk of diabetes

    <b>Objective</b> To explore the reasons for enrolling, experiences of participating and reasons for remaining in a family-based, cluster randomised controlled trial of a dietitian-delivered lifestyle modification intervention aiming to reduce obesity in South Asians at high risk of developing diabetes.<p></p> <b>Design</b> Qualitative study using narrative interviews of a purposive sample of trial participants following completion of the intervention. Data were thematically analysed.<p></p> <b>Setting</b> The intervention was conducted in Scotland and resulted in a modest decrease in weight, but did not statistically reduce the incidence of diabetes.<p></p> <b>Participants</b> We conducted 21 narrative interviews with 24 participants (20 trial participants and four family volunteers).<p></p> <b>Results</b> Many participants were motivated to participate because of: known family history of diabetes and the desire to better understand diabetes-related risks to their own and their family's health; ways to mitigate these risks and to benefit from personalised monitoring. Home-based interventions, communication in the participant's chosen language(s) and continuity in dietitians supported their continuing engagement with the trial. Adaptations in food choices were initially accommodated by participants, although social and faith-based responsibilities were reported as important barriers to persevering with agreed dietary goals. Many participants reported that increasing their level of physical activity was difficult given their long working hours, physically demanding employment and domestic commitments; this being compounded by Scotland's challenging climate and a related reluctance to exercise in the outdoors.<p></p> <b>Conclusions</b> Although participants had strong personal interests in participation and found the information provided by dietitians useful, they nonetheless struggled to incorporate the dietary and exercise recommendations into their daily lives. In particular, increasing levels of physical exercise was described as an additional and in some cases unachievable burden. Consideration needs to be given to strengthening and supporting lifestyle interventions with community-based approaches in order to help overcome wider social and environmental factors

    Electron multiplication CCD detector technology advancement for the WFIRST-AFTA coronagraph

    The WFIRST-AFTA (Wide Field InfraRed Survey Telescope-Astrophysics Focused Telescope Asset) is a NASA space observatory. It will host two major astronomical instruments: a wide-field imager (WFI) to search for dark energy and carry out wide field near infrared (NIR) surveys, and a coronagraph instrument (CGI) to image and spectrally characterize extrasolar planets. In this paper, we discuss the work that has been carried out at JPL in advancing Electron Multiplying CCD (EMCCD) technology to higher flight maturity, with the goal of reaching a NASA technology readiness level of 6 (TRL-6) by early-to-mid 2016. The EMCCD has been baselined for both the coronagraph's imager and integral field spectrograph (IFS) based on its sub-electron noise performance at extremely low flux levels - the regime where the AFTA CGI will operate. We present results from a study that fully characterizes the beginning of life performance of the EMCCD. We also discuss, and present initial results from, a recent radiation test campaign that was designed and carried out to mimic the conditions of the WFIRST-AFTA space environment in an L2 orbit, where we sought to assess the sensor's end of life performance, particularly degradation of its charge transfer efficiency, in addition to other parameters such as dark current, electron multiplication gain, clock induced charge and read noise

    Resource-based view of the firm Complementarities Employee development Customer satisfaction Performance Using the theoretical foundations of the resource-based view of the firm, this study develops and measures marketing employee development capabilities and investigates how it moderates the relationships between brand and customer relationship management capabilities and firm performance outcomes. Based on a random sample survey of chief marketing executives from selected industries, combined with objective firm performance indicators and controls, results demonstrate that marketing employee development capabilities can leverage the relationships between firm-level marketing capabilities and customer satisfaction, market effectiveness, and objective financial performance. Further, these results show that such capabilities can be complementary and, in some cases, even substitute for each other, which improves organizational performance. Implications for researchers and marketing managers are discussed
